A numerical study on the oxidation removal of nitric oxide in a ceramic hollow fiber membrane contactor was performed. To represent the transport and absorption process, the model was created by combining multiphase, species, reaction, and porous models. The numerical results were verified by comparing them with experimental data. The tube and lumen sides both have laminar parabolic velocity distributions. The nitric oxide concentration decreases gradually from the membrane wall to axis at the cross-section except on the inner and outer sides of the membrane tube. The equivalent diffusion length was proved useful for evaluating the entrance effect. At low concentrations, the reduction efficiency was proportional to the absorbent concentration, and at large concentrations, it neared a maximum value. The reduction efficiency was positively affected by elevated operating temperature and pressure. With a gas channel width of 13 mm, the reduction flow rate achieves its maximum. The efficiency of NO reduction per area decreases as the effective membrane length increases. Increasing the operating temperature and membrane length are recommended as design priorities due to high relative enhancements. It is not recommended to improve reduction efficiency by increasing membrane tube diameter and operating pressure in design. Changing the gas flow rate, absorbent concentration and gas channel width are moderate recommended as well.

Glioblastoma is a particularly challenging disease characterized by the connection of tumor cells to functional multicellular networks that effectively resist therapies. In this issue of Biochemical Journal, Pinto et al. report the discovery of two distinct classes of intercellular membrane tube connections, tunneling nanotubes and tumor microtubes, in the same state-of-the-art culture model of patient-derived glioblastoma material. These findings contribute to our understanding of the heterogeneity of intercellular membrane tubes in health and disease, and pave the way for future functional studies on their various roles for disease progression and tumor resistance.

Objective The purpose of this study was to observe feasibility of nano calcium-deficient hydroxyapatite-multi (amino acid) copolymer (n-CDHA-MAC) membrane tubes in repairing goat femurs’ large defects. Methods Twelve goats were divided into two groups, whose femurs were created 30 mm segmental bone defects and then implants were performed. In experimental group, the bone defect of right femur was reconstructed by n-CDHA-MAC membrane tube, while left side was reconstructed by allogenic bone tube in control group. Every three goats were sacrificed at 4, 8, 16, 24 weeks after operation respectively. General observation, X-ray analysis, histology, Scanning electron microscope (SEM) examination and protein level comparison of BMP-2 were conducted to evaluate the effects of repairing segmental bone defects. Results All goats recovered well from anesthesia and surgical interventions. The radiographic evaluations showed that periosteal reaction outside of the membrane tubes and allogenic bone tubes were observed 4 weeks after surgery. At 16 weeks, callus was continuously increased in experimental group, which was more obvious than control group. At 24 weeks, callus outside of the membrane tubes connected together. Histologic evaluation showed fibro-cartilage callus was evolved into bony callus in experimental group, which was more obvious than control group at 8 and 16 weeks. The protein expression level of BMP-2 increased at 4, 8 weeks and peaked at 16 weeks in experimental groups. There were statistical differences at 8 and 16 weeks ( P < 0.05). At each time point in 8, 16, 24 weeks after surgery, the bending stiffness, torsional stiffness and compressive strength of the two groups were similar, and there was no significant difference ( P > 0.05). Conclusions This novel surface degradation n-CDHA-MAC membrane tube has good ability to maintain enough membrane space, which can provide long-term and stable biomechanical support for large bone defects and integrate well with the surrounding bone.

Abstract Dynamin plays an important role in clathrin-mediated endocytosis by cutting the neck of nascent vesicles from the cell membrane. Gold nanorods were used as imaging probes to observe dynamin action on cargo vesicles during live endocytosis events. Invariant is that at the peak of dynamin accumulation, the cargo-containing vesicle always gives abrupt, right-handed rotations that finishes in a short time (~ 0.28 s). The large and quick twist, herein named the super twist, is the result of the coordinated dynamin helix action upon GTP hydrolysis. After the super twist, the rotational freedom of the vesicle drastically increases, accompanied with simultaneous or delayed translational movement, indicating that it detaches from the cell membrane. These observations suggest that dynamin-mediated scission at the final stage involves a large torque generated by coordinated actions of multiple dynamins in the helix, which is the main driving force for scission. The super twist presumably results in membrane tube hemi-fission and partial destruction of the dynamin helix, followed by vesicle fission.

AbstractThe mechano-chemical GTPase dynamin assembles on membrane necks of clathrin-coated vesicles into helical oligomers that constrict and eventually cleave the necks in a GTP-dependent way. It remains not clear whether dynamin achieves this via molecular motor activity and, if so, by what mechanism. Here, we used ensemble kinetics, single-molecule FRET and molecular dynamics simulations to characterize dynamin’s GTPase cycle and determine the powerstroke strength. The results were incorporated into a coarse-grained structural model of dynamin filaments on realistic membrane templates. Working asynchronously, dynamin’s motor modules were found to collectively constrict a membrane tube. Force is generated by motor dimers linking adjacent helical turns and constriction is accelerated by their strain-dependent dissociation. Consistent with experiments, less than a second is needed to constrict a membrane tube to the hemi-fission radius. Thus, a membrane remodeling mechanism relying on cooperation of molecular ratchet motors driven by GTP hydrolysis has been revealed.

Abstract We first characterize strain solitary waves propagating in a fluid-filled membrane tube when the fluid is stationary prior to wave propagation and the tube is also subjected to a finite stretch. We consider the parameter regime where all traveling waves admitted by the linearized governing equations have nonzero speed. Solitary waves are viewed as waves of finite amplitude that bifurcate from the quiescent state of the system with the wave speed playing the role of the bifurcation parameter. Evolution of the bifurcation diagram with respect to the pre-stretch is clarified. We then study the stability of solitary waves for a representative case that is likely of most interest in applications, the case in which solitary waves exist with speed c lying in the interval $$[0, c_1)$$ [ 0 , c 1 ) where $$c_1$$ c 1 is the bifurcation value of c, and the wave amplitude is a decreasing function of speed. It is shown that there exists an intermediate value $$c_0$$ c 0 in the above interval such that solitary waves are spectrally stable if their speed is greater than $$c_0$$ c 0 and unstable otherwise.
