Abstract Purpose To investigate the nature of the materials used in the adhesive layer of the Imago Pietatis painting (end of the fifteenth century—beginning of the sixteenth century) by Domenico Morone as a prerequisite for its restoration. Methods Micro-FTIR spectra of the animal glue and a polished cross-section were acquired by a Jasco IRT3000 spectrometer, equipped with a 32× Cassegrain objective. A dot blot immunoassay was used to characterise a minor component of the adhesive layer. Results Micro-FTIR was used as an effective diagnostic tool to detect the major component of the adhesive layer and the binder of the paint. Despite the ageing, the complex matrix and the micro-size of the sample, using a dot blot immunoassay, it was possible to quantify 3.7 ± 2.0 ng of ovalbumin per microgram of sample (corresponding to 0.004 ± 0.002% of the weight). Conclusions The findings were in line with conservation practices described in the old treatises, confirming the correct interpretation of the adhesive layer compounds added to the painting and suggesting for the cleaning the use of an anionic water-soluble surfactant highly effective in the removing of proteinaceous materials.

Abstract Background The species composition of cattle gastrointestinal nematode (GIN) communities can vary greatly between regions. Despite this, there is remarkably little large-scale surveillance data for cattle GIN species which is due, at least in part, to a lack of scalable diagnostic tools. This lack of regional GIN species-level data represents a major knowledge gap for evidence-based parasite management and assessing the status and impact of factors such as climate change and anthelmintic drug resistance. Methods This paper presents a large-scale survey of GIN in beef herds across western Canada using ITS-2 rDNA nemabiome metabarcoding. Individual fecal samples were collected from 6 to 20 randomly selected heifers (n = 1665) from each of 85 herds between September 2016 and February 2017 and 10–25 first season calves (n = 824) from each of 42 herds between November 2016 and February 2017. Results Gastrointestinal nematode communities in heifers and calves were similar in Alberta and Saskatchewan, with Ostertagia ostertagi and Cooperia oncophora being the predominant GIN species in all herds consistent with previous studies. However, in Manitoba, Cooperia punctata was the predominant species overall and the most abundant GIN species in calves from 4/8 beef herds. Conclusions This study revealed a marked regional heterogeneity of GIN species in grazing beef herds in western Canada. The predominance of C. punctata in Manitoba is unexpected, as although this parasite is often the predominant cattle GIN species in more southerly latitudes, it is generally only a minor component of cattle GIN communities in northern temperate regions. We hypothesize that the unexpected predominance of C. punctata at such a northerly latitude represents a range expansion, likely associated with changes in climate, anthelmintic use, management, and/or animal movement. Whatever the cause, these results are of practical concern since C. punctata is more pathogenic than C. oncophora, the Cooperia species that typically predominates in cooler temperate regions. Finally, this study illustrates the value of ITS-2 rDNA nemabiome metabarcoding as a surveillance tool for ruminant GIN parasites. Graphical Abstract

The manuscript attempts to understand the evolution of NOx precursors: NH3 and HCN from Pyrolysis of furfural residue (FR). The pyrolysis process was carried out in a thermogravimetric analyzer (TGA) coupled to Fourier-transform infrared (FTIR) spectrometer. The combination revealed insightful information on the evolution of NH3 and HCN. This could help us better understand the characteristics of FR derived from furfural production especially with regard to NH3 and HCN. Nitrogen is considered a minor component in biomass wastes; in this study nitrogen content is about 0.57%. However, the pollution potential poised by low nitrogen content is huge through both direct and indirect processes. Thus, this study presents results that were found with regard to FR pyrolysis in pure nitrogen environment. At the heating rate of 40°C/min−1, the only NOx precursor detected was HCN at 713 cm−1 as per the database provided by National Institute of Standards and Technology (NIST). NH3 was not detected. The particle size of FR used ranged between 0.15–0.25 mm.

Morphological transition between hexagonal and lamellar patterns in thin polystyrene–block–poly(4-vinyl pyridine) films simultaneously exposed to a strong in-plane electric field and saturated solvent vapor is studied with atomic force and scanning electron microscopy. In these conditions, standing cylinders made of 4-vinyl pyridine blocks arrange into threads up to tens of microns long along the field direction and then partially merge into standing lamellas. In the course of rearrangement, the copolymer remains strongly segregated, with the minor component domains keeping connectivity between the film surfaces. The ordering tendency becomes more pronounced if the cylinders are doped with Au nanorods, which can increase their dielectric permittivity. Non-selective chloroform vapor works particularly well, though it causes partial etching of the indium tin oxide cathode. On the contrary, 1,4-dioxane vapor selective to polystyrene matrix does not allow for any morphological changes.

Abstract Background The microbiome in plant-soil systems has a significant influence in promoting plant growth. The extent of selectivity that the plant exerts on the microbiome in the continuum of internal plant tissues is not well understood. This study analysed the root microbiome of a legume, Melilotus officinalis (L.) Pall., sweet clover, and focused on dynamic shifts in the microbial community structure through the niches of bulk soil, rhizosphere, periderm, phloem and xylem, and further examined the effects of environmental factors, root exudates and root cell wall development on the microbiome assemblages in different root compartments.ResultsYoung and mature plants were sampled at 24 field sites and the microbial communities in different niches from bulk soil and rhizosphere through to root compartments were analysed by 16S rRNA gene sequencing. The microbiome composition changed from periderm to phloem to a greater extent than across other boundaries (0.95 vs 0.71 based Bray–Curtis distance). Variation in microbiome composition was associated with geographic distance and soil properties for the bulk soil, rhizosphere and periderm niches. The composition of root exudate compounds were correlated with the rhizosphere microbiome assemblages in mature and young plants. The endophyte communities that occupied the phloem and xylem were most conserved and were independent of growing environments and root exudation. Symbiotic rhizobia able to nodulate M. officinalis were prominent colonisers of the periderm (~15%) and xylem (~6.2%), but were only a minor component in other soil-related niches (0.1%-2.5%). In xylem tissues, endophyte diversity was correlated with the total cell wall and lignin content across the sampled sites (r=0.29-0.62). Conclusions Our results demonstrate that selection of microbiome constituents occurs at different boundaries through bulk soil, rhizosphere, periderm, phloem and xylem, and is especially strong across the periderm boundary. The conserved endophyte community in the innermost tissues (phloem and xylem) was identified, and will be advantageous to the development of specific beneficial microbial inoculants.

Caulerpa lentillifera is a type of green seaweed that is cultivated in tropical and subtropical areas. The objectives of this study were to determine the characteristics of the sulfated polysaccharides from C. lentillifera and evaluate its antidiabetic activity. In the initial process of this study, samples were macerated with ethanol (1:10). Then, the maceration residue was extracted with an accumulator at 75 °C for three hours. The crude extract yield was 4.16% based on weight seaweed. Ion chromatography purification with DEAE-Sepharose resin provided a yield of 14.8% of crude extract. The monomer analysis of C. lentillifera from the crude extract and purified extract revealed that galactose monomers were dominant and glucose was a minor component. The total carbohydrate and sulfate contents of purified C. lentillifera were higher than those of crude C. lentillifera. Bioactivity tests revealed that purified polysaccharides had higher antidiabetic activity against α-glucosidase enzyme than crude ones with IC50 values of 134.81± 2.0 µg/mL. Purified sulfated polysaccharides of C. lentillifera could potentially be used as an antidiabetic medication.

Communal roosting by urban birds, such as crows, mynas, and starlings, can be a public nuisance due to excessive noise and fouling of the surroundings with droppings. Food availability within proximity to the roosting area is one of the key factors influencing roosting site preference and fidelity. The diets of roosting mynas and crows have been well-studied, yet little is known about the diet of the Asian glossy starling (AGS), Aplonis panayensis. This study focused on assessing the diet of roosting AGS and food resource availability around the roosting area. The AGS diet was assessed through microscopic analysis of stomach contents and droppings. The diet mainly consisted of fruits (76%) with a minor component of animal materials, i.e. ants, snails, and beetle larvae. Intact seeds found in the samples were identified using DNA barcoding. Seven out of the nine plant species detected were new records for the AGS diet. The most common fruit found in the samples was Trema orientalis, which grows extensively along roadsides within foraging areas of AGS. The availability of fruits of different fast-growing pioneer species around the roosting site ensured a continuous supply of food to the birds. Animal materials, which were consumed by chance during foraging, supplemented the fruit in the bird’s diet. Hence, the birds’ preference for the roosting site may have been influenced by the availability of various food resources around it.

Eosinophils are a minor component of circulating granulocytes, which are classically viewed as end-stage effector cells in host defense against helminth infection and promoting allergic responses. However, a growing body of evidence has emerged showing that eosinophils are versatile leukocytes acting as an orchestrator in the resolution of inflammation. Rheumatoid arthritis (RA) is the most common chronic inflammatory disease characterized by persistent synovitis that hardly resolves spontaneously. Noteworthy, a specific population of eosinophils, that is, regulatory eosinophils (rEos), was identified in the synovium of RA patients, especially in disease remission. Mechanistically, the rEos in the synovium display a unique pro-resolving signature that is distinct from their counterpart in the lung. Herein, we summarize the latest understanding of eosinophils and their emerging role in promoting the resolution of arthritis. This knowledge is crucial to the design of new approaches to rebalancing immune homeostasis in RA, considering that current therapies are centered on inhibiting pro-inflammatory cytokines and mediators rather than fostering the resolution of inflammation.

Abstract. Coccolithophores are potentially affected by ongoing ocean acidification, where rising CO2 lowers seawater pH and calcite saturation state (Ωcal). Southern Patagonian fjords and channels provide natural laboratories for studying these issues due to high variability in physical and chemical conditions. We surveyed coccolithophore assemblages in Patagonian fjords during late spring 2015 and early spring 2017. Surface Ωcal exhibited large variations driven mostly by freshwater inputs. High-Ωcal conditions (max. 3.6) occurred in the Archipelago Madre de Dios. Ωcal ranged from 2.0–2.6 in the western Strait of Magellan and 1.5–2.2 in the inner channel and was subsaturating (0.5) in Skyring Sound. Emiliania huxleyi was the only coccolithophore widely distributed in Patagonian fjords (> 96 % of total coccolithophores), only disappearing in the Skyring Sound, a semi-closed mesohaline system. Correspondence analysis associated higher E. huxleyi biomasses with lower diatom biomasses. The highest E. huxleyi abundances in Patagonia were in the lower range of those reported in Norwegian fjords. Predominant morphotypes were distinct from those previously documented in nearby oceans but similar to those of Norwegian fjords. Moderately calcified forms of E. huxleyi A morphotype were uniformly distributed throughout Patagonia fjords. The exceptional R/hyper-calcified coccoliths, associated with low Ωcal values in Chilean and Peruvian coastal upwellings, were a minor component associated with high Ωcal levels in Patagonia. Outlying mean index (OMI) niche analysis suggested that pH and Ωcal conditions explained most variation in the realized niches of E. huxleyi morphotypes. The moderately calcified A morphotype exhibited the widest niche breadth (generalist), while the R/hyper-calcified morphotype exhibited a more restricted realized niche (specialist). Nevertheless, when considering an expanded sampling domain, including nearby southeast Pacific coastal and offshore waters, even the R/hyper-calcified morphotype exhibited a higher niche breadth than other closely phylogenetically related coccolithophore species. The occurrence of E. huxleyi in naturally low pH–Ωcal environments indicates that its ecological response is plastic and capable of adaptation.
