Background: Little is known about the use of an electronic reminder system for HCV screening among patients with kidney disease. In this study, we tried to determine whether reminder systems could improve the HCV screening rate in patients with kidney disease. Methods: Patients with kidney disease were enrolled from August 2019 to December 2020 to automatically screen and order HCV antibody and RNA testing in outpatient departments. Results: A total of 19,316 outpatients with kidney disease were included, and the mean age was 66.5 years. The assessment rate of HCV antibody increased from 53.1% prior to the reminder system to 79.8% after the reminder system (p < 0.001), and the assessment rate of HCV RNA increased from 71% to 82.9%. The anti-HCV seropositivity rate decreased from 7.3% at baseline to 2.5% after the implementation of the reminder system (p < 0.001), and the percentage of patients with detectable HCV RNA among those with anti-HCV seropositivity decreased from 69.1% at baseline to 46.8% (p < 0.001). Conclusions: The feasibility of an electronic reminder system for HCV screening among patients with kidney disease in a hospital-based setting was demonstrated.

This paper describes the automatic speech recognition (ASR) systems built by the MLLP-VRAIN research group of Universitat Politècnica de València for the Albayzín-RTVE 2020 Speech-to-Text Challenge, and includes an extension of the work consisting of building and evaluating equivalent systems under the closed data conditions from the 2018 challenge. The primary system (p-streaming_1500ms_nlt) was a hybrid ASR system using streaming one-pass decoding with a context window of 1.5 seconds. This system achieved 16.0% WER on the test-2020 set. We also submitted three contrastive systems. From these, we highlight the system c2-streaming_600ms_t which, following a similar configuration as the primary system with a smaller context window of 0.6 s, scored 16.9% WER points on the same test set, with a measured empirical latency of 0.81 ± 0.09 s (mean ± stdev). That is, we obtained state-of-the-art latencies for high-quality automatic live captioning with a small WER degradation of 6% relative. As an extension, the equivalent closed-condition systems obtained 23.3% WER and 23.5% WER, respectively. When evaluated with an unconstrained language model, we obtained 19.9% WER and 20.4% WER; i.e., not far behind the top-performing systems with only 5% of the full acoustic data and with the extra ability of being streaming-capable. Indeed, all of these streaming systems could be put into production environments for automatic captioning of live media streams.

This protocol details steps to extract protein from Gram-negative bacterial or fungal cells (that have been pretreated with zymolyase) in quantitative proteomic workflows by using a Biomek FX liquid handler system. It is a semi-automated protocol that includes several 'pause' steps for centrifugation steps that are conducted manually "off-deck". This protocol works best as part of an automated proteomic sample preparation workflow with: Automated Protein Quantitation with the Biomek-FX liquid handler system and Automated Protein Normalization and Tryptic Digestion on a Biomek-NX Liquid Handler System

Intracellular trafficking through the secretory organelles depends on transient interactions between cargo proteins and transport machinery. Cytosolic coat protein complexes capture specific luminal cargo proteins for incorporation into transport vesicles by interacting with them indirectly through a transmembrane adaptor or cargo receptor. Due to their transient nature, it is difficult to study these specific ternary protein interactions just using conventional native co-immunoprecipitation. To overcome this technical challenge, we have applied a crosslinking assay to stabilize the transient and/or weak protein interactions. Here, we describe a protocol of protein cross-linking and co-immunoprecipitation, which was employed to prove the indirect interaction in the endoplasmic reticulum of a luminal secretory protein with a selective subunit of the cytosolic COPII coat through a specific transmembrane cargo receptor. This method can be extended to address other transient ternary interactions between cytosolic proteins and luminal or extracellular proteins through a transmembrane receptor within the endomembrane system.
