Both inhibitory and excitatory GABA transmission exist in the mature suprachiasmatic nucleus (SCN), the master pacemaker of circadian physiology. Whether GABA is inhibitory or excitatory depends on the intracellular chloride concentration ([Cl−]i). Here, using the genetically encoded ratiometric probe Cl-Sensor, we investigated [Cl−]i in AVP and VIP-expressing SCN neurons for several days in culture. The chloride ratio (RCl) demonstrated circadian rhythmicity in AVP + neurons and VIP + neurons, but was not detected in GFAP + astrocytes. RCl peaked between ZT 7 and ZT 8 in both AVP + and VIP + neurons. RCl rhythmicity was not dependent on the activity of several transmembrane chloride carriers, action potential generation, or the L-type voltage-gated calcium channels, but was sensitive to GABA antagonists. We conclude that [Cl−]i is under circadian regulation in both AVP + and VIP + neurons.

Abstract Striking colorimetric probe (CynH) for abrupt detection of hydrazine under complete aqueous solution was achieved. The water soluble probe was designed with electron “push-pull” strategy by coupling of 4-hydroxy benzaldehyde and 2, 3, 3-trimethylindolinine. The positively charged N-propylated indolinine make the probe completely soluble in water. The probe yields eye catching selective detection of hydrazine over other competing analytes with high sensitivity. Obvious colour change was observed from colourless to appearance of bright pink colour with hydrazine. It reacts quickly with hydrazine within 2 minutes and makes the probe an effective candidate for practical application. The real time application was demonstrated using paper strip to detect hydrazine vapour. This probe is superior to earlier reported probes because of its effective sensing of hydrazine displayed with various applications including real-time strip based sensing, spray test and soil analysis. In all the examinations, the probe yields distinct response with rapid naked eye colour change this overcomes the drawbacks of previous reports.

The use of carbon dots (CDs) with dual emission based on ratiometric fluorescence has been attracting attention in recent times for more accurate ion detection since they help avoid interference from background noise, probe concentration, and complexity. Herein, novel dual-emission nitrogen-doped CDs (NCDs) were prepared by a simple method for Cu2+ and ClO- detection. The NCDs showed excellent anti-interference ability and selectivity for different emissions. In addition, a good linear relationship was observed between the fluorescence intensity (FI) of the NCD solutions in different emissions with Cu2+ (0–90 μM) and ClO- (0–75 μM). The limits of both Cu2+ detection and ClO- were very low, at 17.7 and 11.6 nM, respectively. The NCDs developed herein also showed a good recovery rate in water for Cu2+ and ClO− detection. Hence, they are expected to have a more extensive application prospect in real samples.
