To study the composition of a biofilm a previous extraction method is required to separate cells from the matrix. There are several methods reported in the literature; however they are not efficient or promote leakage of intracellular material. In this work several extraction methods were assayed in mixed culture and pure culture biofilms and their efficiency was evaluated by the amount of organic carbon, proteins and intracellular material extracted. The results showed that the extraction with glutaraldehyde 3% (w/v) was the most suitable method, extracting great amounts of organic carbon without promoting cell lysis or permeabilization. Glutaraldehyde is a bifunctional reagent that binds to cell walls avoiding their permeabilization and the biofilm matrix is solubilized in the solution.

The aim of the work was a comparative study of the antiviral activity of the melanin isolated from submerged culture of basidiomycete birch fungus Inonotus obliquus F-1244 and melanin obtained from natural birch fungus. We showed that the melanin isolated from cultivated birch fungus is similar to the melanin from natural birch fungus in respect of its physicochemical and antiviral properties. Although, the sample`s activity depended not only on the source of melanin, but also on the method of isolating and purification. The toxic dose (CD50) varied from 300 to 2500 µg/ml. The effective antiviral dose in the case of A/H1N1pdm09 (ID50) was one from 10 to 47 µg/ml. The highest therapeutic index was for the melanin of the cultivated birch fungus, purified by dialysis. The index made 160; it is higher in 2, 5 times in comparison with natural birch fungus. The melanin from cultivated birch fungus is a promising substance for the developing of naturally occurring medication.

Abstract This study explores the potential for synchronous extraction of Cu, Cr, Ni and Zn during sewage sludge bioleaching processes, using three types of bacterial cultures: a pure culture of Acidithiobacillus ferrooxidans (A. ferrooxidans); a pure culture of Acidithiobacillus thiooxidans (A. thiooxidans); and a mixed culture of A. ferrooxidans and A. thiooxidans. Variable operating parameters included initial pH, solids concentration, sulfur concentration and ferrous iron concentration, with optimization via Box-Behnken design of response surface methodology. Results indicate that the mixed culture of A. ferrooxidans and A. thiooxidans, was the most effective at bioleaching heavy metals from sewage sludge. The optimal operating conditions were as follows: an initial pH of 2.0, with concentrations of 3% solids, 6.14 g L−1 sulfur and 4.55 g L−1 ferrous iron. Maximum extraction efficiencies obtained after 14 days of bioleaching under optimal conditions, were 98.54% Cu, 57.99% Cr, 60.06% Ni and 95.60% Zn. Bioleaching kinetics were effectively simulated using a shrinking core model to explain the leaching reaction, with modelling results suggesting that the rate was determined by the diffusion step.

ABSTRACT The food industry is in need of rapid, reliable methodologies for the detection of Listeria monocytogenes in ready-to-eat products, as an alternative to the International Organization of Standardization (ISO) 11290-1 reference method. The aim of this study was to evaluate impedanciometry combined with chromogenic agar culture for the detection of L. monocytogenes in dry-cured ham. The experimental setup consisted in assaying four strains of L. monocytogenes and two strains of Listeria innocua in pure culture. The method was evaluated according to the ISO 16140:2003 standard through a comparative study with the ISO reference method with 119 samples of dry-cured ham. Significant determination coefficients (R2 of up to 0.99) for all strains assayed in pure culture were obtained. The comparative study results had 100% accuracy, 100% specificity, and 100% sensitivity. Impedanciometry followed by chromogenic agar culture was capable of detecting 1 CFU/25 g of food. L. monocytogenes was not detected in the 65 commercial samples tested. The method evaluated herein represents a promising alternative for the food industry in its efforts to control L. monocytogenes. Overall analysis time is shorter and the method permits a straightforward analysis of a large number of samples with reliable results.

ABSTRACT Leptospirillum ferriphilum strain UBK03 isolated from a mine in Jiangxi, China, is resistant to Ni2+ (30 to 40 mM). A four-gene nickel resistance cluster was identified and, when transformed into Escherichia coli, enabled growth in 6 mM nickel. Mutation experiments revealed that the genes ncrA, ncrB, and ncrC could confer nickel resistance in Escherichia coli, whereas the gene ncrY could have a negative effect on nickel resistance.

The response of iron-oxidizingLeptospirillum ferriphilumYSK and sulfur-oxidizingAcidithiobacillus thiooxidansA01 to arsenite under pure culture and coculture was investigated based on biochemical characterization (concentration of iron ion and pH value) and related gene expression.L. ferriphilumYSK andAt. thiooxidansA01 in pure culture could adapt up to 400 mM and 800 mM As(III) after domestication, respectively, although arsenite showed a negative effect on both strains. The coculture showed a stronger sulfur and ferrous ion oxidation activity when exposed to arsenite. In coculture, the pH value showed no significant difference when under 500 mM arsenite stress, and the cell number ofAt. thiooxidanswas higher than that in pure culture benefiting from the interaction withL. ferriphilum. The expression profile showed that the arsenic efflux system in the coculture was more active than that in pure culture, indicating that there is a synergetic interaction betweenAt. thiooxidansA01 andL. ferriphilumYSK. In addition, a model was proposed to illustrate the interaction between arsenite and thearsoperon inL. ferriphilumYSK andAt. thiooxidansA01. This study will facilitate the effective application of coculture in the bioleaching process by taking advantage of strain-strain communication and coordination.
