Two steroid compounds, ergosterol peroxide (1) and stigmasterol (2) have been isolated from the stembark of Aglaia simplicifolia belong to Meliaceae family. The chemical structures of 1 and 2 were identified based on spectroscopic evidence including UV, IR, 1D NMR, 2D NMR as well as mass spectra and by comparison with those previously reported spectra data. Both compounds were evaluated for their cytotoxic effects against cervical cancer HeLa cells in vitro. Compounds 1 and 2 showed cytotoxicity activity against HeLa cervical cancer cells with IC50 values of 0.80 and 26.42 µM, respectively.

Eleutherine palmifolia (L.) Merr. is a typical plant found in Central Kalimantan that has been used empirically by the Dayak people as medicine for various diseases, including cancer. The plant contains flavonoid compounds that potentially used as an anticancer. The purpose of this study is to find the most active fraction, indicated by its cytotoxic potency on HeLa cervical cancer cell line, and to identify compounds in E. palmifolia bulbs fraction. E. palmifolia bulbs was extracted by maceration. The extraction with ultrasonic bath and partition fractionation was conducted by using n-hexane, chloroform, and ethyl acetate. Each fraction was tested for toxicity level on HeLa cells using MTT assay. The identification of active compounds was carried out by Ultra Performance Liquid Chromatography-Mass Spectrometry (UPLC-MS). The result showed that based on the IC50 value, the ethyl acetate fraction had the highest bioactivity. IC50 values of n-hexane, chloroform, and ethyl acetate fractions were 250.77±19.01; 720.46±42.38; and 44.34±9.45μg/mL, respectively. The identification of the active compound in ethyl acetate fraction resulted 28 chemical compounds. Compounds with the highest percentage area were isoliquiritigenin and oxyresveratrol. The ethyl acetate fraction of E. palmifolia bulbs is potential to be developed as an anticancer candidate (phytopharmaceutical).Keywords: Compound identification, Anticancer activity, Eleutherine palmifolia (L.) Cervical cancer ranks second of cancer that affects women in the world and the first order for women in developing countries. Plant Salung (Psychotria viridiflora Reinw. Ex. Blume) have been used for generations to treat various diseases. The purpose of the study to determine the effects of leaf extracts and fractions salung as the anti-proliferation and inducer of apoptosis in HeLa cervical cancer cells. Concentration of extracts and fractions of 640; 320; 160; 80; 40 µg/mL, while cisplatin using a concentration of 200; 100; 50; 25 µg/mL. The results showed IC50 extract value 380.7 µg/mL. n-hexane fraction amounted to 229.3 µg/mL. ethyl acetate fraction of 116.8 µg/mL, and amounted to 562.8 µg/mL fraksi methanol water, so that the ethyl acetate fraction had enough categories active cytotoxic activity seen from the IC50. To further test the researchers only tested the active fraction is the fraction of ethyl acetate. Results ethyl acetate fraction doubling time is 58, which means replicating or proliferating cells on hour to 58 while the cells replicate cisplatin on the hour to 64.5 and the control cell on the hour to 41. Results of flowcytometry showed ethyl acetate fraction induce apoptosis by 72.82% while cisplatin 87.96%. It can be concluded that the extracts and fractions of leaves salung have a cytotoxic effect, acts as an anti-proliferation and inducer of apoptosis in HeLa cervical cancer cell.

Modern pharmacology, however, relies on refined chemicals - either obtained from plants, or synthesized. This work investigated the anticancer, antioxidant and Cytotoxicity activities of N. lotus leaves citrate, N. lotus leaves not citrate, P. juliflora leaves commonly used as anti-inflammatory and Ant diabetic. All the plant parts were extracted using 80% methanol, the anticancer activity was examined by using MTT assay against Hela (Cervical Cancer) Cell Line. And determine their antioxidant activities by testing DPPH cytotoxicity using - (4, 5-Dimethyl thiazole-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT), filter and kept in dark, prepared freshly. The examined plants methanolic extracts of P. juliflora leaves is high activity against Hela (Cervical Cancer) Cell Line IC50 is 56.02 µg/ml. The extract C N. lotus leaves citrate, N. lotus leaves not citrate has shown none active anti- Hela (Cervical Cancer) IC50 values 100, and > 100 μg/ml respectively. All the extracts revealed cytotoxicity activity not toxic in N. lotus leaves citrate, N. lotus leaves not citrate, P. juliflora leaves the inhibition percentage with (90.81, 89.33074 , 86.47866 ) (73.17427 , 71.93975 , 60.17069 ) (74.93001, 73.78714 , 71.13981 ) respectively. The following plant parts showed highly potent scavenging activity against DPPH (above 80%); N. lotus leaves citrate, 88.78, While the following revealed a good activity against DPPH (between 60-79); N. lotus leaves not citrate, P. juliflora leaves with inhibition % are 77and 62.96 respectively.

BACKGROUND<br /><p>The leaves of Nerium indicum Mill. have been utilized traditionally to cure cancer. By using Bioassay guided extraction and isolation method, three compounds (NiO-1, NiO-2, NiO-3) were isolated from an active fraction of the CHCl3 extract of N. indicum leaves. NiO-2 identified as 5α- oleandrin is the best cytotoxic compound on HeLa cervical cancer cell in vitro among the other two. However mechanism of action of the compound hasn’t been evaluated yet. The aims of this study were to determine the mechanism of action at molecular level.</p><p>METHODS<br />The action mechanism of 5α-oleandrin on HeLa cervical cancer cells was analyzed by staining the cells with Hoechst 33342; the agarose gel electrophoresis was aimed to determine the DNA fragmentation and the western blotting was aimed to determine the Bcl-2 and Bax protein expression.</p><p>RESULTS<br />Incubation of HeLa cervical cancer cell with 5α-oleandrin at the concentration 3,47x10-4 mM (24 hours) followed by staining with Hoechst 33342, a broken up light blue color of nucleus was observed (compared with intensive color of untreated control). By gel electrophoresis (at the same concentration of the tested compound), a smear band at about 200 bp was observed. In addition, cells treated with 5α-oleandrin displayed a decreasing of the Bcl-2 protein expression and increasing of the Bax protein expression</p><p>CONCLUSION<br />5α-oleandrin induced HeLa cervical cancer cells DNA fragmentation observed by the presence of a smear band at about 200 bp indicative the apoptotic occurrence. 5α-oleandrin induces apoptosis by reducing the Bcl-2 protein expression but the Bax protein expression increases.</p>
