(HP) infection was performed using invasive and non-invasive methods. The study group consisted of 95 patients with dyspepsia. HP infection was detected in 47 patients (49.4 %). The expediency of using a set of diagnostic methods for detecting HP (PCR, immunochromatographic, bacteriological and method for determining urease activity) is proved. Most often (100 %) in patients HP infection was detected in biopsies using the PCR method. Somewhat less frequently it was detected when examining biopsies with an invasive biochemical method (AMA RUT Reader) (82 %) and fecal immunochromatographic method (83 %). Despite the fact that helicobacteriosis was detected bacteriologically in a small number of patients (24 %), this method is of particular value, since it allows you to assess the sensitivity to antimicrobial drugs and probiotics, and does not give false positive results.

ABSTRACT The authors studied the correlation between cortisol production, as measured by an isotope dilution method, and the urinary excretion of total and free Porter-Silber chromogens, as well as of 17-ketogenic steroids. Although a significant correlation exists between total Porter-Silber chromogens, 17-ketogenic steroid excretion and cortisol production, discrepancies are occasionally observed. Hence, different colorimetric methods should be used to assess the glucocorticoid activity of the adrenal cortex.

Diseases of periodontal tissues occupy one of the leading positions among modern dental problems, namely the multifactorial nature of these diseases. In modern dental science, the issue of the development of periodontal pathology against the background of somatic pathology and risk factors remains relevant. Pathology of periodontal tissues in 68–90 % of cases is accompanied by chronic diseases of the gastrointestinal tract. Today, there is no doubt that Helicobacter pylori infection can be present in the biotopes of the oral cavity and can affect the course of periodontal pathology. As you know, smoking is one of the important risk factors for the development of inflammatory-dystrophic diseases of periodontal tissues, which can aggravate the course of the latter. The purpose of the work is to determine the prevalence of oral Helicobacter pylori infection in tobacco-dependent patients with chronic generalized periodontitis on the background of chronic hyperacid gastritis during treatment. Patients who received the proposed therapeutic and prophylactic complex (ultraphonophoresis procedures with the created gel «Apisan», and probiotic drug BioGaia ProDentis and angioprotective drug of natural origin — Detralex) showed a gradual decrease in the level of total urease activity and, as a consequence, a decrease the prevalence of Helicobacter pylori infection in the oral cavity according to the results of a urease rapid test with material from the oral cavity, both in the presence of a risk factor — smoking, and in its absence. The use of the proposed therapeutic and prophylactic complex proved to be effective in reducing the prevalence of oral Helicobacter pylori infection in smoking patients and patients who do not smoke, with chronic generalized periodontitis against the background of chronic hyperacidal gastritis associated with Helicobacter pylori.

The suitability of sedimentary urease activity as a potential tracer for sewage outfall plumes has been examined. Enzyme activity is readily measured in the sediments by a relatively simple procedure and results may be obtained within a few hours of sampling. The results of urease measurements in areas around point source discharges were compared with bacteriological data for the same areas. Three areas were selected for study: a discharge of untreated sewage into a harbour, a discharge from a sewage treatment plant to a river and a discharge from a contaminated drain to a small beach. In all cases, positive correlation between the distribution of the two parameters was observed. Urease activity probably reflects the movement of soluble products from the outfall whereas E.coli represents the particulate phase of the discharge. Although further work will be required, urease activity does offer a potential alternative to E.coli for tracing plumes of faecal pollution.

AbstractProtocols for microbially induced carbonate precipitation (MICP) have been extensively studied in the literature to optimise the process with regard to the amount of injected chemicals, the ratio of urea to calcium chloride, the method of injection and injection intervals, and the population of the bacteria, usually using fine- to medium-grained poorly graded sands. This study assesses the effect of varying urease activities, which have not been studied systematically, and population densities of the bacteria on the uniformity of cementation in very coarse sands (considered poor candidates for treatment). A procedure for producing bacteria with the desired urease activities was developed and qPCR tests were conducted to measure the counts of the RNA of the Ure-C genes. Sand biocementaton experiments followed, showing that slower rates of MICP reactions promote more effective and uniform cementation. Lowering urease activity, in particular, results in progressively more uniformly cemented samples and it is proven to be effective enough when its value is less than 10 mmol/L/h. The work presented highlights the importance of urease activity in controlling the quality and quantity of calcium carbonate cements.

Helicobacter pylori (H. pylori) produces urease in order to improve its settlement and growth in the human gastric epithelium. Urease inhibitors likely represent potentially powerful therapeutics for treating H. pylori; however, their instability and toxicity have proven problematic in human clinical trials. In this study, we investigate the ability of a natural compound extracted from Zingiber zerumbet Smith, zerumbone, to inhibit the urease activity of H. pylori by formation of urease dimers, trimers, or tetramers. As an oxygen atom possesses stronger electronegativity than the first carbon atom bonded to it, in the zerumbone structure, the neighboring second carbon atom shows a relatively negative charge (δ−) and the next carbon atom shows a positive charge (δ+), sequentially. Due to this electrical gradient, it is possible that H. pylori urease with its negative charges (such as thiol radicals) might bind to the β-position carbon of zerumbone. Our results show that zerumbone dimerized, trimerized, or tetramerized with both H. pylori urease A and urease B molecules, and that this formation of complex inhibited H. pylori urease activity. Although zerumbone did not affect either gene transcription or the protein expression of urease A and urease B, our study demonstrated that zerumbone could effectively dimerize with both urease molecules and caused significant functional inhibition of urease activity. In short, our findings suggest that zerumbone may be an effective H. pylori urease inhibitor that may be suitable for therapeutic use in humans.
