Immunohistochemistry (IHC) using mouse retinal cryosections is widely used to study the expression and intracellular localization of proteins in mouse retinas. Conventionally, the preparation of retinal cryosections from mice involves tissue fixation, cryoprotection, the removal of the cornea and lens, embedding and sectioning. The procedure takes 1–2 days to complete. Recently, we developed a new technique for the preparation of murine retinal cryosections by coating the sclera with a layer of Super Glue. This enables us to remove the cornea and extract the lens from the unfixed murine eye without causing the eyecup to collapse. In the present study, based on this new technique, we move a step forward to modify the conventional protocol. Unlike in the conventional protocol, in this method, we first coat the unfixed mouse eyeball on the sclera with Super Glue and then remove the cornea and lens. The eyecup is then fixed, cryoprotected and sectioned. This new protocol for the preparation of retinal cryosections reduces the time for the procedure to as little as 2 h. Importantly, the new protocol consistently improves the morphology of retinal sections as well as the image quality of IHC. Thus, this new quick protocol will be greatly beneficial to the community of visual sciences by expediting research progress and improving the results of IHC.

Immunohistochemistry using mouse retinal cryosections is a routine assay used in vision research. However, retinal tissues are fragile, and it is difficult to obtain an ideal retinal cryosection. Here, we developed a modified method for preparing retinal cryosection. Super Glue was applied on the surface of the sclera before the cornea and the lens are removed from either the unfixed or PFA-fixed mouse eyeballs. The new methods largely prevented retinal detachment in mouse retinal cryosections. Immunostaining of retinal cryosections derived from PFA-fixed mouse eyes using rod and cone markers yielded high-quality immunofluorescent images. Immunolabeling of retinal cryosections obtained from unfixed mouse eyes using a cilium-specific marker had improved orientations of photoreceptor connecting cilia. This new method substantially improves the morphology and immunostaining results of fixed and unfixed mouse eyes.

Pendahulan: Luka bakar kimia mata, luka bakar termal, dan radiasi mewakili kasus-kasus yang sering terjadi pada kasus di unit gawat darurat pada bidang mata. Paparan terhadap cairan tubuh dan cyanoacrylates (super glue) juga sering terjadi. Keadaan-keadaan tersebut jika tidak ditindaklanjuti dengan cepat akan mengakibatkan prognosis yang buruk. Tujuan: Untuk mengetahui diagnosis dan tatalaksana awal pada luka bakar mata. Metode: Artikel ini dibuat dengan metode literature review, melibatkan 22 pustaka baik buku dan jurnal nasional atau internasional. Hasil: Luka bakar oleh bahan kimia mewakili cedera mata yang paling sering dan berpotensi membuat buta, hal ini merupakan keadaan darurat mata yang membutuhkan penanganan segera dan memulai pengobatan. Mayoritas korban masih muda dan paparan bisa terjadi di rumah, tempat kerja dan dapat juga ada hubungannya dengan tindak kriminal. Pembahasan: Cedera alkali terjadi lebih sering daripada cedera asam. Cedera kimia pada mata menghasilkan kerusakan luas pada epitel permukaan mata, kornea, segmen anterior, dan sel batang yang mengakibatkan kerusakan penglihatan permanen unilateral atau bilateral. Tatalaksana yang cepat dan tepat mungkin merupakan faktor terpenting dalam menentukan hasil akhir. Simpulan: Artikel ini mengulas manajemen darurat dan teknik-teknik baru untuk meningkatkan prognosis pasien dengan cedera luka bakar pada mata.   Kata kunci: Alkali, asam, luka bakar kimia, luka bakar termal, mata, radiasi.

The purpose of this article is to highlight the importance of religious nationalism to protect vital national interests, maintain stability and preservation of core values. The expression of the constitution of Indonesia is a guarantor in integrating the people from all groups and strata of Indonesian society. It ensures a favorable internal and external condition. Religious nationalism is super glue that bonds the people, religions and the nation as a single entity. This is still utmost relevant in the pandemic.

A rare case of cyanoacrylate urine bladder urolithiasis in a 60-year-old male is presented. The application of surgical glue (Glubran) as treatment of seroma one month after laparoscopic inguinal hernioplasty led to the instillation of the n-butyl cyanoacrylate into the bladder resulting in the formation of a concretion. Infrared spectroscopy of the urine stone removed by cystoscopic laser lithotripsy four months after the surgery allowed the identification of the nature of the stone and revealed cyanoacrylate as the major component and co-monomer methacryloxy sulfolane as the minor component. Polypropylene from the mesh was not detected.

The objective of this study was to prepare the skeletons of duck to enhance veterinary anatomy teaching. Five aged ducks were used in this experiment, which was collected from the Bangladesh Agricultural University poultry farm. The ducks were skinned and different segments of the body were dissected with the help of a scalpel. Then the bones were wrapped using merkin clothes, heated in 5% solution of soda water for 1 hour, and kept in that solution for 7 days for decomposing the muscles, tendon and ligaments. For proper cleaning, the bones were dipped into 10% bleaching water for 2 hours. After complete sundry, the bones were articulated using super glue for rising into a skeleton. The vertebral column was aligned with 25-gauge steel wire was inserted into the vertebral foramen. Finally, the skeletons were fixed on a wooden stand. Therefore, the skeletons were preserved and used for demonstration of osteology, and avian anatomy. This duck skeleton will be used to study avian morphology and make a comparative analysis with other mammals. Asian J. Med. Biol. Res. June 2019, 5(3): 192-196
