Abstract Study question Is there a significant difference in the clinical results of embryos cultured in time-lapse systems with single-step medium and conventional benchtop incubators with sequential media? Summary answer Embryos cultured in time-lapse systems and single-step media are more likely to achieve an ongoing pregnancy and have higher implantation rates than those cultured otherwise. What is known already One of the strategies for embryo culture in IVF consisted in conventional benchtop incubators combined with sequential culture media (CI-Seq). New generation time-lapse systems provide useful information on the morphokinetics of embryo development, but also a stable culture environment where embryos can develop undisturbed until blastocyst stage when paired with single-step culture media (TLS-SS). These features have the potential to improve embryo development and selection. Nonetheless, there is inconclusive evidence of whether this new culture strategy has a significant effect on clinical results of ICSI treatments. Studies on the matter are heterogeneous and reduced in both number and sample size. Study design, size, duration Unicentric retrospective cohort study. We compared the results of 11471 blastocyst transferences from 10276 ICSI treatments performed during 4 consecutive years, where embryos were cultured either on CI with sequential media (N = 5255) or a TLS with single-step medium (N = 5021). 3922 of the totals were fresh embryo transfers (ET) and 7549 frozen-thawed ET. We compared the implantation rate (IR) and ongoing pregnancy rate (OGPR) in both study groups, stratifying by ovum origin. Participants/materials, setting, methods Three models of TLS were used for embryo culture: EmbryoScope, EmbryoScope Plus (Vitrolife) and GERI (Genea Biomedx), as well as one CI (ASTEC). Sequential media: Cook, Origio, Vitrolife; Single-step media: Gems, Irvine, Life Global. Embryo scoring and selection was performed by ASEBIR criteria in the CI group, and by morphological and morphokinetic assessment for embryos cultured in TLS. Embryos were extracted from the CI only for media change. Statistical analysis: ANOVA tests and Logistic regressions. Main results and the role of chance A general Logistic Regression was performed, including egg origin, PGT-A and culture strategy to explain their impact in OGPR. Egg origin (OR = 1,094 (95%CI: 1,015–1,179); P = 0,019) and culture strategy (OR = 1,141 (95%CI: 1,060–1,229); P < 0,001) were statistically significant, which confirms the need for stratification due to the heterogeneity of the groups. The total IR in the TLS-SS group was 54,68±48,84%, significantly higher than that of CI-Seq (49,18±47,91%; P < 0,001). In ovum-donation treatments, a complete Logistic Regression for OGPR, with all typical confounding variables (age, BMI, nº oocytes, fresh/frozen transfer, number and day of ET) resulted in an OR = 1,187 (95%CI: 1,074–1,313; P = 0,001) favoring culture in TL-SS. IR in these treatments were 61,98±47,68% in TL-SS vs 55,08±46,58% in CI-Seq (P < 0,001) in fresh transfers and 51,48±48,91% in TL-SS vs 44,39±47,67% in CI-Seq (P < 0,001) in frozen-thawed ET. In autologous treatments with PGT a similar regression yielded an OR = 1,055 (95%CI: 0,889–1,252; P = 0,542) for culture strategy. The IR of genetically tested ET was not significantly different: 53,08±49,49% for TL-SS, 50,90±49,07% for CI-Seq, P = 0,246. In autologous procedures without PGT, culture strategy was not significant for OGPR (OR = 0,998 (95%CI: 0,835–1,191), P = 0,979) nor IR of fresh (49,75±48,91% TL-SS vs 44,23±47,36% CI-Seq; P = 0,081) nor frozen-thawed transferences (50,77±48,33% TL-SS vs 50,67±47,33% CI-Seq; P = 0,970). Limitations, reasons for caution After fertilization check, embryos were evaluated exclusively on D5/6. On D3, embryos cultured in CI were taken out only for a quick media change, but not for evaluation, and all handling was done in isolette cabins with controlled environmental conditions. Being a retrospective study, there is high variability in population. Wider implications of the findings: A more homogenous prospective study, including comparison in life-birth rates, is necessary to extract final conclusions. However, our results suggest that the introduction of TLS and SS media in IVF laboratories might be a valid strategy to increase clinical results, especially in fresh embryo, thanks to an improved embryo selection. This paper explores individuals’ motives for using social media when living under ‘social distancing’ conditions imposed during the COVID-19 pandemic, where they were instructed to physically distance from other people. Adopting a ‘uses and gratifications’ approach, and using a previously established five-factor scale, we examine the relationship between individuals’ motives for using social media and their personality traits. Hundred and eighty-nine social media users living in the United Kingdom completed surveys assessing their motives for using social media and their personality. Our findings demonstrate that participants were generally motivated to use social media to ‘pass time’ and to ‘maintain relationships.’ Further, we find that those high in extraversion in particular use social media to ‘maintain relationships.’ By comparing our findings to previous studies where face-to-face interaction was not restricted, our findings indicate that individuals’ motives for using social media change when they are placed under physical distancing restrictions. We reflect on the potential application of our findings for others experiencing similar conditions, such as those working in remote locations, as well as the potential implications for living in a post-pandemic world with increased virtual ‘meetings’ using social media.

Despite having become more visible in popular and academic discourses over the last half decade, trans* selfies are not new. In this article, I examine an early set of trans* selfies featured in a sexploitation periodical published in the United States during the early 1960s. I show how numerous media, including bodies, clothing, cosmetics, photographs and magazines, produced a socio-technical environment through which trans* subjects composed alternative gender expressions and identities, formed intimate networks and created conditions of possibility for the eventual re-emergence of trans* selfies via digital social media platforms. Merging trans* theory with media ecology, I develop trans* media ecology as a conceptual frame from which to locate the always imbricated – but never complete – becoming of gendered bodies and media. Methodologically, trans* media ecology adopts three guiding principles: (1) genders are media, (2) genders depend on media and (3) genders and media change.

The aim of this work was to evaluate the performance of vertical subsurface flow treatment wetlands (VSSF TWs) for treating rural domestic wastewater when strategies such as bed depth reduction and media change are used in combination with bottom saturation. Two treatment wetland systems were implemented: normal (VF-N), with a bed depth of 1.0 m, and modified (VF-M), with a bed depth of 0.5 m and a bottom layer of natural zeolite. Schoenoplectus californicus was used as experimental plant. These two treatment systems were operated at a hydraulic loading rate of 120 mm/d in two phases. Phase I did not use bottom saturation, while Phase II involved a bottom saturation of the zeolite layer of the VF-M system. The results show that bed depth reduction did not have a significant effect (p > 0.05) in terms of organic matter, solids, and ammonium removal. Conversely, it had a significant influence (p < 0.05) on phosphate as well as a negative effect on pathogen removal. This influence could be explained by initial media capacity for phosphorus removal and filtration importance in the case of pathogens. Partial saturation only had a positive influence on total nitrogen removal. The addition of a bottom layer of natural zeolite showed no positive effect on nutrient removal. The plant showed adaptation and positive development in both VF-N and VF-M. The water balance showed that water loss was not influenced by bed depth reduction. Therefore, according to the previous results, a combination of the proposal modifications to VSSF TWs can be introduced for treating rural domestic wastewater.

The emergence of TV in the first half of the 20th century became one of the media for mass audiovisual communication, technologically extending the tradition of electric telegraph and radio. Initially, TV was considered a media for transmitting current  processes. With the introduction of video technology in the 1960s, TV began to capture live content and re-display videos. TV content production technologies have been radically changed by video editing, which has brought TV closer to the cinema. Technological changes in the analog era have had a greater impact on content production processes, and the digital era  sparked a revolution in content consumption. Technological changes in the pre-digital and digital era can be seen as progress, but at the same time raises the question of media perception, even survival, as the technologies of production and distribution of TV content and audience behavior change from time to time.

This study describes a successful protocol for establishing cell lines from the threatened Triturus cristatus in terms of collection, preparing, establishing, cryopreserving, thawing and quality checking. Different parameters such as media, media change, fresh vs. cryopreserved tissue and seeding density were tested to optimize culture conditions for this species. With fresh tissue, no considerable differences in the use of two different media were found, but with cryopreserved tissue, a combination of ITS (insulin/transferrin/selenite) and 2-mercaptoethanol had a positive effect on growth. Real-time measurements on the cell lines were used, for the first time in amphibian cells, to investigate the effect of different treatments such as media change with or without washing. Media change had a positive impact on the cells, whereas the effect was negative when combined with washing. It is concluded that establishment of cell lines is possible from the great crested newt, especially when using fresh tissue, but much more challenging if the tissue has been cryopreserved. Real-time measurement during cell culture is a useful tool to visualize the sensitivity of amphibian cells during different culture treatments.

The article is a review of current studies of emotion transformations under the influence of the Internet and “new media”. The main approaches to this problem, including cognitive research, are considered. It is proved that a significant limitation of the most popular approach in cultural studies, which recognizes considerable differences between the “old” and “new” media, but does not absolutize this opposition, is the tendency to large-scale generalizations — a direct combination of empirical observations and broad theoretical conclusions about the nature of neoliberal society and contemporary digital culture. An important means of overcoming these superficial generalizations is turning to cognitive research, for which it is fundamentally important to verify data and clarify the framework of their correctness, depending on different types of social interaction.
