Esse trabalho teve o objetivo de realizar um monitoramento bacteriológico da água de um poço artesiano localizado na zona urbana da cidade de Remígio-PB. O estudo foi desenvolvido com amostras de água coletadas num poço artesiano localizado na zona urbana da cidade de Remígio-PB. As amostras de água foram coletadas diretamente do poço artesiano, em garrafas de vidro (500 ml) com boca larga, protegidas com papel laminado, previamente esterilizadas em autoclave a 121 oC, por 30 minutos, e foram encaminhadas para o Laboratório do Centro de Formação Profissional do Instituto Albano Franco de Tecnologia de Couro e de Calçado (CTCC) em Campina Grande. Os parâmetros analisados foram: coliformes totais, coliformes termotolerantes, Escherichi coli e Bactérias Heterotróficas. Observou-se que as amostras analisadas apresentaram quantidade de coliformes totais, coliformes termotolerantes e bactérias heterotróficas totalmente fora dos limites estabelecidos pela portaria vigente. Não se detectou a presença de E. coli. Conclui-se que todas as amostras estão em desacordo com as recomendações estipuladas pela Portaria do Ministério da Saúde de nº 5, de 28 de setembro de 2017, pois apresentaram coliformes totais, coliformes termotolerantes e Bactérias heterotróficas, necessitando, portanto, de melhorias significativas no que se refere a proteção da fonte e de tratamento prévio antes de serem fornecidas para consumo humano.

Sanitization by pH adjustment of dairy and pig slurries was evaluated for potential use as organic fertilizer in horticulture. This requires absence of Salmonella in 25 g of slurry and less than 1000 Escherichi coli colony-forming unit per gram of fresh slurry (Regulation (EU) 2019/1009). Additives used in the alkalinization and acidification treatments, included hydroxide-salts and nitrogen-based reactants to increase slurry pH to a basic range (9.0–11.0) and concentrated H2SO4 to decrease slurry pH to an acidic range (5.5–3.5). While low-cost urea was unable to increase the slurry pH above 9.5, ammonia efficiently increased slurry pH to the targeted values (but enhanced the emissions risk), whereas the effect of Ca(OH)2 was hindered by its low solubility. Slurry sanitization by alkalinization was achieved at a pH of 9.5 for both slurries, using similar quantities of KOH or NaOH. KOH was selected for further tests since it provides a plant macronutrient. Acidification with concentrated H2SO4 was able to achieve sanitization by lowering the pH to 5.0. After a 60-d storage experiment with raw and treated slurries, the level of E. coli was below the sanitization limit for all samples. Storage had no significant impact on slurry characteristics, except for ammonium-nitrogen content. Acidification treatment minimized ammonia losses.

A concurrent and promising effects has been achieved by using cow urine as a efficient antibacterial activity against gram-negative and gram-positive human pathogen. The present study aims to evaluate anti-microbial activity photo activated cow urine against pathogenic bacterial strains. It is estimated by agar well diffusion method.It is evident from the data presented that the zone of inhibition of photo activated Gir cow urine and Khillari cow urine against bacteria Escherichi coli and staphulococcus aureus is significant.The present investigation clearly demonstrate that the average width of inhibition zone at 50 micrilitre dose was 1.6cm.Therefore this pathogen can be well managed by using cow urine.

Silk sericin composed of 18 amino acids has been widely used in the fields of cosmetic additives, food, medicine and functional biomaterials because of good hydrophilicity and biocompatibility, making it great possibility in providing abundant nutrients for microbial growth. Sericin (40~200 KDa) was used as culture medium for incubation of E. coli at 37°C to study the effect of sericin concentration on the growth of bacterial Escherichi coli (E. coli). The growth curves of E. coli, surface/inside morphology and protein of E. coli were investigated by UV/vis spectrophotometer (UV/vis), scanning electron microscopy (SEM), transmission electronic microscopy (TEM) and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The cytotoxicity of sercin was also confirmed by MTT assay. The value of OD600 increases with increasing sericin concentration from 0 to 40 g/L. Compared with the control, OD600 of 40 g/L sericin medium increases from 0.013 to 1.269 after incubated 12h. E. coli cell still remains rod shape regardless of concentration of sericin. The content of cellular soluble proteins significantly increases in sericin-treated bacteria, which in turn influenced the cell structure composition and catalyzing activity of enzyme, and finally stimulated the proliferation of E. coli. Results indicate that sericin can independently provide carbon and nitrogen for bacterial growth. Besides, it can promote bacterial protein expression without affecting cell morphology.
