Enhanced light-driven catalytic performance of cytochrome P450 confined in macroporous silica

A light-driven approach combined with a macroporous reactor for the enzymatic biocatalytic reaction has been developed by confining the enzyme/photosensitizer nanohybrids in a macroporous material, which exhibits high bio-conversion efficiency due to the fast diffusion and collision between the enzyme/photosensitizer nanohybrid and the substrate in the reactor.

Ordered mesoporous silica, SBA-15 and MCM-41, and three-dimensionally ordered macroporous SiO2 were used as the supports of H4PMo11VO40 heteropolyacid for methacrolein oxidation. The dispersion and structural evolutions of the heteropolyacid along with thermal treatment were investigated. It was found that the heteropolyacid entered the one-dimensional mesoporous channels of SBA-15 and MCM-41, and the crystallization and growth were limited, leading to high dispersion of the heteropolyacid. However, the thermal stability was decreased under high dispersion. The migration of the heteropolyacid was observed to the end of the one-dimensional channels of SBA-15 and the outer surface of MCM-41 with calcination, accompanied by the decomposition of the heteropolyacid and the formation of MoO3. In comparison, the crystallization and growth of heteropolyacid were not limited in the open macropores of three-dimensionally ordered macroporous SiO2. Dispersed particles on the surface of the macropores with size of about 5 nm exhibited a higher thermal stability. The decomposition of the heteropolyacid in the SBA-15 and MCM-41 supported catalysts resulted in the loss of strong acid sites, causing low selectivity to methacrylic acid in methacrolein oxidation. High thermal stability with high exposure of the active sites in the three-dimensionally ordered macroporous SiO2 supported catalyst contributed to the enhancement in the catalytic performance.

“Smart” polymeric microcapsules with excellent permeability of membranes have drawn considerable attention in scientific and industrial research such as drug delivery carriers, microreactors, and artificial organelles. In this work, hybrid hollow polymeric microcapsules (HPs) containing redox-active gold-sulfide bond were prepared with bovine serum albumin, inorganic metal cluster (AuNCs), and poly(N-isopropylacrylamide) conjugates by using Pickering emulsion method. HPs were transferred from water-in-oil to water-in-water by adding PEGbis(N-succinimidylsuccinate). To achieve redox-responsive membrane, the Au-S bond units incorporated into the microcapsules’ membranes, allowed us to explore the effects of a new stimuli, that is, the redox Au-S bond breaking on the microcapsules’ membranes. The permeability of these hybrid hollow polymeric microcapsules could be sensitively tuned via adding environment-friendly hydrogen peroxide (H2O2), resulting from a fast fracture of Au-S bond. Meanwhile, AuNCs and conjugates could depart from the microcapsules, and enhance the permeability of the membrane. Based on the excellent permeability of the membrane, phosphatase was encapsuled into HPs and p-nitrophenyl phosphate as a substrate. After adding 1 × 10−2 and 1 × 10−4 M H2O2, the catalytic efficiency was nearly 4.06 and 2.22 times higher than that of HPs in the absence of H2O2, respectively. Hence, the unique redox-responsive HPs have potential applications in biocatalytic reaction, drug delivery, and materials as well as in bioscience.

ABSTRACT Discovery of the CYP107Z subfamily of cytochrome P450 oxidases (CYPs) led to an alternative biocatalytic synthesis of 4″-oxo-avermectin, a key intermediate for the commercial production of the semisynthetic insecticide emamectin. However, under industrial process conditions, these wild-type CYPs showed lower yields due to side product formation. Molecular evolution employing GeneReassembly was used to improve the regiospecificity of these enzymes by a combination of random mutagenesis, protein structure-guided site-directed mutagenesis, and recombination of multiple natural and synthetic CYP107Z gene fragments. To assess the specificity of CYP mutants, a miniaturized, whole-cell biocatalytic reaction system that allowed high-throughput screening of large numbers of variants was developed. In an iterative process consisting of four successive rounds of GeneReassembly evolution, enzyme variants with significantly improved specificity for the production of 4″-oxo-avermectin were identified; these variants could be employed for a more economical industrial biocatalytic process to manufacture emamectin.
