Epigenetic changes influence various physiological and pathological conditions in the human body. Recent advances in epigenetic studies of the skin have led to an appreciation of the importance of epigenetic modifications in skin diseases. Cutaneous sarcomas are intractable skin cancers, and there are no curative therapeutic options for the advanced forms of cutaneous sarcomas. In this review, we discuss the detailed molecular effects of epigenetic modifications on skin sarcomas, such as dermatofibrosarcoma protuberans, angiosarcoma, Kaposi’s sarcoma, leiomyosarcoma, and liposarcoma. We also discuss the application of epigenetic-targeted therapy for skin sarcomas.

Background: The clinical application of glabridin in treating skin diseases has been constrained by the limitations of its poor chemical stability and low skin permeability. Objective: Here, we describe tip-loading dissolving microneedles (TDMNs) encapsulating drugs only in the tips of needles for glabridin delivery with improved stability and skin permeability. Methods and Results: The TDMNs fabricated by solvent casting technique had sufficient mechanical strength to penetrate through the excised rat's skin without fracture. Drug delivery efficiency and drug residual in the skin of TDMNs were 63.16% and 49.28%, respectively. Glabridin encapsulated in the tips of TDMNs was effectively delivered into the abdominal skin of rats, and the in vivo delivery efficiency was inversely proportional to the drug doses. Transepidermal water loss (TEWL) significantly increased to 34.80 g/m2•h after the application of TDMNs and returned to normal levels (11.31 g/m2•h) after 8 h, indicating that the TDMNs were well tolerated. The stability of glabridin at room temperature was appreciably improved when loaded in TDMNs. Conclusion: These results suggest that intradermal delivery of glabridin by TDMNs is a safe and efficient alternative to currently available routes of administration.

Epstein's “Constructive Thinking Inventory” (CTI) was developed to measure the construct of experiential intelligence, which is based on his cognitive-experiential self-theory. Inventory items were generated by sampling naturally occurring automatic cognitions. Using principal component analysis, the findings showed a global factor of coping ability as well as six main factors: Emotional Coping, Behavioral Coping, Categorical Thinking, Personal Superstitious Thinking, Esoteric Thinking, and Naive Optimism. We tested the replicability of this factor structure and the amount of statistical independence (nonredundancy) between these factors in an initial study of German students (Study 1, N = 439) and in a second study of patients with chronic skin disorders (Study 2, N = 187). Factor congruence with the original (American) data was determined using a formula proposed by Schneewind and Cattell (1970) . Our findings show satisfactory factor congruence and statistical independence for Emotional Coping and Esoteric Thinking in both studies, while full replicability or independence could not be found in both for the other factors. Implications for the use and further development of the CTI are discussed.
