The parasitoid wasp Muscidifurax raptorellus (Hymenoptera: Pteromalidae) is a gregarious species that has received extensive attention for its potential in biological pest control against house fly, stable fly, and other filth flies. It has a high reproductive capacity and can be reared easily. However, genome assembly is not available for M. raptorellus or any other species in this genus. Previously, we assembled a complete circular mitochondrial genome with a length of 24,717 bp. Here, we assembled and annotated a high-quality nuclear genome of M. raptorellus, using a combination of long-read (104× genome coverage) and short-read (326× genome coverage) sequencing technologies. The assembled genome size is 314 Mbp in 226 contigs, with a 97.9% BUSCO completeness score and a contig N50 of 4.67 Mb, suggesting excellent continuity of this assembly. Our assembly builds the foundation for comparative and evolutionary genomic analysis in the genus of Muscidifurax and possible future biocontrol applications.

Abstract Most parasitoid wasps parasitize herbivorous insects, so nectar from flowers is readily available. However, parasitoid wasps are also an important component of the rich invertebrate communities at livestock facilities in large accumulations of manure, where flowers are largely absent. Little is known about adult parasitoid diet and nutrition in these communities. The present study examined this in Spalangia cameroni Perkins, a pupal parasitoid of filth flies. Like many parasitoid wasps, S. cameroni feed on host fluids, and in the laboratory readily feed on honey or a sucrose solution, which increases their longevity. Here adult longevity in the presence of six potential food sources, bovine manure, sorghum silage, bovine milk, buckwheat inflorescence (Polygonaceae), sweet alyssum inflorescence (Brassicaceae), or dandelion inflorescence (Asteraceae), was compared to that with water or honey. Only parasitoids given buckwheat lived as long as parasitoids given honey, and parasitoids given honey or buckwheat lived longer than parasitoids given water. Parasitoids readily ate buckwheat nectar, avoiding pollen grains. Diet affected the amount of free sugars, glycogen, and lipids in complex ways. Compared to parasitoids that were given just water, parasitoids with access to honey or sucrose had higher sugar and glycogen levels, but not detectably higher lipid levels. Access to buckwheat had no detectable effect on a parasitoid’s free sugar, glycogen, or lipid levels; however, then after 4 d with just water, sugar levels were lower and glycogen levels were higher compared to parasitoids that had been given access to only water the entire time.

Abstract ‘Filth flies’ facilitate the dispersal of pathogens between animals and humans. The objective was to study the intestinal colonization with antimicrobial resistant and enteropathogenic bacteria in ‘filth flies’ from Nigeria. Flies from Southern Nigeria were screened for extended-spectrum β-lactamase producing Enterobacterales (ESBL-E), Staphylococcus aureus, Salmonella sp., Shigella sp., Campylobacter sp. and Yersinia enterocolitica by culture. ESBL-E were tested for blaSHV, blaCTX-M and blaTEM; S. aureus was screened for enterotoxins. Spa typing and multilocus sequence typing (MLST) was done for S. aureus and MLST for Escherichia coli. Of 2,000 flies, 400 were randomly collected for species identification. The most common species were Musca domestica (44.8%, 179/400), Chrysomya putoria (21.6%, 85/400) and Musca sorbens (18.8%, 75/400). Flies were colonized with S. aureus (13.8%, 275/2,000) and ESBL-E (0.8%, 16/2,000). No other enteropathogenic bacteria were detected. The enterotoxin sei was most common (26%, 70/275) in S. aureus, followed by sea (12%, n = 32/275). Four S. aureus isolates were methicillin resistant (mecA positive, t674 and t5305, ST15). The blaCTX-M (n = 16) was the most prevalent ESBL subtype, followed by blaTEM (n = 8). ‘Filth flies’ can carry antimicrobial resistant bacteria in Nigeria. Enterotoxin-positive S. aureus might be the main reason for food poisoning by ‘filth flies’ in the study area.
