Impact of airborne zinc pollution on the antimicrobial activity of olive oil and the microbial metabolic profiles of Zn-contaminated soils in an Italian olive orchard

According to Coldiretti, Italy still continues to hold the European Quality record in extra virgin olive oils with origin designation and protected geographical indication (PDO and PGI). To date, 46 Italian brands are recognized by the European Union: 42 PDO and 4 PGI (Tuscan PGI, Calabria PGI; Tuscia PGI and PGI Sicily). Specific regulations, introduced for these quality marks, include the designation of both the geographical areas and the plant varieties contributing to the composition of the olive oil. However, the PDO and PGI assessment procedures are currently based essentially on farmer declarations. Tuscan PGI extra virgin olive oil is one of the best known Italian trademarks around the world. Tuscan PGI varietal platform is rather wide including 31 specific olive cultivars which should account for at least 95% of the product. On the other hand, while the characteristics of other popular Italian extra virgin olive oils (EVOOs) cultivars from specific geographical areas have been extensively studied (such as those of Coratina based blends from Apulia), little is still known about Tuscan PGI EVOO constituents. In this work, we performed, for the first time, a large-scale analysis of Tuscan PGI monocultivar olive oils by 1H NMR spectroscopy and multivariate statistical analyses (MVA). After genetic characterization of 217 leaf samples from 24 selected geographical areas, distributed all over the Tuscany, a number of 202 micro-milled oil samples including 10 PGI cultivars, was studied. The results of the present work confirmed the need of monocultivar genetically certified EVOO samples for the construction of 1H-NMR-metabolic profiles databases suitable for cultivar and/or geographical origin assessment. Such specific PGI EVOOs databases could be profitably used to justify the high added value of the product and the sustainability of the related supply chain.

The aim of the work is to assess the antimicrobial activities of Cell Free Supernatants (CFS) and Membrane Vesicles (MVs), produced by Lactobacillus reuteri DSM 17938, versus Gram-positive and Gram-negative bacteria and investigate their metabolic profiles. The Minimum Inhibitory Concentration was determined through the broth microdilution method and cell proliferation assay while the Minimum Bactericidal Concentration was determined by Colony Forming Units counts. The characteristics of the antimicrobial compounds were evaluated by pH adjustments, proteinase treatment, and size fractionation of the CFS. The cytotoxicity of CFS was tested on two human cell lines. A detailed snapshot of the L. reuteri metabolism was attained through an untargeted metabolic profiling by means of high resolution Fourier Transform Ion Cyclotron Resonance Mass Spectrometry (FT-ICR MS) coupled with Electrospray Ionization Source (ESI). The results showed (i) a greater efficacy of CFS and its fractions towards Gram-negative compared to Gram-positive bacteria; (ii) an antimicrobial effect related to pH-dependent compounds but not to MVs; (iii) a molecular weight < 3 KDa as well as an a non-proteinaceous nature of the antimicrobial compounds; and (iv) more than 200 and 500 putative metabolites annotated in MVs and supernatants, covering several classes of metabolites, including amino acids, lipids, fatty and organic acids, polyalcohols, nucleotides, and vitamins. Some putative compounds were proposed not only as characteristic of specific fractions, but also possibly involved in antimicrobial activity.

The survival of foodborne pathogens in aqueous extracts of olive oil, virgin olive oil, vinegar, and several beverages was evaluated. Vinegar and aqueous extracts of virgin olive oil showed the strongest bactericidal activity against all strains tested. Red and white wines also killed most strains after 5 min of contact, black and green tea extracts showed weak antimicrobial activity under these conditions, and no effect was observed for the remaining beverages (fruit juices, Coca-Cola, dairy products, coffee, and beer). The phenolic compound content of the aqueous olive oil and virgin olive oil extracts could explain their antibacterial activity, which was also confirmed in mayonnaises and salads used as food models. Virgin olive oil in mayonnaises and salads reduced the counts of inoculated Salmonella Enteritidis and Listeria monocytogenes by approximately 3 log CFU/g. Therefore, olive oil could be a hurdle component in certain processed foods and exert a protective effect against foodborne pathogens when contaminated foods are ingested.

    Olive oil and olive extracts have been used in folk med­icine since ancient times. Romans and Greeks employed olive extracts to treat many diseases and an extract of boiled olive leaves was administered as a drink to malaria patients during the 19th century. Hence, the Mediterranean countries have cultivated the olive tree (Olea europaea L.) to produce olive oil, table olives and olive leaf extracts for centuries. At present, both olive oil and table olives are important com­ponents of the Mediterranean diet and are largely consumed throughout the world. In addition, there are many enterprises that commercialize olive leaf extracts to treat a myriad of diseases, many of them caused by microorganisms. Recently, the importance of preventive medicine has been gradually recognized in the field of orthopaedic surgery with a concept that peak bone mass should be increased in childhood as much as possible for the prevention of osteoporosis (Ohtani et al., 2009). The aim of the current study is to explore the antimicrobial activity of olive oil against microbial infections. To achieve the aim of research, the researcher applied the exploratory approach where dozens of relevant studies were reviewed and explored in order to collect the results needed to enrich the discussion within the current study. The results of study indicated the efficiency of antimicrobial activity of olive oil against microbial infections.

AbstractFatty acids represent important substrates for obtaining microbial lipases and biosurfactants. The yeast strain CMGBRG5 was identified as Rhodotorula glutinis using the BIOLOG MicroLog System. The opacity screening tests showed that R. glutinis CMGB-RG5 was able to produce lipases in presence of 1% Tween 80 after three days of incubation. Lipase induction was estimated as cell growth in presence of Tween 80, Tween 20, olive oil and tributyrin. After 48 hours, best results were obtained in presence of butyric acid, respectively, oleic acid: 2.2 × 107cells/ml on tributyrin and 1.0 × 107cells/ml on olive oil. Biosurfactant production was evaluated as emulsification index (E24%). After one week, high E24 values were obtained on fried sunflower oil (53%) and olive oil (35%). Crude and concentrated biosurfactants were tested against nine Candida strains. Best antimicrobial activity was observed for [20X] biosurfactants against C. tropicalis, C. guilliermondii and C. krusei. In conclusion, R. glutinis CGB-RG5 shows high potential for using fatty acids from various sources as unique carbon substrates for synthesis of biocompounds with high economic and biotechnological value.

Olive mill wastewater (OMW) is a compound originating from oil mills during oil extraction processes. In the Mediterranean area, more than 30 million m3 of OMW are produced each year, which represents 95-97% of the world production. Such volumes of untreated OMW are usually directly disposed of into drainage systems, water bodies (such as streams, lagoons and ponds) or are sprinkled on soils, causing potentially severe environmental problems to soils and groundwater. Consequently, there is a serious waste management problem related to the olive oil industry, because these practices no longer being acceptable. In the case of on-land OMW disposal, the characterization and the identification of this contaminant in soils is a fundamental task especially with a view to maintaining the integrity and quality of agroecosystems. In recent years, soils have been extensively studied to detect contaminants by using various geophysical methods. Among such techniques, time domain reflectometry (TDR) has shown, in different contexts, evident sensitivity and resolution capability for the characterization of contaminated soil sites. In order to further exploit the potential of the TDR technique, in this study we conducted a series of laboratory-controlled tests to explore how OMW influences the dielectric response of contaminated soils. This investigation led to the development of an empirical dielectric model to estimate the presence of OMW in variably saturated-contaminated soils with different textures and pedological features. In particular, the soils selected belong to two typical pedological units in southern Italy which account for approximately 90% of the Italian olive plantations and one of the highest concentrations of the olive oil industry in Italy. In these districts, as well as in other European and Mediterranean countries, there is the controversial habit to spread OMW on soils.

<p><strong>Objective:</strong> The aim of this study was to evaluate the antimicrobial effectiveness of calcium hydroxide associated with a two conventional (anesthetic liquid and olive oil) and a new vehicle (Triethanolamine). <strong>Material and methods:</strong> Initially, microorganisms were collected from four upper incisors presenting unsatisfactory root canal treatments and persistent apical periodontitis from the same patient using sterile absorbent paper cones placed inside the root canal following initial access and root filling removal. Next, they were grown in a BHI culture medium for 24 h. Afterwards, they were placed in 10 Petri dishes with 3 holes, where different formulations of the medications studied were placed. After 48 and 72 h, readings were taken of the growth inhibition halos. <strong>Results:</strong> After using Kruskal-Wallis test with significance level of 1% (α = 0.01) it was observed that, calcium hydroxide associated with Triethanolamine, produced results similar to those produced by the anesthetic liquid. Both combinations were superior to the association with olive oil. <strong>Conclusion:</strong> Considering the results of this <em>in vitro</em> study, it is possible to consider that Triethanolamine can be a viable alternative to be used as a vehicle associated with the calcium hydroxide.</p><p><strong>Keywords:</strong> Vehicles; Calcium hydroxide; Endodontics.</p>
