Role of dog behaviour and environmental fecal contamination in transmission ofEchinococcus multilocularisin Tibetan communities

SUMMARYOn the Eastern Tibetan Plateau region (Sichuan province, China) dogs are regarded as important definitive hosts ofEchinococcus multilocularis. We studied dog spatial behaviour in 4 Tibetan villages in order to determine the role of dogs in environmental contamination and their potential interactions with small mammal intermediate hosts. We identified definitive host species andEchinococcusspp. infection status of feces collected in the field by PCR methods and analysed the spatial distribution of canid feces. Nocturnal space utilization of GPS collared dogs in and around villages was also undertaken.E. multilocularisDNA was amplified in 23% of dog feces (n=142) and in 15% of fox feces (n=13) but this difference was not significant. However, dog feces were more frequently observed (78% of collected feces) than fox feces and are therefore assumed to largely contribute to human environment contamination. Feces were mainly distributed around houses of dog owners (0–200 m) where collared dogs spent the majority of their time. Inside villages, the contamination was aggregated in some micro-foci where groups of dogs defecated preferentially. Finally, small mammal densities increased from the dog core areas to grasslands at the periphery of villages occasionally used by dogs; male dogs moving significantly farther than females. This study constitutes a first attempt to quantify in a spatially explicit way the role of dogs inE. multilocularisperi-domestic cycles and to identify behavioural parameters required to modelE. multilocularistransmission in this region.

SUMMARYChub and eels were experimentally infected via intermediate hosts harbouring cystacanths, with Pomphorhynchus laevis alone, or Acanthocephalus anguillae alone, or simultaneously with mixtures of both species in varying proportions, and sampled at 7, 56 or 112 days post-infection. Examination of chub revealed that both species showed low establishment and growth rates, differing markedly from British field data, where chub is apparently one of the most important hosts, and preventing further meaningful experiments. Both species showed higher establishment rates in eels, but A. anguillae both grew faster and matured to a greater extent than P. laevis, reflecting field data. No evidence of any interactions, competitive or otherwise, was found in the mixed infections in eels at low or high infection levels. The results were interpretated with regard to British field data and current theories of the role of interspecific competition in structuring fish helminth communities.

Abstract Background Cattle are intermediate hosts of six Sarcocystis species, among which Sarcocystis hominis and Sarcocystis heydorni can infect humans through the consumption of raw or undercooked meat. In addition to the zoonotic potential, there is increasing interest in these protozoa because of the evidence supporting the role of Sarcocystis spp. in the occurrence of bovine eosinophilic myositis (BEM), a specific inflammatory myopathy which leads to carcass condemnation and considerable economic losses. Actually, all the prevalence studies carried out on cattle in Italy have been based on either morphological or 18S rDNA-based molecular techniques, most likely leading to misidentification of closely related species. Therefore, there is a strong need for new data on the prevalence of the different Sarcocystis spp. in cattle in Italy and their association with bovine eosinophilic myositis. Methods To reach our aim, individual striated muscle samples from BEM condemned carcasses (N = 54) and diaphragm muscle samples from randomly sampled carcasses (N = 59) were obtained from Northwest Italy slaughterhouses. Genomic DNA was extracted and analyzed by multiplex-PCR targeting 18S rDNA and cox1 genes. PCR products amplified using the genus-specific primer set in absence of the specific fragment for S. hirsuta, S. cruzi, S. hominis or S. bovifelis were sequenced to achieve species identification. Results Sarcocystis DNA was detected in 67.8% of the samples from slaughter cattle and in 90.7% of the samples from BEM condemned carcasses. S. cruzi was identified as the most prevalent species in slaughter cattle (61%), followed by S. bovifelis (10.2%), S. hominis (8.5%) and S. hirsuta (1.7%). Notably, among the different Sarcocystis spp. detected, the presence of S. bovifelis and S. hominis was significantly higher in samples isolated from BEM condemned carcasses (46.3% and 40.7% respectively), while there was no statistically significant difference between the presence of S. cruzi or S. hirsuta in BEM condemned carcasses (42.6% and 1.8%, respectively) and randomly sampled carcasses. Furthermore, DNA sequence analysis revealed the presence of a putative new species in two carcasses. Conclusions Our study contributes to updating the data on the prevalence of the different Sarcocystis spp. in cattle in Italy, highlighting the presence of three Sarcocystis spp., S. cruzi, S. hominis and S. bovifelis, in BEM lesions and allowing us to speculate on the possible role of S. hominis and S. bovifelis as the major sarcosporidian species involved in bovine eosinophilic myositis. Antimicrobial resistance (AMR) is a global problem and there has been growing concern associated with its widespread along the animal–human–environment interface. The farm-to-fork continuum was highlighted as a possible reservoir of AMR, and a hotspot for the emergence and spread of AMR. However, the extent of the role of non-antibiotic antimicrobials and other food-related stresses as selective factors is still in need of clarification. This review addresses the use of non-antibiotic stressors, such as antimicrobials, food-processing treatments, or even novel approaches to ensure food safety, as potential drivers for resistance to clinically relevant antibiotics. The co-selection and cross-adaptation events are covered, which may induce a decreased susceptibility of foodborne bacteria to antibiotics. Although the available studies address the complexity involved in these phenomena, further studies are needed to help better understand the real risk of using food-chain-related stressors, and possibly to allow the establishment of early warnings of potential resistance mechanisms.

Although place-based investigations into human phenomena have been widely conducted in the social sciences over the last decades, this notion has only recently transgressed into Geographic Information Science (GIScience). Such a place-based GIS comprises research from computational place modeling on one end of the spectrum, to purely theoretical discussions on the other end. Central to all research that is concerned with place-based GIS is the notion of placing the individual at the center of the investigation, in order to assess human-environment relationships. This requires the formalization of place, which poses a number of challenges. The first challenge is unambiguously defining place, to subsequently be able to translate it into binary code, which computers and geographic information systems can handle. This formalization poses the next challenge, due to the inherent vagueness and subjectivity of human data. The last challenge is ensuring the transferability of results, requiring large samples of subjective data. In this paper, we re-examine the meaning of place in GIScience from a 2018 perspective, determine what is special about place, and how place is handled both in GIScience and in neighboring disciplines. We, therefore, adopt the view that space is a purely geographic notion, reflecting the dimensions of height, depth, and width in which all things occur and move, while place reflects the subjective human perception of segments of space based on context and experience. Our main research questions are whether place is or should be a significant (sub)topic in GIScience, whether it can be adequately addressed and handled with established GIScience methods, and, if not, which other disciplines must be considered to sufficiently account for place-based analyses. Our aim is to conflate findings from a vast and dynamic field in an attempt to position place-based GIS within the broader framework of GIScience.

The role of polychaetes as intermediate hosts of helminth parasites of aquatic vertebrates is reviewed. Some 18–20 species of trematodes and three or four each of cestodes and nematodes are known to have larval stages in these annelids. There are no records of acanthocephalan larvae from polychaetes. Trematoda use polychaetes as first (three species) or second intermediate hosts. The polychaete hosts are found among various genera of Errantia, Sedentaria, and Myzostomida. Definitive hosts are mainly fishes, with a few species of trematodes and one of cestodes developing in birds, and apparently one genus of nematodes (two species) maturing in the lungs of seals. A list of polychaetes and their larval helminth parasites is given.

The Saccharomyces cerevisiae GCR2 gene affects expression of most of the glycolytic genes. We report the nucleotide sequence of GCR2, which can potentially encode a 58,061-Da protein. There is a small cluster of asparagines near the center and a C-terminal region that would be highly charged but overall neutral. Fairly homologous regions were found between Gcr2 and Gcr1 proteins. To test potential interactions, the genetic method of S. Fields and O. Song (Nature [London] 340:245-246, 1989), which uses protein fusions of candidate gene products with, respectively, the N-terminal DNA-binding domain of Gal4 and the C-terminal activation domain II, assessing restoration of Gal4 function, was used. In a delta gal4 delta gal80 strain, double transformation by plasmids containing, respectively, a Gal4 (transcription-activating region)/Gcr1 fusion and a Gal4 (DNA-binding domain)/Gcr2 fusion activated lacZ expression from an integrated GAL1/lacZ fusion, indicating reconstitution of functional Gal4 through the interaction of Gcr1 and Gcr2 proteins. The Gal4 (transcription-activating region)/Gcr1 fusion protein alone complemented the defects of both gcr1 and gcr2 strains. Furthermore, a Rap1/Gcr2 fusion protein partially complemented the defects of gcr1 strains. These results suggest that Gcr2 has transcriptional activation activity and that the GCR1 and GCR2 gene products function together.

Background: Infection is one of the top ten diseases in Indonesia, so that antibiotic therapy is quite high. The increasing use of antibiotics may lead to an increase in the use of irrational antibiotics. Drug Related Problems (DRPs) are events that are not expected from the patient's experience or are suspected to be due to drug therapy so that they have the potential to interfere with the desired healing success. Objective: This study aims to determine the number of occurrences of DRPs which include dosage inaccuracy, namely under and over dose, indication without drug, drug without indication, potential interactions and inaccuracy of drug selection in inpatient pneumonia treatment for children at the Deli Serdang Regional public hospital. In 2019 Method: This study is retrospective in which data is obtained through secondary data in the form of patient medical records for the period January-December 2019 with a cross-sectional study design. The data collection technique was in the form of total sampling, obtained 50 samples that fit the inclusion criteria. Results: Drung Related Problems (DRPs) research that occurred in the under-dose category of drugs (4.0%), and excessive drug doses (2.0%) and no DRPs (94.0%). This shows that the role of pharmacists is important in monitoring patient drug therapy to minimize the occurrence of DRPs.
