Sturgeons are among the most ancient linages of actinopterygians. At present, many sturgeon species are critically endangered. Surrogate production could be used as an affordable and a time-efficient method for endangered sturgeons. Our study established a method for identifying and isolating type A spermatogonia from different developmental stages of testes using flow cytometric cell sorting (FCM). Flow cytometric analysis of a whole testicular cell suspension showed several well-distinguished cell populations formed according to different values of light scatter parameters. FCM of these different cell populations was performed directly on glass slides for further immunocytochemistry to identify germ cells. Results showed that the cell population in gate P1 on a flow cytometry plot (with high forward scatter and high side scatter parameter values) contains the highest amount of type A spermatogonia. The sorted cell populations were characterized by expression profiles of 10 germ cell specific genes. The result confirmed that setting up for the P1 gate could precisely sort type A spermatogonia in all tested testicular developmental stages. The P2 gate, which was with lower forward scatter and side scatter values mostly, contained type B spermatogonia at a later maturing stage. Moreover, expressions of plzf, dnd, boule, and kitr were significantly higher in type A spermatogonia than in later developed germ cells. In addition, plzf was firstly found as a reliable marker to identify type A spermatogonia, which filled the gap of identification of spermatogonial stem cells in sterlet. It is expected to increase the efficiency of germ stem cell culture and transplantation with plzf identification. Our study thus first addressed a phenotypic characterization of a pure type A spermatogonia population in sterlet. FCM strategy can improve the production of sturgeons with surrogate broodstock and further the analysis of the cellular and molecular mechanisms of sturgeon germ cell development.

Abstract Background: The microbial production of hemicellulasic cocktails is still a challenge for the sector of biorefineries and agro-waste valorization. In this work, the production of hemicellulolytic enzymes by Thermobacillus xylanilyticus has been considered. This microorganism is of interest since it is able to produce an original set of thermostable hemicellulolytic enzymes, and notably a xylanase GH11, Tx-xyn11. However, cell-to-cell heterogeneities impairs the production capability of the whole microbial population.Results: Sequential cultivations of the strain on xylan as a carbon source has been considered in order to highlight and better understand this cell-to-cell heterogeneity. Successive cultivations pointed out a fast decrease of xylanase activity (loss of ~75%) after 23.5 generations. Accordingly, the expression of the Tx-xyn11 gene decreased drastically and followed the same trend as the xylanase activity. Flow cytometry analyses pointed out that two subpopulations, differing at the level of their light scattering properties, were potentially involved in this progressive loss of enzymatic activities. Interestingly, upon successive cultivations on xylan, the subpopulation exhibiting low forward scatter (FSC) signal. Additionally, the evolution of the ratio between the two subpopulations was correlated to the decrease in xylanase activity. Cell sorting and direct observation of the sorted subpopulations revealed that the low-FSC subpopulation was not sporulating, whereas the high-FSC subpopulation contained cells at the onset of the sporulation stage. Serial cultivations on glucose, followed by the addition of a xylan pulse led to a ~1.5-fold to ~15-fold improvement of xylanase, depending on the moment for pulse addition, , suggesting that alternating cultivation conditions could lead to an efficient population management strategy for the production of xylanase. Conclusions: Taken altogether, the data from this study point out that a cheating behaviour is responsible for the progressive reduction in xylanase activity during serial cultivations of T. xylanilyticus. Alternating cultivation condition between glucose and xylan could be used as an efficient strategy for promoting population stability and higher enzymatic productivity from this bacterium.

Abstract Visibility lidar has obvious monitoring advantages over forward scatter visibility sensors or fog droplet spectrometers; it can measure visibility information over a large area. In 2021, two visibility lidar instruments (1064 or 532 nm wavelengths) were installed in Beilun, Ningbo Zhoushan Port, to monitor sea fog. Comparing their monitoring data to those of forward scatter visibility sensors and a fog droplet spectrometer revealed that the visibility lidar instruments could obtain energy progress information section-by-section in the monitoring path, and could directly reflect sea fog changes. The 1064 nm lidar outperformed the 532 nm lidar regarding sea fog detection. The effective detection range decreased significantly with decreasing visibility; the reliability decreased in low-visibility, uneven atmospheres. In a low-visibility but uniform atmosphere, however, lidar data corresponded well with forward dispersion data. The 532 nm and 1064 nm lidar data sometimes differed at the same monitoring position owing to differing heights and particle reflection angles. During a sea fog event on May 9, 2021, the maximum droplet concentration was 14 cm−3, the maximum liquid water content was 0.21 g·m−3, and the maximum equivalent diameter was 49 μm. The formation of this sea fog was dominated by large particles.

Purpose. The role of oxidative stress in Aluminum (Al)-induced apoptotic effects has been investigated and suicidal death of erythrocytes, eryptosis, is characterized by cell shrinkage and phosphatidylserine externalization (PSE) at the surface of the erythrocyte cell membrane. Eryptosis is stimulated by an increase in cytosolic Ca2+ concentration and reactive oxygen species (ROS). This ex vivo study was conducted to evaluate the effect of well-known antioxidants including vitamin C (vit C) and N-acetylcysteine (NAC), against Al-induced hemolysis and eryptosis. Methods. Isolated erythrocytes from the healthy volunteers were partitioned into various groups (6 replicates/group) and treated by various concentrations of Al (3–100 µM) in the presence and absence of vit C (0.6 mM) and NAC (1 mM). After 24 hours of treatment, hemolysis was determined from hemoglobin levels in the supernatant. Flowcytometric methods were applied to measure PSE, cell shrinkage, Ca2+ content, and ROS abundance using annexin V-binding, forward scatter, Fluo3-fluorescence, and DCFDA dependent fluorescence, respectively. Reduced glutathione (GSH) was measured by the ELISA method. Results. The results showed that a 24 hours’ exposure of the erythrocytes to Al (10–100 µM) significantly increased hemolysis in a dose and Ca2+dependent manner. Al also dramatically decreased forward scatter. The percentage of PSE cells, Fluo3-fluorescence, and DCFDA fluorescence were increased by Al. Furthermore, cotreatment with NAC inhibited the effect of Al on hemolysis, eryptosis, and ROS production. Vit C decreased Al-induced ROS production. However, increased Al-induced eryptosis. There were no significant changes in glutathione after the ALCL3 treatment. Conclusions. Al-induced eryptosis and hemolysis through triggering oxidative stress, while NAC could diverse this effect. In contrast, vit C might intensify Al-induced eryptosis at particular doses through a less known mechanism.

The paper analyses the possibility of Forward Scatter Radar (FSR) systems to detect airplanes using cosmic emission from pulsars and planets (pulsar, Sun, Moon). A suboptimal multichannel algorithm for joint detection and evaluation of the parameters of the forward scattering signal created by an airplane (duration and velocity) is proposed, with preliminary compensation of the powerful direct signal emitted by cosmic sources (pulsar, Sun and Moon). The expressions for calculation of the Signal-to-Noise Ratio (SNR) at the input of the detector and the compensator are obtained. The detection characteristics are also obtained, and the requirements for the suppression coefficient of the compensator are evaluated. A methodology for calculating the maximum distance for detecting an aircraft using a described algorithm is proposed. The obtained results show that due to the Forward Scatter (FS) effect, there is the theoretical possibility to detect airplanes at close ranges by FSRs, which use very weak signals from cosmic sources.
