In this work, a 3D computational model based on computational fluid dynamics (CFD) is built to simulate the aerodynamic behavior of a Savonius-type vertical axis wind turbine with a semi-elliptical profile. This computational model is used to evaluate the performance of the wind turbine in terms of its power coefficient (Cp). Subsequently, a full factorial design of experiments (DOE) is defined to obtain a representative sample of the search space on the geometry of the wind turbine. A dataset is built on the performance of each geometry proposed in the DOE. This process is carried out in an automated way through a scheme of integrated computational platforms. Later, a surrogate model of the wind turbine is fitted to estimate its performance using machine learning algorithms. Finally, a process of optimization of the geometry of the wind turbine is carried out employing metaheuristic optimization algorithms to maximize its Cp; the final optimized designs are evaluated using the computational model for validating their performance.

This paper proposes a new surrogate optimization routine for optimal design of a direct on line (DOL) squirrel cage induction motor. The geometry of the motor is optimized to maximize its electromagnetic efficiency while respecting the constraints, such as output power and power factor. The routine uses the methodologies of Latin-hypercube sampling, a clustering technique and a Box–Behnken design for improving the accuracy of the surrogate model while efficiently utilizing the computational resources. The global search-based particle swarm optimization (PSO) algorithm is used for optimizing the surrogate model and the pattern search algorithm is used for fine-tuning the surrogate optimal solution. The proposed surrogate optimization routine achieved an optimal design with an electromagnetic efficiency of 93.90%, for a 7.5 kW motor. To benchmark the performance of the surrogate optimization routine, a comparative analysis was carried out with a direct optimization routine that uses a finite element method (FEM)-based machine model as a cost function.

Burkholderia pseudomallei is a soil-dwelling organism present throughout the tropics. It is the causative agent of melioidosis, a disease that is believed to kill 89,000 people per year. It is naturally resistant to many antibiotics, requiring at least two weeks of intravenous treatment with ceftazidime, imipenem or meropenem followed by 6 months of orally delivered co-trimoxazole. This places a large treatment burden on the predominantly middle-income nations where the majority of disease occurs. We have established a high-throughput assay for compounds that could be used as a co-therapy to potentiate the effect of ceftazidime, using the related non-pathogenic bacterium Burkholderia thailandensis as a surrogate. Optimization of the assay gave a Z’ factor of 0.68. We screened a library of 61,250 compounds and identified 29 compounds with a pIC50 (-log10(IC50)) greater than five. Detailed investigation allowed us to down select to six “best in class” compounds, which included the licensed drug chloroxine. Co-treatment of B. thailandensis with ceftazidime and chloroxine reduced culturable cell numbers by two orders of magnitude over 48 hours, compared to treatment with ceftazidime alone. Hit expansion around chloroxine was performed using commercially available compounds. Minor modifications to the structure abolished activity, suggesting that chloroxine likely acts against a specific target. Finally, an initial study demonstrates the utility of chloroxine to act as a co-therapy to potentiate the effect of ceftazidime against B. pseudomallei. This approach successfully identified potential co-therapies for a recalcitrant Gram-negative bacterial species. Our assay could be used more widely to aid in chemotherapy to treat infections caused by these bacteria.

Unit commitment (UC) is an important problem solved on a daily basis within a strict time limit. While hourly UC problems are currently considered, they may not be flexible enough with the fast-changing demand and the increased penetration of intermittent renewables. Sub-hourly UC is therefore recommended. This, however, will significantly increase problem complexity even under the deterministic setting, and current methods may not be able to obtain good solutions within the time limit. In this paper, deterministic sub-hourly UC is considered, with the innovative exploitation of soft constraints – constraints that do not need to be strictly satisfied, but with predetermined penalty coefficients for their violations. The key idea is the “surrogate optimization” concept that ensures multiplier convergence within “surrogate” Lagrangian relaxation as long as the “surrogate optimality condition” is satisfied without the need to optimally solve the “relaxed problem.” Consequently, subproblems can still be formed and optimized when soft constraints are not relaxed, leading to a drastically reduced number of multipliers and improved performance. To further enhance the method, a parallel version is developed. Testing results on the Polish system demonstrate the effectiveness and robustness of both the sequential and parallel versions at finding high-quality solutions within the time limit.
