The inhibitory activity of Lactobacillus spp. isolated from breast milk on gastrointestinal pathogenic bacteria of nosocomial origin

Neonatal necrotizing enterocolitis (NEC), which is a disease with a poor prognosis, is considered to be caused by the coincidence of intestinal ischemia-reperfusion injury and systemic inflammation due to the colonization of pathogenic bacteria. Interleukin (IL)-8, a proinflammatory cytokine, plays an important role in the pathophysiology of NEC. It was recently reported that IL-1β activates the IL-8 gene by regulating the transcriptional nuclear factor κB (NF-κB) signaling pathways in intestinal cells. The protective role of maternal milk in NEC pathogenesis has been reported in both human and animal studies. In this study, we show that human breast milk dramatically suppressed the IL-1β-induced activation of the IL-8 gene promoter by inhibiting the activation pathway of NF-κB. Moreover, we also show that human breast milk induced the production of IκBα. These results suggest that human breast milk could be protective and therapeutic in neonates with NEC by inhibiting the activation pathway of NF-κB.

In this study 100 samples were collected from infected children with acute and chronic tonsillitis who attended to Al-Yarmook Teaching Hospital (ENT consultation clinic) from 5/12/2013 to 1/3/2014. The result of laboratory culture was positive in 67 samples. Depending on their cultural, morphological and biochemical characterization of bacterial isolate of them were identified as (37.31%) belonged to Streptococcus pyogenes and the diagnosis is confirmed by the use of Remel Rapid STR System, (34.32%) belonged to S.parasanguinis, (11.94%) S.mitis, (11.94%) S.oralis and (4.47%) S.thoraltensis . Results confirmed that cup assay gave highest inhibition zone after 24 hrs compare with well diffusion methods for suspension of L.acidophilus gave highest inhibition zone after 48 hrs for incubation, while ahigh inhibition zone revealed for suspension of L.fermentum after 24 hrs incubation. the study included also the measurement of the inhibition activity for bacteriocins produced by L.acidophilus bacteria against pathogenic bacteria on nutrient agar by well diffusion method in which results revealed stability of the bacteriocins produced towards PH which kept its activity with PH 4-6 for 24 hrs, and the highest stability was with PH 4, however it lost a lot of its activity with acidic PH less than 2 and alkaline PH as 8. The treatment of bacteriocins with salts such as Nacl it revealed little effect in inhibition zone within 1 & 2% concetrations. The salt MgSo4 & Kcl showed reduction in the inhibitory activity in the low concentration, however the higher concentration of salt caused great reduction and 5% concentration led to loss of inhibitory activity for bacteriocins completely.

Two strains of Penicillium nalgiovense, which carried the god gene of Aspergillus niger and had increased glucose oxidase (GOD) activity compared with the wild-type strain, were tested for their ability to suppress the growth of certain food-related pathogenic bacteria. In contrast to the wild type, which showed no antibacterial effect when grown in mixed culture with different bacteria, the two tranformed strains were highly antagonistic. The strain that expressed higher amounts of GOD in general had higher inhibitory activity. Both strains showed antibacterial activity against Listeria monocytogenes, Salmonella Enteritidis, and Staphylococcus aureus. The inhibitory activity was dependent on the glucose concentration in the medium. S. aureus was completely inhibited at 1% glucose in the presence of the higher GOD-producing transformant. In contrast, if arabinose was used as a carbon source, no inhibition occurred. If catalase was added to the medium, the inhibitory activity of the transformants was completely inactivated, indicating that the hydrogen peroxide produced was responsible for the antibacterial activity of the transformants.

ABSTRACT Several human mucosal fluids are known to possess an innate ability to inhibit human immunodeficiency virus type 1 (HIV-1) infection and replication in vitro. This study compared the HIV-1 inhibitory activities of several mucosal fluids, whole, submandibular/sublingual (sm/sl), and parotid saliva, breast milk, colostrum, seminal plasma, and cervicovaginal secretions, from HIV-1-seronegative donors by using a 3-day microtiter infection assay. A wide range of HIV-1 inhibitory activity was exhibited in all mucosal fluids tested, with some donors exhibiting high levels of activity while others showed significantly lower levels. Colostrum, whole milk, and whole saliva possessed the highest levels of anti-HIV-1 activity, seminal fluid, cervicovaginal secretions, and sm/sl exhibited moderate levels, and parotid saliva consistently demonstrated the lowest levels of HIV-1 inhibition. Fast protein liquid chromatography gel filtration studies revealed the presence of at least three distinct peaks of inhibitory activity against HIV-1 in saliva and breast milk. Incubation of unfractionated and fractionated whole saliva with antibodies raised against human lactoferrin (hLf), secretory leukocyte protease inhibitor (SLPI), and, to a lesser extent, MG2 (high-molecular-weight mucinous glycoprotein) reduced the HIV-1 inhibitory activity significantly. The results suggest that hLf and SLPI are two key components responsible for HIV-1 inhibitory activity in different mucosal secretions. The variation in HIV inhibitory activity between the fluids and between individuals suggests that there may be major differences in susceptibility to HIV infection depending both on the individual and on the mucosal fluid involved.

Aim of the study: To present an analysis of the literature data on the role of Lactobacillus fermentum CECT5716 in the prevention and treatment of postpartum lactation complications. Materials and methods: literature sources published in the Pubmed, Scopus database were searched. Results: according to randomized clinical studies, the use of Lactobacillus fermentum CECT5716 in the postnatal period by mothers leads to the correction of breast microbiota by reducing the number of pathogenic bacteria and increasing lactic-acid bacteria, reducing the content of inflammatory factors in breast milk. Conclusion: Lactobacillus fermentum CECT5716 is an effective and safe way to prevent and treat postpartum lactation complications.

This study aimed to study the inhibition activity of purified bacteriocin produced from the local isolation Lactococcuslactis ssp. lactis against pathogenic bacteria species isolated from clinical samples in some hospitals Baghdad city. Screening of L. lactis ssp. Lactis and isolated from the intestines fish and raw milk was performed in well diffusion method. The results showed that L. lactis ssp. lactis (Lc4) was the most efficient isolate in producing the bacteriocin as well observed inhibitory activity the increased that companied with the concentration, the concentration of the twice filtrate was better in obtaining higher inhibition diameters compared to the one-fold concentration. The concentrated bacteriocin was purified using the gel filtration column and Sephacryl S-200. The results showed the high inhibitory activity of the purified bacteriocin after the purification against the positive and negative bacteria of the Gram stain under study compared to the one-fold concentration and two-fold before purification , The diameters of the inhibition zones after gel-filtering of the purified bacteriocin reached S. aureus, S. epidermidis, P. aeruginosa, E. coli, E. clocae and S. marcescens (23, 25, 26, 20, 22 and 28) Mm respectively. The carbohydrate content of purified bacteriocin from L. lactis ssp. lactis (Lc4) isolate was 6.02% with a molecular weight of 6310 Dalton. The results showed that purified bacteriocin retained its inhibitory activity at pH 2-10 and showed the highest inhibition at pH 4-6 and lost at pH 12. The purified bacteriocin was characterized by thermal stability. It retained its effectiveness when exposed to 40, 60, 80, 100°C for 30, 15, 5 minutes and 120°C for 15.5 minutes and lost 50% of its effectiveness when exposed to 120°C for 30 minutes. Results The purified bacteriocin was effectively retained when treated with enzyme pepsin and trypsin of 37°C for one hour and at pH 7.
