 A method for revealing genetic affinity between laboratory rats has been developed, which is based on the transplantation of ear skin-chondrocyte allografts onto the surface of a full-thickness skin defect. On the example of Wistar rats (laboratory rats were taken from three research institutes), it was shown that the rejection of allografts occurs in the interval between 8 and 21 days with the same genetic differences that existed between the animals. However, at the same time, only those structures of ear allografts such as the dermis, epidermis and its derivatives die and are rejected, but the layers of chondrocytes are submerged under the granulation tissue and remain in a viable position for at least 210 days. The latter phenomenon is probably due to the membranes that cover the chondrocytes and act as an immunological barrier.

The microbiological, physical, chemical, and immunological barriers of the gastrointestinal tract (GIT) begin developing in utero and finish maturing postnatally. Maturation of these barriers is essential for the proper functioning of the GIT. Maturation, particularly of the immunological barrier, involves stimulation by bacteria. Segmented filamentous bacteria (SFB) which are anaerobic, spore-forming commensals have been linked to immune activation. The presence and changes in SFB abundance have been positively correlated to immune markers (cytokines and immunoglobulins) in the rat ileum and stool samples, pre- and post-weaning. The abundance of SFB in infant stool increases from 6 months, peaks around 12 months and plateaus 25 months post-weaning. Changes in SFB abundance at these times correlate positively and negatively with the production of interleukin 17 (IL 17) and immunoglobulin A (IgA), respectively, indicating involvement in immune function and maturation. Additionally, the peak in SFB abundance when a human milk diet was complemented by solid foods hints at a diet effect. SFB genome analysis revealed enzymes involved in metabolic pathways for survival, growth and development, host mucosal attachment and substrate acquisition. This narrative review discusses the current knowledge of SFB and their suggested effects on the small intestine immune system. Referencing the published genomes of rat and mouse SFB, the use of food substrates to modulate SFB abundance is proposed while considering their effects on other microbes. Changes in the immune response caused by the interaction of food substrate with SFB may provide insight into their role in infant immunological barrier maturation.

AbstractWhole organ perfusion decellularization has been proposed as a promising method to generate non-immunogenic organs from allogeneic and xenogeneic donors. However, the ability to recellularize organ scaffolds with multiple patient-specific cells in a spatially controlled manner remains challenging. Here, we propose that replacing donor endothelial cells alone, while keeping the rest of the organ viable and functional, is more technically feasible, and may offer a significant shortcut in the efforts to engineer transplantable organs. Vascular decellularization was achieved ex vivo, under controlled machine perfusion conditions, in various rat and porcine organs, including the kidneys, liver, lungs, heart, aorta, hind limbs, and pancreas. In addition, vascular decellularization of selected organs was performed in situ, within the donor body, achieving better control over the perfusion process. Human placenta-derived endothelial progenitor cells (EPCs) were used as immunologically-acceptable human cells to repopulate the luminal surface of de-endothelialized aorta (in vitro), kidneys, lungs and hind limbs (ex vivo). This study provides evidence that artificially generating vascular chimerism is feasible and could potentially pave the way for crossing the immunological barrier to xenotransplantation, as well as reducing the immunological burden of allogeneic grafts.

The skin is the biggest organ and provides a physical and immunological barrier against pathogen infection. The distribution of primary cilia in the skin of mice has been reported, but which cells in human skin have them has not, and we still know very little about how they change in response to immune reactions or disease. This review introduces several studies that describe mechanisms of cilia regulation by immune reaction and the physiological relevance of cilia regulating proliferation and differentiation of stroma cells, including skin-resident Langerhans cells. We discuss the possibility of primary cilia pathology in allergic atopic dermatitis and the potential for therapies targeting primary cilia signaling.

Eyelids represent a complex structure; its principal function is to provide physical and immunological barrier. The complexity of this structure lies in the blinking and in the range of movement they carry out respecting the integrity of the cornea. The objective of this study is to provide a surgical alternative with minimal morbidity for the reconstruction of the upper eyelid, as well as to present the available tools for the treatment of this complex area of the facial anatomy. We present the case of a female patient of 62 years, with 50% of necrosis on the surface of the upper eyelid and infection data, surgical management was initiated with debridement, systemic antibiotic therapy and negative pressure system device, once the infectious process was resolved and according to the principles of reconstruction by aesthetic subunits, it was decided to reconstruct 100% of the subunit affected by a skin graft of total thickness of the contralateral eyelid through a blepharoplasty with good aesthetic-functional results. The reconstruction of the eyelids should be managed by a plastic surgeon, taking into account the principles of the aesthetic subunits, optimally the donor tissue should have similarity with the tissue to be reconstructed, with respect to the size, color and flexibility, to obtain an adequate aesthetic and functional result.

The studies were carried out with the aim of revealing histological reactions in the shoots of remontant raspberries as an aspect of the growth-stimulating effect of pre-planting treatment of the root system of seedlings with strains of saprotrophic bacteria of the genus Bacillus. The observations in the model experiment were carried out in 2017-2018 in the production plantings of remontant raspberries of the agricultural artel “Sady Sibiri” in the Novosibirsk Region. Plants were treated before planting by soaking the root system in a working fluid containing a bioagent at a concentration of 1×105 CFU / ml. In terms of the totality of growth-stimulating effects, the most effective complex effect on remontant raspberry plants was provided by pre-planting treatment with B subtilis strain VKPM B-10641 and a mixture preparation Fitop 8.67. The number of replacement shoots increased by 45–69%, the length of replacement shoots - by 16–20%, the amount internodes - 1.2 times relative to control. Under the influence of the same bacterial bioagents in the shoots of remontant raspberries, an increase in the thickness of the peridermis by 20–25% relative to the control, an increase in the number of peridermal layers by 12–35%, suberinized (matured) layers of the peridermis by 30–48%, and the thickness of xylem by 12 –22% was observed. B subtilis VKPM B-10641 strain and Fitop 8.67 influence raspberry plants, the regularity of simultaneous stimulation of growth processes in the replacement shoots and the formation of mature peridermis. So it acts as an immunological barrier against phytopathogens, pests and abiotic stress factors. So the research proved it.

A study of solid-phase structures of blood serum using wedge-shaped and marginal dehydration methods (Litos system technology) was conducted in order to find out the causes of an inflammatory reaction followed by fibrosis in the second operated joint in a patient with bilateral knee arthritis. The study was aimed at identifying specific morphological markers that characterize the bodys response to the endoprosthesis material. Its solid-phase structures indicated the activation of a hyperergic reaction with daily incubation of blood serum with an alloy of titanium, aluminum, and vanadium. On the contrary, the immunological activity of blood serum can be suppressed and the structures present in it can be transformed into amorphous detritus with the incubation of an alloy of cobalt, chromium, and molybdenum. It was observed from the study that the nature of the immunological reaction of a sensitized organism depends on the type of metals that are part of the endoprosthesis. The immune response causes inflammation of the periarticular tissue, followed by its fibrosation and the formation of a scar demarcation shell that separates the periarticular tissue from the endoprosthesis and performs the function of an immunological barrier on the alloy of titanium, aluminum, and vanadium. On the other hand, an immunological reaction causes the destruction of inflamed periarticular tissue, followed by gradual destruction of the articular bag on the alloy of cobalt, chromium, and molybdenum.
