Antifungal activity of ethanolic extracts of Ocimum gratissimum and Vernonia amygdalina leaves against dermatomycotic agents isolated from domestic animals in South Eastern Nigeria

The study investigated fungi associated with post harvest tomato fruits sold in the open market in Port Harcourt Metropolis. The antifungal activity (bitter leaf) extracts against spoilage was studied using well-in-agar diffusion method. Results showed that mean percentage incidence of fungi isolated from tomato fruits were Rhizopus stolonifer (56%), Aspergillus niger (62%) and Altermaria altermata (35%). Aqueous and ethanolic extracts of Vernonia amygdalina at different concentrations (20%, 40%, and 60%) (w/v) were used against fungi isolates, while water and ketoconzole (0.5 mg/mL) served as negative and positive control, respectively. Aqueous and ethanolic extracts of V. amygdalina inhibited the growth of all three test fungi. There were significant differences (p > 0.05) in the mean inhibitory effects of plant extracts and control. Antifungal activity measured as diameter of zone of inhibition revealed that V. amygdalina acqueous extract at 60% was very active against Aspergillus niger (16.50 mm) and Alternaria altermata (16.00 mm), while being moderately active against Rhizopus stolonifer (13.00 mm). However, 60% ethanolic extract of V. amygdalina was very active against A. niger (19.00 mm), A. altermata (17.00 mm) and Rhizopus stolonifer (15.80 mm). Phytochemical screening of V. amygdalina revealed the presence of tannins, oxalate, saponnins, flavanoid, cynogenic glucoside, phytate and Alkaloids V. amygdalina could serve as a potentially viable alternative to chemical fungicides in the preservation of post harvest tomato fruits (Lycopersicum esculentum).

Sporotrichosis is a serious public health problem in Brazil that affects human patients and domestic animals, mainly cats. Thus, the search for new antifungal agents is required also due to the emergence and to the lack of effective drugs available in the therapeutic arsenal. The aim of this study was to evaluate the in vitro antifungal profile of two synthetic series of coumarin derivatives against Sporothrix schenckii and Sporothrix brasiliensis. The three-components synthetic routes used for the preparation of coumarin derivatives have proved to be quite efficient and compounds 16 and 17 have been prepared in good yields. The inhibitory activity of nineteen synthetic coumarins derivatives 16a-i and 17a-j were evaluated against Sporothrix spp. yeasts and the most potent compounds were 16b and 17i. However, according to concentrations able to inhibit (minimum inhibitory concentrations) and kill (minimum fungicidal concentrations) the cells, 17i was more effective than 16b against Sporothrix spp. Thus, 17i exhibited good antifungal activity against S. brasiliensis and S. schenckii, suggesting that it is an important scaffold for the development of novel antifungal agents.

Postharvest fruits including tomatoes are commonly infected by gray mold disease resulting in significant economic losses in the fruit industry. Therefore, this study aimed to develop botanical fungicide derived from Vernonia amygdalina leaf extract to control gray mold on tomato. The emulsion formulation containing surfactant, oil carrier and water was optimized at different non-ionic alkyl polyglucoside surfactants through eleven combinations of oil to surfactant ratio (0:10, 1:9, 2:8, 3:7, 4:6, 5:5, 6:4, 7:3, 8:2, 9:1 and 10:0 w/w). From eight selected formulations, two formulations, F5 and F7 showed stable in storage, remarkable thermodynamic stability, smaller particle size (66.44 and 139.63 nm), highly stable in zeta potential (−32.70 and −31.70 mV), low in polydispersity index (0.41 and 0.40 PdI), low in viscosity (4.20 and 4.37 cP) and low in surface tension (27.62 and 26.41 mN/m) as compared to other formulations. In situ antifungal activity on tomato fruits showed F5 formulation had a fungicidal activity against B. cinerea with zero disease incidence and severity, whereas F7 formulation reduced 62.5% disease incidence compared to a positive control with scale 1. Based on these findings, F5 formulation exhibited pronounced antifungal activity and may contribute to the development of new and safe antifungal product against gray mold on tomato.

Background: The article is a review of three bitter vegetables consumed in South-eastern Nigeria (Gongronema latifolium, Vernonia amygdalina and Vitex doniana). These vegetables are not very palatable to taste but are highly famed because of the myriad of illnesses that they are reputed to cure and manage. The different aspects of these vegetables reviewed are their ethnobotany, zoo pharmacology, bioactivity, phytochemistry and toxicity. Materials and method: The literatures consulted were searched using electronic search engines (Google, Google scholar and Pubmed). Substantial amount of literature was consulted but only those directly related to the main review were selected. Result: The review revealed that the three bitter vegetables possessed phytochemicals like saponins, flavonoids, alkaloids among others that justify the claims of curing and managing many of the illnesses that traditional folks attribute to them. From literature, antimicrobial, antinflammatory, antidiabetic, antioxidant and anti cancer activities were attributed to these three bitter herbs. Antinutritional substances like phytate and oxalate reported in these herbs were not regarded as harmful for human consumption because of the traditional method of processing the vegetables by squeeze washing in several changes of water. Conclusion: The three bitter herbs are regarded as beneficial for human consumption and the information should be disseminated to a large audience. Key words: V. amygdalina, V. doniana, G.

The study evaluated nutritional and organoleptic properties of Clarias gariepinus pre-treated with aqueous leave extracts of Ocimum gratissimum and Vernonia amygdalina before processing. Thirty-six (36) C. gariepinus (live weight = 1.2± 0.2kg) samples were procured and processed for the study. Fish samples were divided into three parts of twelve fish each. Each part was grouped into three treatments having four fish in each group using a completely randomized design. Treatment one (T1) had fish immersed in 5% brine without herbal extract (control). In Treatment two (T2), fish were soaked in mixture of 5% brine and 5% O. gratissium (scent leaf) extracts while Treatment three (T3) had fish soaked in mixture of 5% brine and 5% V. amygdalina (bitter leaf) extracts all for 30 minutes. Treated fish samples were smoke-dried using charcoal kiln for 5 hours at temperature of 80 – 90° C. Samples were then cooled under ambient condition (30+3oC) for 4 hours. The smoke-dried fish samples were labeled according to treatments and stored for 7 days under aseptic condition. Organoleptic properties of the samples were conducted at every 24-hour of storage using a 9-point hedonic scale. Mean values of proximate composition of samples were significantly different (p<0.05). Protein content (70.01%) in T3 was higher than 68.31% and 66.62% in T2 and T1 respectively. Similarly, fat (5.12%) and ash (4.68%) levels for T3 were higher than 4.85% and 4.46% fat with 4.36% and 3.98% ash contents for T2 and T1 respectively. Results of mean scores for each organoleptic property decreased significantly (p<0.05) during storage period with no consistent trend in score pattern.
