Microorganisms, particularly bacteria, in the ruminant's rumen are valuable genetic resources that many scientists interested in. In recent years, the application of next-generation sequencing technologies allows direct decoding an extracted DNA metagenome in each ecological community without culture, increasing the efficiency of exploiting interested genes. Notably, the quantity and quality of extracted DNA play an important role in getting a reliable metagenome database. In this study, DNA metagenome from goat rumen fluid was extracted by five different methods RBB (repeated bead beating plus column), RBBC (repeated bead beating), PSP1, PSP2 (PSP®Spin Stool DNA Kit, protocol 1, 2, Germany) và QIA (QIAamp® DNA Stool Mini Kit, Germany). The results showed that DNA metagenome obtained by all methods had A260/280 greater than 1.8. DNA extracted by the RBB method had high DNA concentration but low A260/230 values (less than 1.4) and still contained Taq polymerase inhibitor. After purifying by QIA column, A260/230 values of RBB-extracted DNA significantly increased up to 2.0 and Taq polymerase inhibitor in samples were removed. However, the concentrations decreased by 57% that nearly equivalent to concentration of DNA metagenome obtained by QIA. The method using PSP®Spin Stool DNA kit produced the highest DNA concentrations (from 149.7 to 195.5 ng/µl) with A260/280 ratios of 1.9 and A260/230 ratios of 1.8 to 1.9. Morever, this method was able to remove polymerase inhibitor and be performed on short time. Therefore, the PSP®Spin Stool DNA kit is a suitable method for DNA metagenome extraction of bacteria from goat rumen. DNA obtained by this method fulfilled all criteria about quality and concentration for sequencing by next-generation sequencing Illumina.

Three major distinct types of lysozymes have been identified in the animal kingdom and most lysozymes cloned from ruminants belong to the chicken-type (c-type). In this study, a new c-type lysozyme gene, named LZRLyz, was cloned and sequenced from the Lezhi black goat rumen. The LZRLyz cDNA has a 444 bp open reading frame (ORF) encoding a 147 amino acid polypeptide. The encoded polypeptide is predicted to have an 18 amino acid signal peptide, and a 129 amino acid mature protein with an isoelectric point (pI) of 6.08. The LZRLyz amino acid sequence shares 70.27% identity with the Capra hircus blood lysozyme and is grouped with other ruminants c-type lysozymes using the phylogenetic tree estimated by Neighbor-Jointing method. The recombinant expressed LZRLyz protein (pET-rLZR) shows a molecular mass of ∼33 kDa, which is consistent with the predicted fusion protein molecular mass and shows antimicrobial activity. Quantitative real-time RT-PCR analyses revealed that LZRLyz transcripts are expressed in all tested tissues with the predominant expression being observed in rumen and the weakest one in spleen. Results of this study suggest that the LZRLyz gene represents a new c-type lysozyme gene that likely functions in Lezhi black goat host immunity and digestive systems.

The production of starch generates, as a by-product, the cassava wastewater (manipueira), which can be treated by anaerobic digestion to provide biogas and minimize its polluting potential. The most commonly utilized biomass in the anaerobic digestion is the anaerobic sludge. The literature presents, as an alternative to sludge, bovine manure and ruminal fluids, being scarce the studies with the cassava wastewater. This research evaluated the influence of temperature on the microbial ability of cattle and goat rumen in anaerobically biodegrading the manipueira in substitution to the anaerobic sludge. The cattle and goat rumen specific methanogenic activities (SMA) were compared with that of the anaerobic sludge. Subsequently, by using the inoculum which had the best SMA results, cassava wastewater biodegradability tests were performed, investigating the kinetics of the organic matter removal and methane production at 32 ° C and 39 ° C. The bovine rumen presented better results in the SMA (0,315 g COD-CH4 g VSS.d-1) and methane production (1,026 mL). The temperature of 32 °C did not influence the activity of bovine ruminal inoculum as the kinetics of the biodegradation of the manipueira did not differ for the evaluated temperatures (0.1799 d-1 at 32°C and 0.1781 d-1 at 39°C). Bovine rumen achieved glucose reduction of 76% and 80% and methane yield of 77% and 79% for the tests at 32°C and 39°C, respectively. It is inferred that this type of inoculum might be used in reactors of anaerobic digestion processes for the treatment of the cassava wastewater at the ambient temperature of the semiarid region.
