The role of aerobic metabolism and intragel oxygen in hypoxia tolerance of three ctenophores: Pleurobrachia bachei, Bolinopsis infundibulum and Mnemiopsis leidyi

Ctenophores are important members of planktonic communities that are often abundant in dysaerobic environments. Previous studies have shown that ctenophores are not adversely affected by extended periods of hypoxia. The three species used in this study, Pleurobrachia bachei, Bolinopsis infundibulum, and Mnemiopsis leidyi, were all able to oxyregulate to very low partial pressures of oxygen (PO2s). These species were found to have mean critical oxygen tensions of 7.7, 10.6, and 7.2 hPa respectively. In general, ctenophores are better oxyregulators than medusae and many species of shrimps, fish and squid. Intragel oxygen was measured using a fibre optic oxygen optode. All these ctenophores have intragel subsurface [O2]s of 5–10% below that of the surrounding seawater. Intragel oxygen measurements of P. bachei showed a gradient of decreasing PO2 from surface tissues to the gut. Specimens of P. bachei over 14 mm in diameter had anaerobic guts. Survival times in anoxia ranged from 0 h for M. leidyi to up to 6 h for P. bachei. Ctenophores rely on aerobic metabolism to tolerate hypoxia.

Background/AimsThe role of DHRS3 in human cancer remains unclear. Our study explored the role of DHRS3 in gastric cancer (GC) and its clinicopathological significance and associated mechanisms.MaterialsBisulfite-assisted genomic sequencing PCR and a Mass-Array system were used to evaluate and quantify the methylation levels of the promoter. The expression levels and biological function of DHRS3 was examined by both in vitro and in vivo assays. A two-way hierarchical cluster analysis was used to classify the methylation profiles, and the correlation between the methylation status of the DHRS3 promoter and the clinicopathological characteristics of GC were then assessed.ResultsThe DHRS3 promoter was hypermethylated in GC samples, while the mRNA and protein levels of DHRS3 were significantly downregulated. Ectopic expression of DHRS3 in GC cells inhibited cell proliferation and migration in vitro, decreased tumor growth in vivo. DHRS3 methylation was correlated with histological type and poor differentiation of tumors. GC patients with high degrees of CpG 9.10 methylation had shorter survival times than those with lower methylation.ConclusionDHRS3 was hypermethylated and downregulated in GC patients. Reduced expression of DHRS3 is implicated in gastric carcinogenesis, which suggests DHRS3 is a tumor suppressor.

Background: Hypoxia is a condition in any stage in the delivery of oxygen to cells which include decreased partial pressures of oxygen, less diffusion of oxygen in the lungs, insufficient hemoglobin, inefficient blood flow to the end tissue, and breathing rhythm. Secretin is an amino acid which plays proper functioning of gastro intestinal system.Methods: The current study was conducted to evaluvate the effect of exogenously administrated secretin on chronic hypoxic damage of brain in rat model. Experimental design consists of control animals, Control animals + secretin hypoxia exposed animals; hypoxia exposed animals +secretin (20ng/kg.bw).Results: The results of this study point to a possible role of Secretin as neuroprotectant.Conclusions: Further research on secretin needs to be conducted in order to confirm the deductions made by this study.

✓ In this analysis, the authors review studies over the last 50 years addressing the association between long-term survival and type of surgical management in adults with supratentorial intermediate or high-grade astrocytomas. Earlier reports are included because they are repeatedly referenced in current works and clearly are an important basis upon which present attitudes are predicated. Because recent work has definitively demonstrated the significance of prognostic variables on outcome, the handling of such factors in studies that investigated survival data according to degree of surgery is emphasized. Study design, experimental methods used, and methods of data analysis are also examined. This analysis shows that there is little justification for dogmatic statements concerning the relationship between increasing patient survival times and aggressive surgical management in adults with supratentorial intermediate or high-grade astrocytomas, if patients receive postoperative radiotherapy.

AbstractThe properties of ferroelectric thin films can be significantly influenced by the presence of point defects. The concentration of vacancies presented in these thin films is known to be one of the key parameters causing the degradation of these films when these films are subjected to polarization reversals.To study the effects of the vacancy concentration on the ferroelectric properties, sol gel PZT films and powders were annealed in different oxygen partial pressures. For the PZT films, the reduction of oxides to pure metals was not observed even with films annealed at 2×10−5 atmosphere of oxygen partial pressure. Samples annealed at low oxygen partial pressure (for instance, 10−3 and 2×10−5 atmosphere), which has more Pb and O2 depletions and consequently has more Pb and O2 vacancies, cannot be switched easily. The ratios of coercive field after and before fatigue increase as the defect concentrations of the annealed samples increase.
