Abstract Background Epigenetic reprogramming plays an important role in the occurrence, development, and prognosis of hepatocellular carcinoma (HCC). DNA methylation is a key epigenetic regulatory mechanism, and DNA methyltransferase 1 (DNMT1) is the major enzyme responsible for maintenance methylation. Nevertheless, the role and mechanism of DNMT1 in HCC remains poorly defined. Methods In the current study, we conducted pan-cancer analysis for DNMT1’s expression and prognosis using The Cancer Genome Atlas (TCGA) data set. We conducted gene Set Enrichment Analysis (GSEA) between high-and-low DNMT1 expression groups to identify DNMT1-related functional significance. We also investigated the relationship between DNMT1 expression and tumor immune microenvironment, including immune cell infiltration and the expression of immune checkpoints. Through a combination series of computer analyses (including expression analyses, correlation analyses, and survival analyses), the noncoding RNAs (ncRNAs) that contribute to the overexpression of DNMT1 were ultimately identified. Results We found that DNMT1 was upregulated in 16 types of human carcinoma including HCC, and DNMT1 might be a biomarker predicting unfavorable prognosis in HCC patients. DNMT1 mRNA expression was statistically associated with age, histological grade, and the level of serum AFP. Moreover, DNMT1 level was significantly and positively linked to tumor immune cell infiltration, immune cell biomarkers, and immune checkpoint expression. Meanwhile, Gene Set Enrichment Analysis (GSEA) revealed that high-DNMT1 expression was associated with epithelial mesenchymal transition (EMT), E2F target, G2M checkpoint, and inflammatory response. Finally, through a combination series of computer analyses the SNHG3/hsa-miR-148a-3p/DNMT1 axis was confirmed as the potential regulatory pathway in HCC. Conclusion SNHG3/miR-148a-3p axis upregulation of DNMT1 may be related to poor outcome, tumor immune infiltration, and regulated malignant properties in HCC.

Unreinforced masonry walls (URM) in old buildings are vulnerable to collapse upon receiving high lateral inertia force due to earthquakes. However, a high strength fiber material such as Kevlar fiber has been identified as able to improve the URM-Wall performance, especially in shear strength enhancement. In this research, the seismic performance of an URM-Wall was analysed using SAP2000 by modeling the wall with three dimensional solid elements. Solids and springs as link connectors were assigned to represent the masonry behavior. The aim of this research is to compare between results obtained from the computer analyses and the previously conducted laboratory experiments. The effectiveness of Kevlar material installed in the diagonal directions of both wall surfaces was investigated. It was found that the failure mechanism shown by the SAP2000 model is similar to the laboratory test results.

Little is known about how play affects the development of children with a chronic condition. Studying play poses major methodological challenges in measuring differences in play behaviour, which results in a relative scarcity of research on this subject. This pilot study seeks to provide novel directions for research in this area. The effectiveness of a play- and sports-based cognitive behavioural programme for children (8–12 years) with a chronic condition was studied. The children and parents completed a battery of measurement tools before and after the programme. Moreover, the application of automated computer analyses of behaviour was piloted. Behaviour (Child Behavior Checklist) seemed to be positively affected by the programme. An increase in psychological well-being was observed (KIDSCREEN). Perceived competence (Self-Perception Profile for Children) and actual motor competence (Canadian Agility and Movement Skill Assessment) did not show any positive trends. These results of 13 participants suggest that children might learn to better cope with their illness by stimulating play behaviour. For the analysis of the effectiveness of programmes like this, we therefore propose to focus on measuring behaviour and quality of life. In addition, pilot measurements showed that automated analysis of play can provide important insights into the participation of children.

In this paper bio-tissue mathematical modeling serves as a central repository to interface design, simulation, and tissue fabrication. Finite element computer analyses will be used to study the role of local tissue mechanics on endochondral ossification patterns, skeletal morphology and mandible thickness distributions using single and multi-phase continuum material representations of clinical cases of patients implanted with the traditional protocols. New protocols will be hypothesized for the use of the new biologically techno-structured hybrid materials.

Abstract This article deals with computer analyses of output quantities of a railway vehicle depending on changing of parameters of suspension system. A passenger car was chosen for dynamic analyses. An analysed passenger railway vehicle uses two stage suspension system composed of coil springs and hydraulic dampers. Stiffness of coil springs of primary and secondary suspensions were defined for two states and its influence on output values in terms of quality and quantity was evaluated. As output variables, values of forces in a wheel/rail contact and accelerations in several locations on a wagon body floor were chosen. Values of forces in a wheel/rail contact indicate dynamic response of a railway vehicle running in terms of running safety and values of accelerations serve as important input for evaluation of passenger ride comfort.

Nearly 25% of incoming UT-Austin students are unable to get their first two choices for a college major. Historically, these students have been given an extensive list of all potential majors from which to choose. Many students simply lack awareness of the various majors and have no background knowledge that could be helpful in determining whether specific majors would suit their interests or skills.The purpose of this project was to rely on students’ admissions essays to provide a more coherent and tailored set of recommendations when students are selecting an alternative college major. The logic underlying this project is based on decades of empirical research demonstrating that psychological information can be extracted from the language of students’ admissions essays via automated computer analyses. The results of these analyses can then be used to inform the “college major options” webpage so that potential majors most closely aligned with their interests and skills will be displayed first in a recommendation system.

The aim of this study was to compare the levels of nesfatin-1 in healthy subjects with those in prediabetic and diabetic patients who have different glucose tolerance levels. Overall, 100 subjects were divided into 5 groups healthy control (C), impaired fasting glycemia (IFG), impaired glucose tolerance (IGT), metabolic syndrome (MS) and type 2 diabetes mellitus, (Type 2 DM). Glycated hemoglobin (HbA1c) assessed the glycemic control. Homeostasis model assessment of insulin resistance (HOMA-IR) was determined using computer analyses. Nesfatin-1 levels were measured using ELISA method. IFG and IGT (prediabetic groups) from MS and Type 2 DM (diabetic groups) differed significantly in HOMA-IR. The nesfatin-1 levels were lower, although not statistically significant, in IFG (0.937±0.03 ng/ml, p=0.07) and IGT (1.039±0.06 ng/ml, p=0.5) groups compared to healthy subjects (1.094±0.07 ng/ml). However, the nesfatin-1 levels were lower in patients with Type 2 DM (0.867±0.02 ng/ml, p=0.007) and MS (0.885±0.01 ng/ml, p=0.01) compared to healthy subjects. Nesfatin-1 levels were significantly lower in diabetic patients compared to healthy subjects. This study supports the role of insulin resistance in decreased nesfatin-1 levels in patients with Type 2 DM and MS.
