Lipid droplets (LDs) are essential organelle in most eukaryotes, and tracking intracellular LDs dynamics using synthetic small molecules is crucial for biological studies. However, only a limited number of fluorescent markers that satisfy all requirements, such as the selective staining of LDs, high photostability, and sufficient biocompatibility, have been developed. Herein, we report a series of donor-p-acceptor dyes based on the thiophene-containing fused polycyclic scaffold [1]benzothieno[3,2-<i>b</i>][1]benzothiophene (BTBT), in which either or both thiophene rings are oxidized into thiophene-<i>S</i>,<i>S</i>-dioxide to form an electron-accepting building block. Among these dyes, LAQ1 satisfied all the aforementioned requirements, and allowed us capturing ultra-small LDs on the endoplasmic reticulum (ER) membrane by stimulation emission depletion (STED) microscopy with a super-resolution below the diffraction limit of light. Moreover, the extremely high photostability of LAQ1 enabled recording the lipolysis of LDs and the concomitant lipogenesis as well as long-term trajectory analysis of micro LDs at the single particle level in living cells.

Evidence-based canine decontamination protocols are underrepresented in the veterinary literature. Aerosolized microbiological and chemical contaminants can pose a risk in deployment environments highlighting the need for improved canine field decontamination strategies. Prior work has established the efficacy of traditional, water-intensive methods on contaminant removal from the coat of the working canine; however, it is not known if similar reductions can be achieved with simple field expedient methods when resources are limited. The objective of this study was to measure the reduction of aerosolized contamination via a practical “wipe-down” procedure performed on working canine coats contaminated with a fluorescent, non-toxic, water-based aerosol. Disposable, lint-free towels were saturated with one of three treatments: water, 2% chlorhexidine gluconate scrub (CHX), or 7.5% povidone-iodine scrub (PVD). Both CHX and PVD were diluted at a 1:4 ratio. Treatments were randomly assigned to one of three quadrants established across the shoulders and back of commonly utilized working dog breeds (Labrador retrievers, n = 16; German shepherds, n = 16). The fourth quadrant remained unwiped, thus serving as a control. Reduction in fluorescent marker contamination was measured and compared across all quadrants. PVD demonstrated greater marker reduction compared to CHX or water in both breeds (p < 0.0001). Reduction was similar between CHX or water in Labradors (p = 0.86) and shepherds (p = 0.06). Effective wipe-down strategies using common veterinary cleansers should be further investigated and incorporated into decontamination practices to safeguard working canine health and prevent cross-contamination of human personnel working with these animals.

Several models have been developed to study neutrophil degranulation. At the most basic level, phospholipid vesicles have been used to investigate the lipid interactions occurring during membrane fusion. The two major forms of assays used to measure phospholipid vesicle fusion are based either on the dilution of tagged phospholipids within the membrane of the two fusing partners or the mixing of the aqueous contents of the vesicles. Although problems exist with both methods, the latter is considered to be more accurate and representative of true fusion. Using 8-aminonaphthalene-1,3,6-trisulphonic acid (ANTS) as a fluorescent marker, we have taken advantage of the quenching properties of p-xylenebispyridinium bromide (‘DPX’) to develop a simple aqueous-space mixing assay that can be used with any sealed vesicle. We compared our new assay with more conventional assays using liposomes composed of phosphatidic acid (PA) and phosphatidylethanolamine (PE), obtaining comparable results with respect to Ca2+-dependent fusion. We extended our studies to measure the fusion of neutrophil plasma-membrane vesicles as well as azurophil and specific granules with PA/PE (1:3) liposomes. Both specific granules and plasma-membrane vesicles fused with PA/PE liposomes at [Ca2+] as low as 500 μM, while azurophil granules showed no fusion at [Ca2+] as high as 12 mM. These differences in the ability of Ca2+ to induce fusion may be related to differences observed in whole cells with respect to secretion.

Abstract Background and Aims Renal micropuncture, which requires the direct access to the renal tubules, has been for long time the technique of choice to measure the single nephron glomerular filtration rate (SNGFR) in animal models, but this approach is challenging by virtue of complex animal preparation and numerous careful steps. The introduction of intravital multiphoton microscopy (MPM) permitted to improve the study of renal functions exploiting the high laser penetration and the optical sectioning capacity. Previous MPM studies measuring in vivo the SNGFR relied on fast full frame acquisition during the filtration process obtainable with microscope resonant scanners, which represent optional expensive equipment able to reach very high acquisition speed. In this work we propose an innovative linescan-based MPM method to calculate SNGFR in rodents doable without using the fast acquisition rate offered by resonant scanners. Method An in vivo MPM approach was used to measure the SNGFR in control Munich Wistar Frömter rats (MWF) and to test the feasibility of the innovative linescan approach. In order to validate this method in conditions known for reduced and increased SNGFR, it was applied to ischemia reperfusion injury (IRI) and low-dose dopamine treated conditions, respectively. Results The glomeruli connected to S1 proximal tubules extending at least 100 μm from the exit of the Bowman’s space were chosen for the measurement. A linescan path starting from the urinary pole and crossing many times the tubular lumen orthogonally to the cellular wall was hand drawn. The linescan was acquired soon after a i.v. bolus of low-molecular weight fluorescent marker was injected. The tubular length, the mean diameter and the transit time of the fluorescent marker within two lines of interest (called cross1 and cross2) were measured to obtain the SNGFR. SNGFR measured in control rats was comparable with previous reported data both at MPM and micropuncture. Significantly higher values compared to control were obtained in 3 μg/kg/min dopamine-treated rats. In IRI-treated rats the SNGFR was reduced about 35% compared to the controls. Conclusion The results achieved with our linescan method were quite similar to those obtained with conventional micropuncture, suggesting that the two methods overlap for the normal, dopamine and IRI treatment. Our results show that linescan approach is a promising and cheap alternative to the fast full frame acquisition for the investigation of SNGFR in health and disease, offering results comparable to conventional micropuncture with unprecedent temporal resolution.

Abstract. The Mg/Ca ratio of Foraminifera calcium carbonate tests is used as proxy for seawater temperature and widely applied to reconstruct global paleo-climatic changes. However, the mechanisms involved in the carbonate biomineralization process are poorly understood. The current paradigm holds that calcium ions for the test are supplied primarily by endocytosis of seawater. Here, we combine confocal-laser scanning-microscopy observations of a membrane-impermeable fluorescent marker in the extant benthic species Ammonia aomoriensis with dynamic 44Ca-labeling and NanoSIMS isotopic imaging of its test. We infer that Ca for the test in A. aomoriensis is supplied primarily via trans-membrane transport, but that a small component of passively transported (e.g., by endocytosis) seawater to the site of calcification plays a key role in defining the trace-element composition of the test. Our model accounts for the full range of Mg/Ca and Sr/Ca observed for benthic Foraminifera tests and predicts the effect of changing seawater Mg/Ca ratio. This places foram-based paleoclimatology into a strong conceptual framework.

SummaryGenetic engineering of quintuple transgenic brain tissue was used to establish a low background, Cre-dependent version of the inducible Tet-On system for fast, cell type-specific transgene expression in vivo. Co-expression of a constitutive, Cre-dependent fluorescent marker selectively allowed single cell analyses before and after inducible, tet-dependent transgene expression. Here, we used this method for acute, high-resolution manipulation of neuronal activity in the living brain. Single induction of the potassium channel Kir2.1 produced cell type-specific silencing within hours that lasted for at least three days. Longitudinal in vivo imaging of spontaneous calcium transients and neuronal morphology demonstrated that prolonged silencing did not alter spine densities or synaptic input strength. Furthermore, selective induction of Kir2.1 in parvalbumin interneurons increased the activity of surrounding neurons in a distance-dependent manner. This high-resolution, inducible interference and interval imaging of individual cells (high I5, ‘HighFive’) method thus allows visualizing temporally precise, genetic perturbations of defined cells.

The research aims to assess the effects of climate change on the livelihood of people living around the coastal areas and coping strategies used. The study uses a quantitative method with semi-structured interview questionnaire and convenient sampling method and conducts a survey of coastal community residents in Banjul, Barra, Bakau, Tanji, Sanyang, Gunjur, and Kartong and uses a descriptive analysis. Majority of respondents are self-employed as fishermen, and fish dryers, fish smokers, ship builders and boat riders. Most of the borrowing is done individually and the line of credit is mostly below D20,000 and more than 85% receive less than that amount. More than 60% of the respondents have no access to land and 4.71% stated that they do not have access to water. Most of the respondents rank their friends as their source of borrowing followed by families and banks. About 93% stated that it takes them less than 40 minutes to get to the nearest health facility. Adaptation strategies employed to deal with health problems during variable and extreme climate are going to hospital, cleanliness, eating healthy using herbal medicine. More than 85% of the respondents did not experience shortage of food in a year. But price of food has been increasing over the last 5 years. Finally, respondents favored adaptation strategy is saving followed by involving in other livelihood activities, government assistance, cleaning the environment, tree planting and protection of coastal areas.
