Single muscle fibres from the giant barnacle, Balanus nubilis, were chemically skinned (2% Tween 20), then equilibrated for 40 h in salt solutions ranging in ionic strength from 0·025 to 0·6M at pH 7·0. The water content of the fibres and the net charge on the myofilaments increased with increasing salt concentration. Cation accumulation in the fibres was about equal to anion exclusion at all salt concentrations. When an organic solute (trimethylamine oxide, glycine, alanine, serine, proline, or glycerol) in the concentration range from 0·1 to 0·6 M was added to the salt solution, cation accumulation increased and water content decreased. Myofilament architecture was disrupted when the fibres were equilibrated in high salt (> 0·4 M) solutions and preserved when 0·5 M-triethylamine oxide was also added. The results are consistent with the view that organic solutes enhance the association between the fixed charge sites and their counterions. This hypothesis is examined quantitatively using the Oosawa relationship between the volume and the counterion association for cylindrical polyelectrolytes. The results illustrate that organic solutes can influence fibre volume in a way other than through osmo-regulation.

The potential role of naringenin (NAR), a natural flavonoid, in the treatment of chronic wound has prompted the present research to deliver the drug in nanoemulsion (NE) form, where synergistic role of chitosan was achieved through development of chitosan-coated NAR NE (CNNE). The NE consisted of Capryol 90, Tween 20 and Transcutol P, which was fabricated by low-energy emulsification method to encapsulate NAR within the oil core. The optimization of the formulated NEs was performed using Box–Behnken statistical design to obtain crucial variable parameters that influence globule size, size distribution and surface charge. Finally, the optimized formulation was coated with different concentrations of chitosan and subsequently characterized in vitro. The size of the CNNE was found to be increased when the drug-loaded formulation was coated with chitosan. Controlled release characteristics depicted 67–81% release of NAR from the CNNE, compared to 89% from the NE formulation. Cytotoxicity study of the formulation was performed in vitro using fibroblast cell line (NIH-3T3), where no inhibition in proliferation of the cells was observed with CNNE. Finally, the wound healing potential of the CNNE was evaluated in an abrasion-created wound model in experimental animals where the animals were treated and compared histologically at 0 and 14 days. Significant improvement in construction of the abrasion wound was observed when the animals were treated with formulated CNNE, whereas stimulation of skin regeneration was depicted in the histological examination. Therefore, it could be summarized that the chitosan coating of the developed NAR NE is a potential platform to accelerate healing of wounds.

Abstract Nonsteroidal anti-inflammatory hydrophobic drugs (NSAIDs) are amongst the most commonly given categories of drugs worldwide in the treatment of pain, irritation and some of them even fever in many conditions. Critical micelle concentration of NSAIDs with Naproxen sodium (NS) and its mixture with non-ionic surfactants i.e. Tween 20, Tween 40 and Tween 60 was investigated at 293 K, 303 K, 313 K and 323 K at different concentrations using surface tension and dynamic laser light scattering (DLS) techniques. Due to amphiphilic behaviour in aqueous solution NS form aggregates at sufficiently high concentration. Thermodynamic/adsorption properties like free energy of micellization (ΔGmic), enthalpy of micellization (ΔHmic), entropy of micellization (ΔSmic), Γmic and area per molecule (A2) of NS in the presence of surfactants were also measured at different temperatures. The results showed that the presence of surfactants favoured the ΔGmic and become more enhanced with increase in temperature. Further the solubility of drug is more favourable with increase in polyethylene chain in basic surfactant molecules i.e. 20–60, which indicates that Tween-60 enhanced the solubility of NS more comparatively to Tween-40 and Tween-20 and may be applied as best additive for solubilisation of NS.

Strong aggregation occurred in the refolding route of arginine kinase (AK) denatured with 3 mol GdnHCl/L (GdnHCl, guanidine hydrochloride). The activity recovery of GdnHCl-denatured AK was very low and dependent on the protein concentration in the process of refolding. For denatured AK at 1.2 µmol/L concentration, the recovered activity yield was about 45.2% of the native enzyme, whereas at 5.2 µmol/L the activity recovery yield was only 20% of native activity. The nonionic detergent Triton X-100 and Tween 20 (≤100 mmol/L concentration) not only effectively blocked the aggregation but also enabled the denatured AK to recover most of its native activity. The kinetics of aggregate solubilization showed that there was an induction phase dependent on the detergent, but there was no dependency when detergent was absent. The apparent activity recovery had a cooperative relation with detergents in the process of refolding, which suggested the existence of some interaction between the detergent and the refolding intermediate. On the basis of the study results, a scheme of refolding was proposed.Key words: arginine kinase, guanidine-denatured, refolding, detergent, aggregation.

Background: The role of cerebrospinal fluid (CSF) alpha-synuclein as a potential biomarker has been challenged mainly due to variable preanalytical measures between laboratories. To evaluate the impact of the preanalytical factors contributing to such variability, the different subforms of alpha-synuclein need to be studied individually.Method: We investigated the effect of exposing CSF samples to several preanalytical sources of variability: (1) different polypropylene (PP) storage tubes; (2) use of non-ionic detergents; (3) multiple tube transfers; (4) multiple freeze-thaw cycles; and (5) delayed storage. CSF oligomeric- and total-alpha-synuclein levels were estimated using our in-house sandwich-based enzyme-linked immunosorbent assays.Results: Siliconized tubes provided the optimal preservation of CSF alpha-synuclein proteins among other tested polypropylene tubes. The use of tween-20 detergent significantly improved the recovery of oligomeric-alpha-synuclein, while multiple freeze-thaw cycles significantly lowered oligomeric-alpha-synuclein in CSF. Interestingly, oligomeric-alpha-synuclein levels remained relatively stable over multiple tube transfers and upon delayed storage.Conclusion: Our study showed for the first-time distinct impact of preanalytical factors on the different forms of CSF alpha-synuclein. These findings highlight the need for special considerations for the different forms of alpha-synuclein during CSF samples’ collection and processing.

The binding of fibronectin to gelatin-agarose was strictly dependent on pH, having a pH optimum of 7-9. The binding was strongly inhibited by increasing ionic strength. A chemical modification of lysyl and arginyl groups of fibronectin abolished the binding activity. The anionic detergents sodium dodecyl sulphate and sodium deoxycholate in concentrations of 10-100mM had the same effect. The binding was not affected by the non-ionic detergents Triton X-100, Tween 20 or Lubrol WX. The results demonstrate an important role of ionic interactions in the binding of fibronectin to gelatin. Absence of inhibition by non-ionic detergents suggests that hydrophobic interactions contribute relatively little to the binding of fibronectin to gelatin.
