The coupling of the microfluidic chip to mass spectrometry (MS) has attracted considerable attention in the area of chemical and biological analysis. The most commonly used ionization technique in the chip–MS system is electrospray ionization (ESI). Traditional chip-based ESI devices mainly employ direct electrical contact between the electrode and the spray solvent. In this study, a microchip ESI source based on a novel polarization-splitting approach was developed. Specifically, the droplet in the microchannel is first polarized by the electric field and then split into two sub-droplets. In this process, the charge generated by polarization is retained in the liquid, resulting in the generation of two charged droplets with opposite polarities. Finally, when these charged droplets reach the emitter, the electrospray process is initiated and both positive and negative ions are formed from the same solution. Preliminary experimental results indicate that the coupling of this polarization-splitting ESI (PS-ESI) chip with a mass spectrometer enables conventional ESI-MS analysis of various analytes.

Aim: To improve the killing efficacy of head and neck squamous cells (SAS) by boric acid-mediated boron neutron capture therapy (BNCT). Materials & methods: Boric acid-containing chitosan/alginate/polyvinyl alcohol nanoparticles (B-capNPs) were manufactured using the nano-electrospray process. Results: Less than 10% of the boric acid leaked from the B-capNPs over 2 days. The B-capNPs killed up to 2.8-fold more SAS cells and reduced cytotoxicity tenfold when compared with pure boric acid alone. B-capNPs show selective uptake in tumor cells with tumor/normal ratios of SAS to normal (NIH 3T3) and macrophage (RAW 264.7) cells of 4.0 and 3.5, respectively, which are greater than the minimum acceptable tumor/normal ratio for BNCT of 2.5. Conclusion: These findings illustrate that B-capNPs may be more superior as BNCT drugs than pure boric acid.

A neurodegenerative disorder, glaucoma is a leading cause of blindness in the world. The conventional treatment strategies do not allow the significant penetration of the drug in the cornea. Therefore, we prepare a brinzolamide (Brz) loaded core-shell nanoparticles (NPs) to enhance the coronial penetration of the drug and thus treating the glaucoma. The shell of the NPs was composed of phosphatidylserine (PS; 1,2-diacyl-sn-glycero-3-phospho-L-serine), whereas the core of the NPs contains the Brz encapsulated in brinzolamide–phosphatidylserine–polymer poly-(DL-lactic acid-co-glycolic acid)–phosphatidylserine (Brz-PS-PLGA). The synthesis of Brz-PS-PLGA was achieved by using a coaxial electrospray process (CEP), which allows the preparation of the particles in a single step. The size of Brz-PS-PLGA with PS shell and brinzolamide–poly (lactic-co-glycolic) acid (Brz-PLGA) without shell was 571 ± 27.02 nm and 456 ± 19.17 nm, respectively. The charges on the surface of Brz-PS-PLGA and Brz-PLGA were (-) 27.45 ± 2.98 mV and (-) 19.47 ± 2.83 mV. The transmission electron microscopy images clearly reveal the PS shell as a light black layer over the dark black PLGA core. The CEP allows the high encapsulation of Brz in Brz-PS-PLGA where percentage of entrapment efficiency for Brz-PS-PLGA was 88.13 ± 6.43%. The release study conducted in a simulated tear fluid revealed the sustained release patterns of Brz from Brz-PS-PLGA and these were nontoxic to the cells as revealed by the cytotoxicity studies. Further, the Brz-PS-PLGA enhanced the coronial penetration of Brz and was capable of significantly reducing the intraocular pressure (IOP) after administration to the rabbit eye in comparison to the Brz-PLGA and free Brz. The results clearly suggest that the PS coating significantly enhances the capability of the particles in reducing IOP.

AbstractYeast cells patterned by pulsed jet electrospray showed a high alcoholic fermentation rate. Multi-dimensional patterns of individual yeast cells were produced by varying the experimental parameters of the electrospray system. The electrospray process, which employed a vibrational electric field, could control patterns of viable yeast cells at a cellular resolution. This novel system for electrospraying viable cells can be applied to biological process engineering including whole cell biochip techniques and micro fermentation processes for biochemical studies.

AbstractAn investigation on the droplet characteristics of ethanol in small-scale combustors with two different systems was conducted experimentally and theoretically. The classical capillary-mesh electrode arrangement was applied in Type A electrospray system, and for Type B, an additional ring electrode is included. The droplet size and velocity were measured by a Phase Doppler Anemometer. The electric filed intensity was theoretically calculated in the two electrospray systems. Compared with Type A, Type B system has smaller droplet size and velocity in the same spraying mode. Meanwhile the electrospray process in Type B system is more stable than that in Type A with its smaller root mean square velocity. By measuring the spraying current, the average specific charge of the droplets for the two systems was obtained in different spraying modes. And it was found that the addition of the ring electrode can help to increase the droplet charge, which is the fundamental reason for Type B electrospray system to perform better. The corona charge of the droplets was theoretically calculated for the two electrospray systems. It was found that the calculated specific charge generated by corona charging was in good agreement with the experimental results.
