The effect of oxidation on the iodine values of phospholid in milk, butter and washed-cream serum

SummaryPhospholipid was isolated from milk, butter, and washed-cream serum by solvent extraction followed by simple counter-current distribution and thin-layer chromatography. Iodine values from fresh samples, determined by a micro-Wijs technique, ranged, for the cephalin fraction, from 70 to 86, for the lecithin fraction from 44 to 55, and for the sphingomyelin fraction from 36 to 44. In washed-cream serum, oxidation with copper and ascorbic acid led to reduction in extractable phosphorus, decreased chromatographic mobility of phospholipid and significant falls in the iodine values of the 3 phospholipid fractions. In milk, slight reductions in phospholipid iodine values were observed following copper-catalysed oxidation but they were not consistently significant. Iodine values of butter phospholipids remained unchanged even after gross oxidative quality deterioration.

Objective: To synthesize benzimidazole derivatives, characterize them by 1HNMR and ATIR techniques and evaluate their antioxidant activity. Methods: In the present study 19 benzimidazole derivatives were synthesized by reacting O-phenylenediamine as the primary reactant with different aromatic aldehydes and benzoic acids. Reactions were monitored using thin layer chromatography technique, and the newly synthesized derivatives were characterized by ATIR and 1HNMR techniques. The antioxidant assay was performed using ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)] method and DPPH [2,2-diphenyl-1-picrylhydrazyl] method. Results: Compounds BNZ-1, BNZ-2, BNZ-3, BNZ-9, and BNZ-10 showed comparable antioxidant activity to ascorbic acid at higher dose. Conclusion: The synthesized benzimidazole derivatives have significant radical scavenging potential.

Acetone extracts from the seeds of Pisum sativum L. sensu lato (Leguminoseae) separated by thin layer chromatography revealed the occurrence of hands with chromatographic mobility and color reaction with Fast Blue B characteristic for 1,3-dihydroxy-5-alkyIbenzenes. These polyketide metabolites have been isolated and identified by Spectroscopic means. The occurrence of homologous series of saturated (approximately 70%) and enoic (mono and diunsaturated) homologs with chain length of C15 to C25 has been revealed with C17 as the main homolog

A simplified method for aflatoxin production studies is described. The mold was cultured in 4-dram (15 ml) vials containing 5 ml of yeast extract sucrose broth, and aflatoxin levels were determined by direct spotting of the broth on thin layer chromatography (TLC) plates and quantitating by spectrodensitometry. Equivalent levels of aflatoxins were produced in vials as compared to flasks. When compared to conventional TLC after solvent extraction, direct spotting was rapid, economical and statistically equivalent. Heating broth cultures (121°C, 15 s) before TLC improved the release of aflatoxin from mycelial mats. Aflatoxins were unstable in YES broth during 3 months of frozen storage.

The present communication reports the occurrence of high-mannose oligosaccharides on Tamm-Horsfall glycoprotein prepared from human pooled urine. The Pronase digest of the glycoprotein was fractionated by gel filtration and a high-mannose glycopeptide species was separated from complex-type glycopeptides. When high-mannose glycopeptides were digested with endo-β-N-acetylglucosaminidase H, followed by reduction with [3H]KBH4, three oligosaccharides were resolved by thin-layer chromatography. On the basis of chromatographic mobility and exoglycosidase digestions the composition Man7-, Man6-, and Man5-GlcNAc was assigned to the three oligosaccharides. Man6GlcNAc is by far the major component.

Objective: The main objective was to focus on qualitative and quantitative analysis of isolated okra mucilage by High-performance Thin Layer Chromatography to set up the quality control parameters for the isolated mucilage. Methods: High-performance thin-layer chromatography (HPTLC) were applied for the identification of components present in methanolic and ethanolic fruit extract of okra at 254 nm and 356 nm. Quantitative analysis of amino acid ascorbic acid and total polyphenol content was determined. Results: The results showed that the yield percentage for methanolic and ethanolic fruit extract of the okra fruit mucilage was found to be 13.5 and12.5% respectively. HPTLC determination of methanolic and ethanolic okra fruit extract showed the presence of 8 components with Rf values in the range of 0.14 to 0.62 and 0.14 to 0.54 respectively when detected at wavelengths 254 nm and at 356 nm. The total amino acid in okra fruit methanolic extract was found to be 11.45%w/w. The ascorbic acid content in methanolic okra fruit extract and ethanolic okra fruit extract was found to be 0.24 %w/w and 0.18% w/w respectively. The total phenolic contents (tannic acid equivalents, mg/g) in the methanolic and ethanolic okra fruit extracts were calculated to be 4.6 % w/wand 5.3% w/w respectively. Conclusion: The data revealed dual benefits like it can act as a potential novel functional ingredient with health-promoting application due to the presence of ascorbic acid and total phenolics contents and at the same time the data provided guidelines for quality control parameters for the isolated okra fruit
