This study evaluated the effects of the feeding of spent mushroom substrate from Agaricus blazei on Nile tilapia (Oreochromis niloticus). The safety of 0–1000 μg/mL A. blazei spent substrate water extract (ABSSE) was demonstrated in the primary hepatic and splenic macrophages and the THK cell line (a cell line with characteristics of melanomacrophages) using a cytotoxicity assay. Here, 10 μg/mL of crude ABSSE promoted the phagocytic activity of macrophages and THK cells. Stimulating ABSSE-primed THK cells with lipopolysaccharides or peptidoglycan resulted in higher expression levels of four cytokine genes (e.g., interleukinz (IL)-1β, IL-12b, IL-8 and tumor necrosis factor α (TNFα)) and one cytokine gene (TNFα), respectively. An in vitro bacterial growth inhibition assay demonstrated that ABSSE could inhibit the growth of Streptococcus agalactiae. In the first feeding trial, Nile tilapia were fed with experimental feed containing 0, 1, or 5% of A. blazei spent substrate (ABSS) for seven and fourteen days followed by bacterial challenge assay. The best result was obtained when Nile tilapia were continuously fed for seven days on a diet containing 1% ABSS, with the survival rate being higher than in groups with 0% and 5% ABSS after challenge with S. agalactiae. In the second trial, fish were fed diets supplemented with 0% or 1% ABSS for seven days, and then all the groups were given the control feed for several days prior to bacterial challenge in order to investigate the duration of the protective effect provided by ABSS. The results showed that the protective effects were sustained at day 7 after the feed was switched. Overall, spent mushroom substrate from A. blazei is a cost-effective feed additive for Nile tilapia that protects fish from S. agalactiae infection.

The synthetic steroid 17α-Methyltestosteron (MT) is commonly used as a feed additive to produce male population of Nile tilapia (Oreochromis niloticus). The use of synthetic testosterone hormone is not recommended in Indonesia. This study was conducted to evaluate the effect of natural testosterone hormone in bull testes meal (BTM) on the masculinization of Nile tilapia using validated aceto carmine squash method of gonads of the fish. Experimental design was utilized two factors experiments in completely randomized design. Fry kept in 40-L glass aquaria at a density of 40 fry/aquarium. Fry (7 dph) received the BTM  for 7 days (T1), 14 days (T2) and 21 days (T3) and doses 0% (D1), 3% (D2), 6% (D3), and 9% (D4).  When treatment was these results, indicated that significant (P≥0.05) masculinization occurred only in the group treated of BTM and no treated of BTM. In the group treated of BTM, doses and duration treatment is not significant. The percentage of male fish 83.3% (9%-7d, 9%-21d, 6%-21d: doses and duration, respectively), higher than all group. Survival rate of fry (95-99.5%) is not affected by treatment BTM (no significant P≥0.05). Fish growth was significantly affected by treatment BTM compare with no treated of BTM. The highest growth performance of fry were obtained with the 9% BTM.Key words: masculinization, nile tilapia, bull testes meal ABSTRAKSteroid sintetik 17α-Methyltestosteron (MT) umumnya digunakan sebagai aditif pakan untuk menghasilkan populasi ikan nila jantan (Oreochromis niloticus). Penggunaan hormon testosteron sintetis tidak dianjurkan di Indonesia. Penelitian ini dilakukan untuk mengevaluasi pengaruh hormon testosteron alami dalam makanan testis banteng (BTM) pada maskulinisasi ikan nila menggunakan metode divalidasi aceto carmine squash, dari gonad ikan. Desain eksperimental dimanfaatkan dua eksperimen faktor dalam desain benar-benar acak. Fry disimpan dalam 40-L akuarium kaca pada kepadatan 40 fry/akuarium. Fry (7 DPH) menerima BTM selama 7 hari (T1), 14 hari (T2) dan 21 hari (T3) dan dosis 0% (D1), 3% (D2), 6% (D3), dan 9% (D4). Ketika pengobatan hasil ini, menunjukkan signifikan (P ≥ 0,05) hanya terjadi maskulinisasi pada kelompok perlakuan dari BTM dan tidak diperlakukan BTM. Pada kelompok diobati BTM, dosis dan durasi pengobatan tidak signifikan. Persentase ikan jantan 83,3% (9%-7d, 9%-21d, 6%-21d: dosis dan durasi, masing-masing), lebih tinggi dari kelompok semua. Tingkat kelangsungan hidup benih (95-99,5%) tidak dipengaruhi oleh pengobatan BTM (tidak ada P yang signifikan ≥ 0,05). Pertumbuhan ikan secara signifikan dipengaruhi oleh BTM pengobatan dibandingkan dengan tidak diobati BTM. Kinerja pertumbuhan tertinggi fry diperoleh dengan BTM 9%.Kata kunci: maskulinisasi, ikan nila, tepung testis sapi

This work examines the economic advantages of probiotic use in the diet of Nile tilapia broodstock during the reproductive period. For this purpose, Bacillus subtilis was applied as a feed additive. The experimental design was completely randomized with three treatment groups: the T0 control (without probiotic), the T1 continuous probiotic intake, and the T2 alternate probiotic intake at a dose of 0.50 g kg-1 of feed (1010 CFU g-1) with four replicates. For the reproduction assay, 118 females and 48 males of Nile tilapia (proportion 4 males:9 females. hapa-1) (weight 527.65 g ± 185.98 g and length 30.16 cm ± 3.57 cm) were distributed into 12 hapas (3.5 × 2.0 × 1.5 m). Reproductive variables (spawning female percentage, egg production, and fry production) were used to calculate the economic feasibility indexes (total cost of nutrition [TcN], gross revenue [GR], and total operational profit [ToP]). The results show increasing values for spawning female number, collected eggs, and surviving fry in the probiotic groups. We recommend continuous intake of probiotic (feed with addition of probiotic) at a dose of 0.5 g kg-1 of feed (1010 CFU g-1) during the breeding season of Nile tilapia, due to the suitable reproductive indexes and profitability.
