Abstract The effectiveness of two methods of collecting semen from honeybee Apis mellifera drones was compared, and the reasons for problems with ejaculating semen were analysed. Among 275 drones, 100 were stimulated to release semen using a manual method, 100 with the use of chloroform, and from 75 drones the reproductive organs were dissected for analysis and evaluation. It was found that the principal causes of problems that drones had with ejaculating their semen were anatomical changes or a delay in the development of the mucus glands. It was also found that the method employing chloroform was less efficient in the first phase of eversion of the endophallus, compared with the manual method. The method with the use of chloroform allows the determination of the proportion of drones, which do not evert the endophallus because of poor or delayed development of mucus glands, as well as the proportion of drones which evert the organ, but do not ejaculate semen because of the absence of semen in the seminal vesicles.

The effects of veterinary drugs, dietary supplements and non-protein amino acids on the European honey bee (Apis mellifera ligustica Spinola, 1806) ventriculum microbial profile were investigated. Total viable aerobic bacteria, Enterobacteriaceae, staphylococci, Escherichia coli, lactic acid bacteria, Pseudomonas spp., aerobic bacterial endospores and Enterococcus spp. were determined using a culture-based method. Two veterinary drugs (Varromed® and Api-Bioxal®), two commercial dietary supplements (ApiHerb® and ApiGo®) and two non-protein amino acids (GABA and beta-alanine) were administered for one week to honey bee foragers reared in laboratory cages. After one week, E. coli and Staphylococcus spp. were significantly affected by the veterinary drugs (p < 0.001). Furthermore, dietary supplements and non-protein amino acids induced significant changes in Staphylococcus spp., E. coli and Pseudomonas spp. (p < 0.001). In conclusion, the results of this investigation showed that the administration of the veterinary drugs, dietary supplements and non-protein amino acids tested, affected the ventriculum microbiological profile of Apis mellifera ligustica.

O mel é um alimento muito apreciado e de excelente valor nutritivo, constituído por uma mistura complexa de carboidratos, enzimas, aminoácidos, ácidos, minerais, substâncias aromáticas, vitaminas, pigmentos, cera e grãos de pólen. O produto pode apresentar alterações devido a formas inadequadas de armazenamento e conservação. Tais procedimentos ocasionam sua deterioração, comprometendo seriamente o alimento. Quando os méis de abelha são submetidos a condições de temperatura e luminosidade intensa podem sofrer algumas modificações nas suas propriedades físico-químicas. A proposta deste trabalho é verificar a presença e o grau de deterioração dos méis de abelha comercializados em São Luís do Maranhão. Foram realizadas as análises físico-químicas: umidade, acidez, atividade diastásica e hidroximetilfurfural em 30 amostras de méis de Apis mellifera e os valores foram analisados segundo a Instrução Normativa n.º 11 de 2000 do MAPA. As amostras em estudo apresentaram valores fora dos padrões estabelecidos pela legislação, em relação à determinação de atividade diastásica e teor de hidroximetilfurfural, parâmetros físico-químicos que fornecem indicações sobre o grau de conservação e superaquecimento, o que indica que as amostras apresentaram deterioração.Palavras-chave: Mel. The purpose of this study is to verify the presence and degree of deterioration of honeys of bee commercialized in São Luis do Maranhão. Physico-chemical analyses were done: moisture, acidity, diastase activity and hydroxymethylfurfural in 30 honey samples of Apis mellifera and the values were analyzed according to Normative Instruction No. 11 of 2000 of MAPA. The samples studied showed values outside the standards established by legislation in relation to the determination of diastase activity and hydroxymethylfurfural content, physico-chemical parameters that provide indications about the degree of conservation and overheating, which indicates that the samples showed deterioration.Keywords: Honey. The complementary sex determiner (csd) gene plays an essential role in the sex determination of Apis mellifera L. Females develop only if fertilized eggs have functional heterozygous genotypes at this gene whereas males, being haploids, are hemizygous. Two identical csd alleles produce non vital males. In light of the recent decline in honey bee populations, it is therefore important to monitor the allele variability at this gene. In this study, we tested the application of next generation semiconductor-based sequencing technology (Ion Torrent) coupled with environmental honey DNA as a source of honey bee genome information to retrieve massive sequencing data for the analysis of variability at the hypervariable region (HVR) of the csd gene. DNA was extracted from 12 honey samples collected from honeycombs directly retrieved from 12 different colonies. A specifically designed bioinformatic pipeline, applied to analyze a total of about 1.5 million reads, identified a total of 160 different csd alleles, 55% of which were novel. The average number of alleles per sample was compatible with the number of expected patrilines per colony, according to the mating behavior of the queens. Allele diversity at the csd could also provide information useful to reconstruct the history of the honey.
