Isotopic signatures of carbon and nitrogen are widely used for analysis of the structure of food webs in aquatic ecosystems. The study of animals raises a number of methodological questions, including choice of representative tissues and organs for sampling as well as storage of the studied organisms. Furthermore, the impacts of preservation methods can be tissue-specific, age-specific, and even taxon-specific; thus, studies of these impacts on particular taxa are necessary. We focused on the C and N isotope composition of the common toad (Bufo bufo), one of the most widespread European anuran amphibians. We hypothesized that its different tissues and organs may vary in isotopic composition, and ethanol and freezing may have different effects on isotopic values. Our results showed that both “tissue” and “storage method” factors significantly affected the δ13C values of tadpoles and postmetamorphic juveniles, whereas only the “tissue” factor had a significant effect on the δ15N values. The two stages, tadpoles and postmetamorphs, should be analyzed separately despite the brief postmetamorphic period of the juveniles. The skin, legs, muscles, and tail in tadpoles and legs, muscles and heart in juveniles can be used for δ13C and δ15N analysis regardless of the method of storage. The results will serve for the optimization of future study designs in isotopic ecology.

Sommers Island is an abandoned 20 ha patch of land in the Baltic Sea. It is inhabited by isolated populations of smooth newt (Lissotriton vulgaris) and common toad (Bufo bufo). The island demonstrates the small area in which it is possible to have a stable population of newts and toads for a period of at least several decades, probably much longer. Most reproduction of both species occurred in one pool with an area of about 100 m². The total area of optimal terrestrial habitat is about 2.5 ha, with an additional 3.5 ha of sub-optimal habitat available. The island is inhabited by about 123 adult newts and several dozen adult toads.

Biological invasions are on the rise, with each invader carrying a plethora of associated microbes. These microbes play important, yet poorly understood, ecological roles that can include assisting the hosts in colonization and adaptation processes or as possible pathogens. Understanding how these communities differ in an invasion scenario may help to understand the host’s resilience and adaptability. The Asian common toad, Duttaphrynus melanostictus is an invasive amphibian, which has recently established in Madagascar and is expected to pose numerous threats to the native ecosystems. We characterized the skin and gut bacterial communities of D. melanostictus in Toamasina (Eastern Madagascar), and compared them to those of a co-occurring native frog species, Ptychadena mascareniensis, at three sites where the toad arrived in different years. Microbial composition did not vary among sites, showing that D. melanostictus keeps a stable community across its expansion but significant differences were observed between these two amphibians. Moreover, D. melanostictus had richer and more diverse communities and also harboured a high percentage of total unique taxa (skin: 80%; gut: 52%). These differences may reflect the combination of multiple host-associated factors including microhabitat selection, skin features and dietary preferences.

Abstract The high load of agrochemicals and antibiotics coexisting in aquatic environments within agroecosystems represents a risk for wildlife. As the gut microbiota plays a key role on its host’s functioning and is sensitive to a wide variety of pollutants, its study allows evaluating organisms’ health and therefore, the ecosystem. We studied toxic effects of commercial formulations of a glyphosate-based herbicide (GBH) and the antibiotic ciprofloxacin (CIP) on gut bacterial microbiota diversity of the common toad (Rhinella arenarum) tadpoles, considered a sentinel species. The study was carried out by classic microbiological analysis and matrix-assisted laser desorption ionization-time of flight mass spectrometry. The microbiota from GBH treatment had greater taxa diversity and richness, including some genera, such as Proteus spp. and Yersinia spp. that were absent in control. In contrast, microbiota from CIP treatment registered a decrease of diversity indexes, dominance of Aeromonas spp. and presence of Leclercia spp. The GBH-CIP treatment showed changes in taxa composition, including decrease of Klebsiella spp. and Pseudomonas spp. and trends of individual pollutant treatments. For all cases, changes in the composition of bacterial community (dysbiosis) were linked to a significant decrease in tadpoles’ weight. To the best of our knowledge, this is the first report on the disruption of gut bacterial microbiota of tadpoles by a mixture of two contaminants of emerging concern worldwide. These findings contribute to understanding how the presence of two co-occurring pollutants in freshwaters results in deleterious effects on the amphibian community and potentially affect the microbiota of those environments.

AbstractIntroductionDespite the growing number of studies describing digit ratio patterns in tetrapods, knowledge concerning certain basic issues is still scarce. In lower vertebrates such as tailless amphibians (Anura), the numbering of individual fingers on the forelimbs and their homology with the fingers of other vertebrates pose an unsolved problem. Based on reviewed data on anuran limb development, we argue that the correct finger numbering scheme should be based on the assumption that the first finger, not the fifth finger, was reduced on the forelimbs. We analyzed the digit ratio in the common toad (Bufo bufo, Bufonidae), a species characterized by well-developed sexual dimorphism whereby females are larger than males, using both numbering schemes present in the literature.ResultsWe found that the digit ratio on hindlimbs differed significantly between the sexes only in the cases of left 2D:3D, with lower digit ratios in females, and of left 3D:4D, with lower digit ratios in males. We found that sex was the only significant variable for forelimbs, differentiating 2D:3D on the left forelimb, with lower digit ratios in females; 2D:4D on the right forelimb, with lower digit ratios in males; and 3D:4D on both forelimbs, with lower digit ratios in males. These results relate to variant II reflecting the hypothesis that the first digit was reduced during phylogenesis. There was no relationship between the body size (SVL) of individuals and any digit ratio, excluding 2D:4D on the right forelimbs in models with age variables. Additionally, for a subset of data where individual age was known, the models indicated that age was linked to significant differences in 2D:4D and 3D:4D on the left hindlimbs, while age, SVL, and sex influenced 2D:4D on the right forelimbs.ConclusionWe emphasize the importance of the problem of the correct numbering of forelimb digits in Anura and, under the assumption that it was the fifth digit that was reduced, argue that earlier results on digit ratio in this group should be interpreted with caution. The detected relationship between digit ratio and age in amphibians expands our knowledge, indicating that the age of individuals should be included in future digit ratio studies. This relationship may also apply to studies using digit ratio as a noninvasive indicator of endocrine disruption in amphibians.
