Effect of avitaminosis C on the state of the anticlotting system of the blood in guinea pigs

Considering the particular danger of remote skin reactions to ultraviolet irradiation (UVI), it is advisable to use ointments with antioxidant activity to reduce its negative effect on the skin. The rationale for the choice of ointments with antioxidant activity was the fact that they reduce the damaging effect of ultraviolet radiation in the erythemal and early post-erythemal period. The presence of a regular connection between the development of the early and late periods has given reason to assume the protective effect of ointments on the remote skin reactions. Objective: to study the effect of thiotriazoline ointment and thiotriazoline ointment with silver nanoparticles on the state of the morphological structures of the skin of guinea pigs after local UVI. Material and methods of research. The study involved 132 albino guinea pigs weighing 400-500 g, divided into 4 groups: 1 - intact, 2 - control (guinea pigs subjected to local UVI), 3 and 4 main ones. The third main group included guinea pigs that after UVI were administered thiotriazoline ointment in the treatment and prophylactic regime, the fourth main group included guinea pigs that after UVI were administered thiotriazoline ointment with silver nanoparticles in the same mode as Group 3. Ointments were applied 1 hour before irradiation and daily until erythema disappeared. Ultraviolet erythema was caused by irradiation in 1 minimum erythemal dose. After 2, 4 hours, on the 3rd, 8th, 15th, 21st, 28th day, the fragments of irradiated skin were investigated using histochemical and morphometric methods (fibroblast density and epidermis thickness). Results. Morphological changes in the skin after applying ointments with antioxidant activity were unidirectional. It was revealed that in the early periods after irradiation, thiotrazoline ointment and thiotrazoline ointment with silver nanoparticles do not affect changes in the thickness of the epidermis, but statistically significantly reduce the density of fibroblasts in the dermis on the 3rd day of the experiment compared to the control group. In the later periods, under the influence of thiotriazoline ointment, a gradual decrease in the thickness of the epidermis, which reached the norm by the end of the experiment, was observed. On the 8th day, the maximum density of fibroblasts was recorded, in the subsequent periods of the experiment, the index gradually decreased, which was accompanied by collagenization of the papillary layer in the loci of damage to collagen and elastic fibers detected in 50% of cases. In later times, under the influence of thiotriazoline ointment with silver nanoparticles, the processes of restoring the morphological structures of the skin occurred faster. In parallel with the decrease in the density of fibroblasts in the loci of the previous damage to the collagen and elastic fibers of the papillary layer, thickening of collagen fibers was observed, replacing them with segments of destruction of elastic fibers. In this group, at the end of the experiment, the collagenization locus was small, single, occurring in 16.7% of cases. Conclusions Ointments with antioxidant activity exert a positive effect on the state of morphological structures of the skin, damaged as a result of local UVI, in erythemal and post-erythemic periods. In the early periods after the local UVI, there was a general tendency for the effect of both ointments, as they reduced the density of fibroblasts on the 3rd day, but did not result in complete normalization. In the late period after local UVI , under the influence of thiotriazoline ointment and thiotriazoline ointment with silver nanoparticles, thickness of the epidermis (by 21st and 15th day, respectively) and density of fibroblasts (by the 28th day) decreased to normal while without treatment both indicators exceeded the norm by several times for 28 days of the experiment.

Carried out was morphometric analysis of cells' reaction of APUD system (neuroendocrinal cells) of guinea pigs immunized with live plague vaccine by aerogenic method. The results of investigations were compared with characteristics of the state of biomodel's internal organs. Elucidated was that the changes of morphofunctional state of neuroendocrinal cells in lungs and organs of immunity were of phase character suggesting the involvement of APUD system elements in the inhalant method of antigenic material intake. Assessment of APUD-system cell's reaction enables to estimate indirectly the nature of adaptive and compensatory processes in biomodel in the course of anti-plague vaccination.

1. Tubercle bacilli were isolated by the Petroff method from 64.8 per cent of the sputa which were found to be positive by the direct microscopic method. 2. The organism was recovered from 69.2 per cent of the samples of milk that were artificially infected. 3. Of the milk obtained through the New Haven Health Department Laboratory 8.4 per cent of the samples were found to contain tubercle bacilli, while none of the twenty-nine samples from the State Laboratory gave positive cultures. 4. All the guinea pigs (thirteen), except one, which were used as control animals died without any visible signs of tuberculosis. 5. Petroff's method for the isolation of tubercle bacilli from sputum may be applied successfully to the examination of milk. With slight modifications, the method should prove constant and reliable. The need of such a method for the examination of milk and milk products is apparent.

Background: The capybara (Hydrochoerus hydrochaeris) is the largest South American rodent species alive, and it preferentially inhabits floodable environments. Currently, capybara populations have been increasing owing to lack of predators and increased availability of food. This favors reemergence of Rocky Mountain spotted fever in the state of São Paulo, as these animals can play host to the Cayenne tick (Amblyomma cajennense), a vector of the bacteria that transmit this disease. The objective of this work was to perform anatomical and radiographic analyses on the teeth of capybaras to broaden knowledge on the morphology of this species, as scientific interest on capybaras has been growing owing to reemergence of Rocky Mountain spotted fever.Materials, Methods & Results: Six capybara carcasses from the state of São Paulo, Brazil, were taken to the Laboratory for Animal Anatomy of the School of Agricultural Sciences and Veterinary Medicine of UNESP - Jaboticabal for obtainment of heads by disarticulation of the cervical region. The heads were frozen at -18°C, and then sectioned medially with a band saw to avoid tissue overlap in radiographs. The head halves were radiographed in the Diagnostic Imaging Section at the Veterinary Hospital of UNESP-Jaboticabal in laterolateral projection. After taking the radiographs, the teeth were extracted from the dental arches using anatomical forceps and pliers, placed in 0.1% thymol, and kept at 4°C for up to 30 days. The teeth were dried using an air jet produced by a compressor, and placed in molds made of aluminum foil. The specimens were embedded in alternating layers of clear polymethylmethacrylate and liquid polymerization catalyst until each tooth was completely covered. After 24 h, the molds were removed, and the blocks were sectioned using a band saw. Transversal and longitudinal calcified tooth sections with a thickness of 1.5 mm were prepared. The sections were sanded using an automotive sander with 80-grit sandpaper, and then sanded by hand until a thickness of 0.12 mm was reached. The tooth fragments were removed from the resin, dehydrated in ethanol, clarified in xylene, and placed in a Petri dish containing glycerin for evaluation using a mesoscope and a negatoscope. Capybaras were found to have one incisor, one premolar, and three molar teeth in each dental hemiarch (superior and inferior). All teeth contained large, open roots.  The root of the superior incisor is rostral to the root of the premolar tooth, and the root of the inferior incisor lies between the roots of the premolar and the first molar teeth. The inferior incisors are much larger than their superior counterparts, and their ends are chisel-shaped. The pulp cavity extends from the apical region to up to two-thirds of the longitudinal length of the tooth, and the enamel is arranged in layers. The roots of the four molar teeth are large and resemble several aggregated laminae arranged longitudinally - five laminae in the inferior molars, four laminae in the premolar, first and second molars, and 13 or 14 laminae in the third superior molar.Discussion: The dental formula of capybaras is similar to that of guinea pigs, chinchillas, and pacas, but different from that of agoutis. All teeth are aradicular as in guinea pigs and chinchillas because of the great tooth wear owing to feeding. In chinchillas and guinea pigs, the apex of the mandibular incisor tooth is located between the second and the third molar; in capybaras, it is located between the premolar and the first molar. The premolar and molar teeth of capybaras are similar to their counterparts in the paca and to the molars of elephants because they exhibit parallel internal laminae. The teeth of capybaras are similar to those of guinea pigs in terms of number and type of teeth, and similar to those of pacas and elephants regarding external morphology.

A fire involving an electric service transformer and its cooling fluid, a mixture of PCBs and chlorinated benzenes, contaminated an office building with a fine soot. Chemical analysis showed PCDDs and PCDFs including the highly toxic tetra isomers. Guinea pigs were chosen as an experimental animal to test the soot's toxicity because of their sensitivity to these compounds, and the liver was examined because it is a target organ. The soot was suspended in 0.75% methyl cellulose and administered in a single dose by gavage at levels of 1,10,100, and 500mgm soot/kgm body weight. Each dose group was composed of 6 males and 6 females. Control groups included 12 (6 male, 6 female) animals fed activated carbon in methyl cellulose, 6 males fed methyl cellulose, and 16 males and 10 females untreated. The guinea pigs were sacrificed at 42 days by suffocation in CO2. Liver samples were immediately immersed and minced in 2% gluteraldehyde in cacadylate buffer at pH 7.4 and 4°C. After overnight fixation, samples were postfixed in 1% OsO4 in cacodylate for 1 hr at room temperature, embedded in epon, sectioned and stained with uranyl acetate and lead citrate.
