SummaryAmong the numerous existing computer-based systems for processing pathological data, none contains sufficient space for encoding data on the basic cytological or histological changes of a certain organ or tissue, upon which the final diagnosis is based.An “analytical record” was constructed listing all the basic changes that can be encountered in the various pathological conditions of the vascular wall. The data collected on the “analytical record” were coded by means of an alphanumeric code and stored in an Apple II 48 K minicomputer.The advantages of this system include the computerization of the data by non-specialized personnel and the possibility to’ quantitatively analyze the histocytopathological parameters used for diagnosis in vascular pathology. This coding system may easily be adapted, with minor modifications, to the histopathological study of other organs and tissues.

AbstractThe Technical Committee on “Medical Informatics” of the European Committee for Standardization (CEN{TC251) is supporting developers ofterminological systems in healthcare by a series of standards. The dream of “universal” coding system was abandoned in favor of a coherent family of terminologies, diversified according to tasks; two ideas were introduced: (1) the “categorical structure”, i.e. a model of semantic categories and their relations within a subject field and (2) the “cross-thesaurus”, i.e. a system of descriptors to build a systematic representation (called here “dissection") for each terminological phrase, coherent across diverse terminologies on a given subject field.The goal is to assure coexistence and interoperability (and reciprocal support for development and maintenance) to three generations of systems: (1) traditional paper-based systems (first generation); (2) compositional systems built according to a categorical structure and a cross-thesaurus (second generation) and (3) formal models (third generation).Various scenarios are presented, on the exploitation of computer-based terminological systems. The idea of “operational meaning” of terminological phrases within administrative and organizational contexts and the idea of “task-oriented details” are also introduced, to justify and exploit design constraints on terminological systems.

Inspection of fabrics is a major consideration in fabric manufacture, as well as in manufacture of garments and other fabric-based goods. In this research, a computer-based system for objective assessment of fabric defects was designed with emphasis placed on fabric defects occurring in the Sri Lankan industry. Image processing techniques were used to analyse scanned images of the test fabric, compare it with an ideal sample, and identify defects according to pre-learnt rules. The information gathered was then used to grade the fabric, either by determining the frequency of defect occurrence or assigning points. A new classification method for common defects was designed, thereby facilitating grading according to commonly used grading systems. A coding system for defects was also designed to help report defects to the user. The fabric defects were classified and stored according to the developed classification method and coding system.

The morphology of the exine of many pollen grains, at the time of flowering, is such that one can suppose that transport of substances through the exine occurred during pollen development. Holes or channels, microscopic to submicroscopic, are described for a large number of grains. An inner part of the exine of Epilobium angustifolium L. and E. montanum L., which may be referred to as the endexine, has irregularly shaped channels early in pollen development although by microspore mitosis there is no indication of such channeling in chemically fixed material. The nucleus in microspores used in the experiment reported here was in prophase of microspore mitosis and the endexine, while lamellated in untreated grains, did not contain irregularly shaped channels. Untreated material from the same part of the inflorescence as iron treated stamens was examined following fixation with 0.1M glutaraldehyde in cacodylate-HCl buffer at pH 6.9 (315 milliosmoles) for 24 hrs, 4% formaldehyde in phosphate buffer at pH 7.2 (1,300 milliosmoles) for 12 hrs, 1% glutaraldehyde mixed with 0.1% osmium tetroxide for 20 min, osmium tetroxide in deionized water for 2 hrs and 1% glutaraldehyde mixed with 4% formaldehyde in 0.1M cacodylate-HCl buffer at pH 6.9 for two hrs.

The use of serial sections and computer-based 3-D reconstruction techniques affords an opportunity not only to visualize the shape and distribution of the structures being studied, but also to determine their volumes and surface areas. Up until now, this has been done using serial ultrathin sections.The serial-section approach differs from the stereo logical methods of Weibel in that it is based on the Information from a set of single, complete cells (or organelles) rather than on a random 2-dimensional sampling of a population of cells. Because of this, it can more easily provide absolute values of volume and surface area, especially for highly-complex structures. It also allows study of individual variation among the cells, and study of structures which occur only infrequently.We have developed a system for 3-D reconstruction of objects from stereo-pair electron micrographs of thick specimens.

Rapid freezing and freeze-substitution have been employed as alternatives to chemical fixation because of the improved structural preservation obtained in various cell types. This has been attributed to biomolecular immobilization derived from the extremely rapid arrest of cell function. These methods allow the elimination of conventionally used fixatives, which may have denaturing or “masking” effects on proteins. Thus, this makes them ideal techniques for immunocytochemistry, in which preservation of both ultrastructure and antigenicity are important. These procedures are also compatible with cold embedding acrylic resins which are known to increase sensitivity in immunolabelling.This study reveals how rapid freezing and freeze-substitution may prove to be useful in the study of the mobile allergenic proteins of rye grass and ragweed. Most studies have relied on the use of osmium tetroxide to achieve the necessary ultrastructural detail in pollen whereas those that omitted it have had to contend with poor overall preservation.

The Information SuperHighway, Email, The Internet, FTP, BBS, Modems, : all buzz words which are becoming more and more routine in our daily life. Confusing terminology? Hopefully it won't be in a few minutes, all you need is to have a handle on a few basic concepts and terms and you will be on-line with the rest of the "telecommunication experts". These terms all refer to some type or aspect of tools associated with a range of computer-based communication software and hardware. They are in fact far less complex than the instruments we use on a day to day basis as microscopist's and microanalyst's. The key is for each of us to know what each is and how to make use of the wealth of information which they can make available to us for the asking. Basically all of these items relate to mechanisms and protocols by which we as scientists can easily exchange information rapidly and efficiently to colleagues in the office down the hall, or half-way around the world using computers and various communications media. The purpose of this tutorial/paper is to outline and demonstrate the basic ideas of some of the major information systems available to all of us today. For the sake of simplicity we will break this presentation down into two distinct (but as we shall see later connected) areas: telecommunications over conventional phone lines, and telecommunications by computer networks. Live tutorial/demonstrations of both procedures will be presented in the Computer Workshop/Software Exchange during the course of the meeting.
