Two new prenylated flavanones, isomaackiaflavanone A (1), isomaackiaflavanone B (2) and a new prenylated stilbene maackiastilbene (3), along with five known flavanones were isolated from the root bark of Maackia amurensis using repeated column chromatography on silica gel, Sephadex LH-20 and octadecyl silica (ODS-A) sorbents. The structures of the new compounds were elucidated by HPLC–PDA–MS, HR-ESI-MS, 1H, 13C, 1H–1H COSY, HSQC, ROESY and HMBC NMR analyses. The cytotoxicity of compounds 1–2, 4–8 against two human cancer cell lines HeLa and SK-MEL-5 was tested using the MTS method. Compounds 4, 6 and 8 showed the strongest cytotoxic activity among the compounds tested with IC50 values of 6.5, 8.8 and 7.7 μM, against SK-MEL-5 cells and 8.2, 18.8 and 12 μM against HeLa cells, respectively.

The suspensions of alcoholic extract of root bark of the plant Calotropis gigantea in 0.6% carboxy methyl cellulose (CMC) were evaluated for hepatoprotective activity in Wistar albino rats by inducing hepatic injury with D-galactosamine (400 mg/kg). Alcoholic extract of root bark of the plant Calotropis gigantea at an oral dose of 200 mg/kg and 400 mg/kg exhibited a significant (P<0.001, P<0.01 and P<0.05) protection effect by normalizing the levels of aspartate amino transferase (ASAT/ GOT), alanine amino transferase (ALAT/GPT), alkaline phosphatase (ALP), total bilirubin (TB), lactate dehydrogenase (LDH), which were significantly (P<0.001) increased in rats by treatment with 400 mg/kg i.p. of D-galactosamine. Silymarin (25 mg/kg), a known hepatoprotective drug used for comparison exhibited significant activity (P<0.001).

Seven isoflavonoids, including a new glycoside, (6a R,11a R)-medicarpin-3- O-gentiobioside (6), were isolated from the roots of Maackia amurensis using repeated column chromatography on a Toyopearl HW-50F sorbent and identified by HPLC–PDA–MS, 1H NMR, 13C, 1H–1H COSY, HSQC NMR and HMBC NMR analyses as daidzin (1), genistein-7- O-gentiobioside (2), pseudobaptigenin-7- O-gentiobioside (3), formononetin-7- O-gentiobioside (4), (6a R,11a R)-maackiain-3- O-gentiobioside (5), and 5- O-methylgenistein-7- O-gentiobioside (7). In the model of oxidative stress induced by formalin injection, the isolated isoflavone and pterocarpan glucosides 1-7 were shown to reduce the formation of malondialdehyde (MDA) and other thiobarbituric acid reactive substances (TBARS), as well as glutathione peroxidase (GPO) activity in rats.

The histochemical distribution of sialoglycoconjugates in the CA1 region in the hippocampus formation of 9-week-old rats and 30-month-old rats was examined using electron microscopy in combination with two lectins, Maackia amurensis lectin, specific for Siaα2–3Gal, and Sambucus sieboldiana agglutinin, specific for Siaα2–6Gal. Each lectin stained the plasma membranes of pyramidal cells, indicating that the Siaα2–3Gal and Siaα2–6Gal groups were expressed on their plasma membranes. These lectins also bound to synapses in the stratum lacunosum moleculare. The staining intensity of the lectins in the synapses in these layers was downregulated in the 30-month-old rats. These results indicated that both the Siaα2–3Gal and Siaα2–6Gal groups are expressed on these synapses and that the expression of these sialyl linkages decreases in the aged brain.
