In humans, glutathione-dependent conjugation of halomethanes is polymorphic, with 60% of the population classed as conjugators and 40% as non-conjugators. We report the characterization of the genetic polymorphism causing the phenotypic difference. We have isolated a cDNA that encodes a human class Theta GST (GSTT1) and which shares 82% sequence identity with rat class Theta GST5-5. From PCR and Southern blot analyses, it is shown that the GSTT1 gene is absent from 38% of the population. The presence or absence of the GSTT1 gene is coincident with the conjugator (GSST1+) and non-conjugator (GSTT1-) phenotypes respectively. The GSTT1+ phenotype can catalyse the glutathione conjugation of dichloromethane, a metabolic pathway which has been shown to be mutagenic in Salmonella typhimurium mutagenicity tester strains and is believed to be responsible for carcinogenicity of dichloromethane in the mouse. In humans, the enzyme is found in the erythrocyte and this may act as a detoxification sink. Characterization of the GSTT1 polymorphism will thus enable a more accurate assessment of human health risk from synthetic halomethanes and other industrial chemicals.

The Role of GSTM1 and GSTT1 Polymorphism in Patients with Renal Cell CarcinomaMembers of the glutathione S-transferase (GST) superfamily exhibit polymorphic expression. GSTs are investigated as biomarkers of risk for various cancers, including renal cell carcinoma (RCC). The aim of this study was to test the association betweenGSTM1andGSTT1polymorphism and susceptibility to RCC, independently or in conjunction with known risk factors. Genomic DNA was isolated from 182 controls and 76 patients with RCC.GSTM1andGSTT1genotypes were determined by multiplex PCR. Data obtained were analyzed with respect to RCC risk factors including smoking and occupational exposure. The frequency ofGSTM1-null genotype was higher in patients with RCC (60.5%) compared to controls (47.2%).GSTT1-null genotype was found in 28.6% controls and 27.6% of cases.GSTM1-null individuals exhibit 1.9-fold increased risk of RCC (95% CI: 1.06-3.33). The presence ofGSTT1active genotype was associated with increased risk of RCC in occupationally exposed subjects when unexposedGSTT1-null subjects were used as a comparison group (OR: 2.48; 95% CI: 1.05-5.86). No association was found between the inactive form ofGSTM1andGSTT1and smoking in RCC patients. In a Serbian cohort of patients, the presence of aGSTM1active genotype is protective against RCC, whereas aGSTT1active genotype increases RCC risk in occupationally exposed subjects.

Abstract. Background: Aortic dilatation in Marfan syndrome (MFS) is progressive. It is associated with oxidative stress and endothelial dysfunction that contribute to the early acute dissection of the vessel and can result in rupture of the aorta and sudden death. We evaluated the participation of the glutathione (GSH) system, which could be involved in the mechanisms that promote the formation and progression of the aortic aneurysms in MFS patients. Patients and methods: Aortic aneurysm tissue was obtained during chest surgery from eight control subjects and 14 MFS patients. Spectrophotometrical determination of activity of glutathione peroxidase (GPx), glutathione-S-transferase (GST), glutathione reductase (GR), lipid peroxidation (LPO) index, carbonylation, total antioxidant capacity (TAC), and concentration of reduced and oxidized glutathione (GSH and GSSG respectively), was performed in the homogenate from aortic aneurysm tissue. Results: LPO index, carbonylation, TGF-β1, and GR activity were increased in MFS patients (p < 0.04), while TAC, GSH/GSSG ratio, GPx, and GST activity were significantly decreased (p < 0.04). Conclusions: The depletion of GSH, in spite of the elevated activity of GR, not only diminished the activity of GSH-depend GST and GPx, but increased LPO, carbonylation and decreased TAC. These changes could promote the structural and functional alterations in the thoracic aorta of MFS patients.

Cadmium (Cd) is a notorious environmental pollutant known for its wide range of toxicities to organisms. Thus, the present study is designed to examine whether melatonin, a potent antioxidant, protects against Cd-induced oxidative damage in the heart, liver and kidney of rats. Cd treatment at a dose of 0.44 mg/kg for 15 days caused severe damage in all these organs. These included significantly increased activities of SGPT, SGOT, lactate dehydrogenase- 1 and 5 and ALP and levels of total lactate, creatinine, lipid peroxidation, protein carbonyl content and reduced glutathione while the activities of superoxide dismutases, catalase, glutathione peroxidase, glutathione reductase and glutathione-S-transferase along with mitochondrial pyruvate dehydrogenase, isocitrate dehydrogenase, α-keto glutarate dehydrogenase, succinate dehydrogenase, NADH-cytochrome-c-oxidoreductase and cytochrome-c-oxidase were significantly reduced by Cd. However, if melatonin was given orally 30 min before Cd injection, all these alterations induced by Cd were significantly preserved by melatonin. Histological observations also demonstrated that Cd exposure caused cellular lesions, promoting necrotic or apoptotic changes. Notably, all these changes were significantly protected by melatonin. The results suggest that melatonin is a beneficial molecule to ameliorate Cd-induced oxidative damage in the heart, liver and kidney tissues of rats with its powerful antioxidant capacity, heavy metal chelating activity and competition of binding sites with Cd to the GSH and catalase.

he present work was designed to investigate the reproductive toxicity induced by oral administration of chlorpyrifos (CPF), cypermethrin (CYP) and their combination in adult male albino rats. Forty mature male albino rats were separated into four groups (10 each), the first group was used as control, while second, third and fourth groups received orally 1/20 LD50 of CPF (10 mg/kg b.wt), 1/20 LD50 of CYP (17.22 mg/kg b.wt) and 1/40 LD50 of CPF plus 1/40 LD50 of CYP (5 mg/kg b.wt CPF plus 8.61 mg/kg b.wt CYP) respectively for 26 days. The results revealed that exposure to CPF and/or CYP induced a significant decrease in the reproductive organs weight. Moreover, a significant decrease in spermatic picture (sperm cell concentration and viability) was observed with high percent of sperm abnormalities. Serum levels of testosterone and pituitary gonadotropins (FSH and LH) have been declined significantly in all treated groups. Significant elevations were observed in malondialdehyde and nitric oxide concentrations, while antioxidant enzymes superoxide dismutase and glutathione-S-transferase activities were decreased significantly as a result of induced oxidative stress. A significant drop in prostatic acid phosphatase activity was observed. Additionally, the results showed some histopathological alterations in the reproductive organs as well as neurological lesions in brain and pituitary glands. In conclusion, CPF and CYP induce deleterious effects on reproductive efficiency of male rats which reflect more obvious impacts when both combined
