Effects of pH and aluminium on the growth of temperate pasture species. II. Growth and nodulation of legumes

Ten temperate pasture legumes inoculated with appropriate rhizobia were grown for 31 days in flowing solution culture. Solution ionic strength was approximately 2700 8M and contained inorganic nitrogen (150 , 8M NO3-) only at the commencement of the experiment. Solution pH was maintained at 4.5, 5.0, 5.5 and 6.0. Also, five aluminium (Al) treatments were imposed, with nominal Al concentrations of 0, 3, 6, 12 and 24 8M (2.5, 7.1, 8.3, 11.2 and 24.7 8M Al measured) at pH 4.5. Solution pH <6 . 0 markedly reduced total dry mass (TDM) in all cultivars of white clover (Trifolium repens) cvv. 'Grasslands Pitau, Huia, G18 and Tahora' and red clover (Trifolium pratense) cvv. 'Grassland Turoa and Pawera', and to a lesser extent in the two subterranean clover (Trifolium subterraneum) cvv. 'Tallarook and Woogenellup'. In contrast, solution pH had no effect on the growth of Lotus corniculatus cv. Maitland, while Lotus pedunculatus cv. Maku grew best at pH 4.5. Lotus pedunculatus cv. Maku grew best in solution where the sum of the activities of the monomeric Al species {Alm} was maintained at 5.9 8M. The growth of all other species was decreased with Al in solution, a 50% reduction in TDM being associated with c. 6 8M {Alm] for white clover and subterranean clover, and c. 3 8M in red clover and Lotus corniculatus cv. Maitland.

A flowing solution culture experiment was conducted in which four temperature pasture grasses and 11 temperate pasture legumes were grown in solutions of low ionic strength for 29 days at six aluminium (Al) levels with average Al3+ activities {Al3+} of 1.0, 5.1, 5.9, 9.9, 20.8, 41.6 8M at pH 4.5. Growth of these species was also measured at pH 5.5 in the absence of Al. In the absence of added Al, lucerne (Medicago sativa) cv. Hunter River and phalaris (Phalaris aquatica) cv. Grasslands Maru grew better at pH 5.5 than at 4.5. The reverse was so for ryegrass (Lolium perenne) cv. Grasslands Nui and subterranean clover (Trifolium subterraneum) cv. Tallarook. Increasing the pH from 4.5 to 5.5 had no effect on the growth of the other 11 species and cultivars tested. Micromolar activities of Al (<10 8M) markedly reduced the growth of all species. Four groups were tentatively identified in terms of their tolerance to Al. The two cultivars of ryegrass and the two red clover (Trifolium pratense) cultivars, together with Lotus corniculatus cv. Maitland, were relatively sensitive to Al. A 50% reduction in total dry matter (TDM) was associated with {Al3+} of 2-3 8M. The four white clover (Trifolium repens) cultivars, tall fescue (Festuca arundinacea) cv. Grasslands Roa, phalaris cv. Grasslands Maru, and Lucerne cv. Hunter River were intermediate in their tolerance, with {Al3+} of 3-5 8M reducing TDM by 50%. The two subterranean clover cultivars were more tolerant to Al than white clover (50% TDM reduction at 5-6 8M {Al3+}. Lotus pedunculatus CV. Grasslands Maku was exceptional in its tolerance to Al. Its growth was unaffected by {Al3+} up to 5 8M and 50% growth reduction was associated with {Al3+} of 7 8M The results of this study emphasize the need to conduct solution culture experiments at realistic solution concentrations and ionic strengths when examining the effects of Al on plant growth.

Studies have been made of the digestion and metabolism in Merino wethers of the isoflavones in subterranean clover (Trtfolium subterraneurn cv. Clare) and red clover (T. pratense). The dietary intake of isoflavones with both clovers was about 9 g per day. With the subterranean clover, the isoflavones were predominantly genistein and biochanin A, and slight teat length increases in the wethers ingesting this clover indicated a low level of oestrogenicity. With the red clover formononetin represented 60% of the isoflavone present and the wethers on this diet exhibited maximal teat length increases indicating a high level of oestrogenicity. Less than 1 % of the daily intake of the isoflavones was excreted as such in the faeces and urine; hence most of these compounds were metabolized or retained in the sheep. The dietary isoflavones were found to disappear rapidly from the rumen, and it was estimated by using marker techniques that the removal of these compounds from the stomach (rumen, reticulum, omasum, and abomasum) was virtually complete. Equol (7,4'-dihydroxyisoflavan), a metabolite of formononetin, was the predominant phyto-oestrogen in the digesta and excreta when red clover was given. The excretion of 3.9 g/day of this compound, mainly in urine, was equivalent to 70% of the intake of formononetin. It was calculated that about 86% of the equol produced in the rumen was absorbed from that organ; the mean residence time for equol in the rumen was estimated to be 1.7 hr. The isoflavones were present in blood plasma mainly in conjugated forms. Equol predominated with both clovers. The levels of equol were much lower with the subterranean clover than with the red clover diet; the concentrations of the conjugated form were respectively 13 and 300-440 �g/100 ml. Equol in the free form, although not detectable with the subterranean clover, was present at 4-10 �g/100 ml with red clover. The data were considered to be consistent with the conclusion that equol accounts for most of the phyto-oestrogenic activity in sheep fed on clovers containing high levels of formononetin.

Alternate 2·4 m wide strips of a mixture of birdsfoot trefoil (Lotus corniculatus L.) with white clover (Trifolium repens L.) (BW), and of red clover (Trifolium pratense L.) (RC) were offered in the proportions (by area) of 0·80[ratio ]0·20, 0·67[ratio ]0·33, 0·33[ratio ]0·67 and 0·20[ratio ]0·80 to yearling heifers in groups of three for periods of 3 days over four replicates in time, balanced for effects of previous treatments. Observations on the distribution of grazing activity and biting rate were made over 3 h periods each evening, and on the morning of Day 2. Biting rates were consistently higher on BW than RC (52·3 v. 46·3±0·59 bites/min, P<0·001). Initially the animals showed a strong tendency to concentrate grazing on the sward of smaller proportional area, but the selection coefficient (log θ) changed from positive to negative with time as herbage on minority strips was depleted. These results are discussed in the context of concepts of partial grazing preference.

A collection of subterranean clover (Trifolium subterraneum L.) genotypes from Sardinia was evaluated for the content of oestrogenic isoflavones to assess differences in total and individual compounds. Daidzein, genistein, and their respective 4′-methoxy derivatives biochanin A and formononetin, were detected in leaves and quantified by GC/FID and GC/MS methods. A large variation among genotypes for individual compounds and for the total isoflavone concentration was observed. Total isoflavones were measured in the range 0.8-13.4 mg/g fresh weight (0.3-4.5% dry matter). Daidzein was usually present in lower amount compared to the other isoflavones. Genistein and biochanin A were found in most cases in higher concentration than formononetin. Phyto-oestrogens have been shown to have both oestrogenic and anti-oestrogenic properties, and their effects entail a positive role for the human health. T. subterraneum revealed the presence of higher concentration of isoflavones compared to other sources of these compounds, e.g. soybean (Glycine max (L.) Merrill) and red clover (Trifolium pratense L.). This study provided information on the biodiversity of the natural strains from Sardinia, emphasizing the importance of these genetic resources for their variation in the amount of total and individual isoflavones.

Seedlings of 3 perennial grasses, Danthonia linkii Kunthcv. Bunderra, D. richardsonii Cashmore cv. Taranna(wallaby grasses), and Phalaris aquatica L. cv. Sirosa,were each grown in replacement series mixtures with seedlings ofTrifolium repens L. (white clover),Trifolium subterraneum L. var. brachycalycinum (Katzn.et Morley) Zorahy & Heller cv. Clare (subterraneanclover), and Lolium rigidum L. (annual ryegrass). Plantswere sown 5 cm apart in boxes (45 by 29 by 20 cm) at a density of 307plants/m2. Maximum likelihood estimates were usedto derive parameters of a non-linear competition model using the dry matterweights of perennial grasses and competitors at 3 harvests, approximately 168,216, and 271 days after sowing. Intra-plant competition was examined inmonocultures of each species, grown at plant spacings of 2, 5, and 8 cm apartwith plants harvested at the above times.Competition occurred in all perennial grass–competitor mixtures, exceptin those of each perennial grass with white clover and thephalaris–subterranean clover mixture (Harvest 1) and those withD. richardsonii and phalaris grown with white clover(Harvest 2). For D. richardsonii (Harvests 1 and 2) andD. linkii (Harvest 1 only) grown with white clover andthe phalaris–subterranean clover (Harvest 1), the two species in themixture were not competing. In the phalaris–white clover mixture, eachspecies was equally competitive (Harvests 1 and 2). These differences incompetition and aggressiveness reflected differences in individual plantweights in monocultures where there was an effect (P < 0.05) of species ondry matter weight per box, but no significant effect of plant spacing.These data indicated that for successful establishment,D. richardsonii and D. linkiishould not be sown in swards with either subterranean clover or white clover,or where populations of annual ryegrass seedlings are likely to be high.Phalaris was more compatible with both white clover and subterranean clover,but aggressively competed with by annual ryegrass.

SummaryDuring 3 years, three groups of 20 Scottish half-bred ewes were grazed on perennial ryegrass, white clover or red clover. During this period the two legumes were oestrogenic, in terms of teat-length extension of wethers, but the perennial ryegrass was not. The white clover was only markedly oestrogenic when there was a high proportion of diseased leaves. The mean litter size and lambing percentage of the ewes grazed on red clover was significantly reduced. White clover, although it was oestrogenic, had no effect on mean litter size or lambing percentage. The mean litter size of the ewes on the perennial ryegrass was 2·35, 2·30 and 2·24, on the white clover was 2·20, 2·33 and 2·23 and on the red clover was 1middot;60, 1·67 and 1·78 for the three lambings. Lambing percentage of the ewes on the perennial ryegrass was 175, 225 and 218, on the white clover was 210, 222 and 189, and on the red clover was 160, 128 and 118 for the three lambings. The lambing date of the ewes grazing on the red clover was significantly later at the third lambing.

The effects of manipulation of sward maturity on ingestive behaviour and diet selection of grazing cattle were tested in two experiments using legumes which are equally preferred by cattle. Yearling heifers, in groups of three, grazed plots formed by alternate 2·4 m wide strips of a mixture of birdsfoot trefoil (Lotus corniculatus L.) and white clover (Trifolium repens L.) (BW) and strips of red clover (Trifolium pratense L.) (RC). The treatments comprised four combinations of each of the two swards at two contrasting levels of maturity/height replicated over four successive 3-day periods, in an order which balanced the effect of previous treatments. Observations of the distribution of grazing activity and biting rate were made over 3-hour periods each evening and in the morning of Day 2.Rate of biting was generally greater on immature than mature swards, and on BW than on RC, with little evidence of interactions between these effects. The animals demonstrated partial preference throughout the two studies, the proportion of grazing time devoted to BW being 0·40±0·010 in Expt 1 and 0·47±0·017 in Expt 2. The distribution of grazing time between sward type and maturity combinations was influenced progressively by herbage mass and height contrasts as strips were grazed down. An overall analysis using the results from these studies and a companion experiment indicated differences between studies in partial preference for BW and RC which appeared to be related to variations in sward morphology.

An in sacco (nylon bag) technique was used to estimate the degradability of dry matter (DM) and crude protein, and to estimate the effective rumen degradability of protein (ERDP), for 3 irrigated clover herbages. Pasture characteristics (nutritive values and leaf: stem ratios) were also described, and relationships to ERDP established. The nutritive value characteristics and degradabilities of white clover (Trifolium repens L.), Persian clover (Trifolium resupinatum L.), and subterranean clover (Trifolium subterraneum L.) were also compared for various regrowth periods (of 3, 4, 6, and 12 weeks). Nutritive value characteristics of clover herbages varied from 9.9 to 11.9 MJ/kg DM for metabolisable energy, 153 to 304 g/kg DM for crude protein, and 209 to 377 g/kg DM for neutral detergent fibre. The leaf: stem ratio correlated well with the crude protein content of the clovers, with herbages consisting of more leaf generally having superior crude protein content (R2 = 0.64, P < 0.001). The effective rumen degradability of protein for clover herbages ranged from 60 g/kg DM for mature (12 week regrowth) subterranean clover to 195 g/kg DM for vegetative (3 weeks regrowth) Persian clover. For clover herbages with a 3-week regrowth period, after initial cutting to 5 cm, and at ruminal outflow rates of 0.08/h, the effective rumen degradability of protein varied from 136 to 195 g/kg DM. A positive relationship between crude protein and ERDP (R2 = 0.82) suggested that ERDP could be estimated for clover herbages that have been previously assessed for crude protein content, obviating the need for in sacco studies. Calculated degradabilities were lower for all clovers when higher ruminal outflow rates were assumed. Using the metabolisable protein system, metabolisable protein supply and metabolisable protein in excess of animal requirements were calculated. Ruminal losses of nitrogen were also estimated for cows consuming white clover of varying regrowth periods in both early and late lactation. It was concluded that metabolisable protein supply is unlikely to limit production in these examples. At regrowth periods of 3 weeks, metabolisable protein contributions from microbial and dietary sources were similar, and twice that required by the animal. Ruminal losses of nitrogen were substantial and amounted to 66—23% of nitrogen intake. In the experiments reported here, if all of the energy required to excrete excess protein (as urea) could instead be used for the production of milk, cows may have produced 0.5mp;mdash;2.0 kg more milk per day. Such losses could potentially be reduced if the protein content and/or degradabilities of clover herbages were reduced, and/or energy rich supplements were offered.

SUMMARYRed clover (Trifolium pratense L.) silage usually contains lower contents of non-protein nitrogen (NPN) compared with other forage legumes. This is often attributed to the polyphenol oxidase (PPO) activity in red clover, although in most field studies the PPO activity was not measured. Therefore, a laboratory ensiling experiment with three red clover cultivars and one white clover cultivar as control grown in two management systems (with and without mechanical stress) over 2 consecutive years was conducted. Fresh, wilted and ensiled clover herbage was sampled at four cutting dates per year to determine the crude protein (CP) fractions according to the Cornell Net Carbohydrate and Protein System. The specific PPO activity was measured photometrically in fresh clover leaves. The content of CP fraction A (NPN) increased from fresh over wilted to ensiled clover herbage at the expense of the content of CP fraction B (true protein), irrespective of species, cultivar and year. The most important source of variation for all CP fractions and the calculated rumen-undegradable protein contents was generally the herbage condition, except for CP fraction C (unavailable protein). White clover silage consisted of higher contents of CP fraction A and lower contents of CP fraction B3 in CP compared with red clover silage. As a result, the calculated rumen-undegradable protein content of white clover silage was lower than that of all red clover cultivars. In conclusion, the extent of proteolysis during ensiling among the silages made from the herbage of different red clover cultivars was primarily influenced by the stage of maturity at harvesting and the degree of wilting at ensiling. The variation in specific PPO activity could not be related to the extent of proteolysis.
