Objective: The main objective of the study was to investigate the effect of ethanol seed extracts of Nelumbo nucifera Gaertn (Nymphaeaceae) as antihyperglycemic, anti-hyperlipidemic and antioxidant activity in streptozotocin-induced diabetic rats. Methods: Diabetes was induced in wistar albino rats by administration of streptozotocin (single intraperitoneal dose of 60 mg/kg B. W). The ethanol extract of N. nucifera seed at a dose of 200 and 400 mg/kg body weight was administrated at a single dose per day to STZ induced diabetic rats for a period of 42 d. The different pharmacological parameters were evaluated. The effect of ethanol seed extract of N. nucifera on insulin, blood glucose, urea, creatinine, HbA1C, serum protein, albumin, globulin, serum enzymes, serum lipid profiles, lipid peroxides (LPO) and other antioxidant enzymes like catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD) and reduced glutathione (GSH) were measured in the diabetic rats. Results: In the acute toxicity study ethanol seed extract of Nelumbo nucifera were non-toxic at 2000 mg/kg in rats. The ethanol extract of Nelumbo nucifera seed showed significant reduction in blood glucose (p<0.05), serum enzymes (SGPT, SGOT, ALP) (p<0.05), lipid parameters (TC, TG, VLDL, LDL) (p<0.05) except HDL and significantly increased insulin (p<0.05), HDL (p<0.05), GPx, GSH, SOD and CAT (p<0.05) at the dose of 400 mg/kg when compared with the diabetic-induced control. Conclusion: The present study suggested that the NNSE has significant (p<0.05) antihyperglycemic, antihyperlipidemic and antioxidant activity in STZ induced diabetic rats. These results clearly indicate that Nelumbo nucifera is effective against free radical-mediated diseases, thus replacing the synthetic ones.

This study is aimed at comparing the antidiabetic and antioxidant potential of fenugreek and buckthorn which are commonly used in modulating diabetes in the Middle East. In this study, the antioxidant and antidiabetic activity of the aqueous extracts of the leaf and seed of fenugreek and buckthorn was tested in streptozotocin-induced diabetic male rats fed with a fat-rich diet for 8 weeks. Thirty-six male albino rats were divided into 6 groups ( n = 6 ); the 1st group was the negative control. Diabetes was induced in the other 30 rats using streptozotocin, which were then divided into 5 groups; the 2nd was the untreated positive diabetic group, the 3rd was treated with fenugreek leaf aqueous extract, the 4th was treated with the fenugreek seed aqueous extract, the 5th was treated with buckthorn leaf aqueous extract, and the 6th was treated with buckthorn seed aqueous extract. The positive control group showed an increase in blood sugar, glycated hemoglobin, liver function enzymes, lactate dehydrogenase, kidney indices, total cholesterol, triglycerides, low- and very-low-density lipoprotein, immunoglobulins, and lipid peroxidation and a decrease in high-density lipoprotein, albumin, and antioxidant activity. The histology of the liver and testes showed severe histopathological alterations. Rats of groups 4-6 that were treated with the aqueous extract of the leaf and seed extract of fenugreek and buckthorn showed improvement of all biochemical and histopathological parameters. The seed extract of fenugreek and buckthorn showed more antioxidant activity than their leaves.

Dates are a component of dates that have 6.10-11.47% of all dates. In some countries, dates are a major problem in the processing industry because the seeds currently only become waste. This research was conducted to determine the solvent that produces the largest yield using the soxhletation method and determine its antioxidant activity. Dates are disoxletasion with a variety of solvents n-hexane, diethyl ether, and ethanol. Also carried out the variation of extraction time for 1.5; 2; and 2.5 hours. Based on the results of the study showed that the results of the largest date seed extract were obtained using ethanol of 4.93% for 2.5 hours. Antioxidant activity of date palm seed extract (IC50) using DPPH ethanol extract method has IC50 of 2.27 ppm; n-hexane extract 8396.56 ppm; and diethyl ether extract 9826.88 ppm. Inhibition of date palm seed extract using the TBA method derived from ethanol extract of 98.22% at a concentration of 250 ppm. The results of the analysis using LC-MS showed that ethanol extract was thought to contain 13-hydroxyabscisic acid compound, 1-ethylidene-4-methylidene-2oxo-7-(propan-2-il)-tocahydro-1H-indent-5-il- 3-methylpent-2-enoate, 12-hydroxy-acid (8,10,14) -eicosatrienoic and 1-hydroxy-3- (pentadecanoiloxy) propan-2-il (5,8,11,14) -icosa-5, 8,11,14-tetraenoate. The results of the analysis using GC-MS on n-hexane extract showed 2,4-di-tert-butylphenol and 9-octadecenoic acid. Diethyl ether extract contains 9-octadecenoic acid and mono (2-ethylhexyl) benzocarboxylic acid.
