249 Increasing Structural Fiber Improves Growth Performance of Nursery Pigs

Abstract Fiber ingredients in swine diets have various components that affect the intestinal tract distinctively. The objective of this study was to determine the effect of structural fiber sources on growth performance of nursery pigs. A total of 1,760 weanling pigs (initial BW = 6.12 ± 0.3 kg, 16 reps/trt, 22 pigs/pen) were used in a study with 5 dietary treatments: 1) Complex nursery diet with no additional fiber (CON); 2) CON + 2% rice hulls; 3) CON + 4% rice hulls; 4) CON + 6% wheat middlings; 5) CON + 12% wheat middlings. Fiber ingredients were added at the expense of corn and some processed soybean meal to maintain similar dietary protein levels. Using an in vitro fermentation estimation of fermentation for rice hulls and wheat middlings, diets were constructed to add similar levels of structural (non-fermentable NDF) fiber between low and high additions of rice hulls and wheat middlings. Pens across two barns were randomly allotted independent of one another and within a location block to one of the five treatments on d 0 of a two-phase study (d 0–11 and d 11–20.5 post-weaning). Data were analyzed by general linear model in R. Mortality and removal data were analyzed as a generalized linear mixed model with a binomial distribution. Contrasts tested the effect of additional fiber (CON vs treatments 2–5), effect of medium vs high fiber (treatment 2/4 vs 3/5), effect of source of fiber (treatment 2/3 vs 4/5), and the interaction of level and source of fiber. Overall (Table 1), additional fiber resulted in increased ADFI (P < 0.05) and tended to increase ADG (P < 0.10). Probability of mortality and removal was reduced (P < 0.05) when additional fiber was included. In summary, increasing the level of structural fiber improved performance and livability of nursery pigs.

Abstract This study evaluated the progeny of sows fed a control or probiotic diet with Bacillus subtilis C-3102 (Calsporin®, Calpis Co. Ltd., Tokyo, Japan) at 500,000 and 1,000,000 CFU/g diet in gestation and lactation, respectively. A total of 358 weaned pigs (DNA 241 × 600) were used in a 42-d trial with 4–5 pigs/pen and 18–19 pens/treatment. Pens were allotted to treatments in a completely randomized design based on BW at weaning. Treatments were a 2×2 factorial with main effects of sow diet (control vs. probiotic) and nursery diet (control vs. probiotic). In the nursery probiotic diet, a product based on probiotic Bacillus subtilis C-3102 at 500,000 CFU/g diet and prebiotics β-glucans and mannan oligosaccharides was included at 0.05% (BacPack ABF™, Quality Technology International, Inc., Elgin, IL). Diets were corn-soybean meal-based. Growth performance and fecal consistency on a 1-to-5 scale were evaluated weekly. Fecal samples were collected for microbial analysis by culture method and bacterial quantification of Bacillus subtilis C-3102, total Bacillus sp., Lactobacillus sp., Clostridium perfringens, Salmonella spp., Enterococcus sp., Enterobacteriaceae, total aerobes, and total anaerobes. Data were analyzed using a linear mixed model (PROC GLIMMIX, SAS®) with pen as experimental unit. There was no evidence for effect of sow diet, nursery diet, or interactions (P > 0.10) on overall nursery growth performance and fecal consistency. However, growth performance from d 21 to 42 and final BW were greater (P < 0.05) in progeny of control-fed sows compared to probiotic-fed sows. Microbial analysis revealed an increase (P < 0.01) in Bacillus subtilis C-3102 and, consequently, total Bacillus sp. in fecal microflora of probiotic-fed pigs. In conclusion, probiotic inclusion to sow diets lowered growth performance of the progeny in late nursery. The probiotic diet provided to sows or nursery pigs did not influence fecal consistency, but altered the fecal microbial population in nursery pigs by increasing total Bacillus sp.

Abstract The objective of this experiment was to evaluate the growth performance and bone mineral content (BMC) of nursery pigs in response to increasing total calcium (Ca) to available phosphorus (aP) ratios in diets containing phytase (250 FTU/kg; Natuphos E, BASF, Florham Park, NJ). A total of 480 nursery pigs (body weight (BW) = 5.7 ± 0.6 kg) with 10 pigs per pen and 7 pens per treatment (6 pens fed 2.75:1 diet) were allotted to seven treatments consisting of increasing ratios of calcium to available phosphorus (Ca:aP): 1.25, 1.50, 1.75, 2.00, 2.25, 2.50, and 2.75. From day −7 to 0, pigs were fed a common diet. They were then fed the treatment diets during two experimental phases from day 1 to 14 and 15 to 28, respectively. Available P was formulated to 0.33% and 0.27% (approximately 90% of requirement) in dietary phases 1 and 2, respectively. BW, average daily gain (ADG), average daily feed intake (ADFI), and gain-to-feed ratio (G:F) were determined. BMC of the femur was measured on day 28 on one pig per pen using dual x-ray absorptiometry. Data were analyzed as a linear mixed model using PROC MIXED (SAS, 9.3). Orthogonal polynomial contrasts were used to determine the linear and quadratic effects of increasing the Ca:aP. Over the 28-d experimental period, increasing Ca:aP resulted in a linear decrease in ADG (353, 338, 328, 304, 317, 291, and 280 g/d; P &lt; 0.01), ADFI (539, 528, 528, 500, 533, 512, and 489 g/d; P &lt; 0.05), and G:F (0.68, 0.66, 0.64, 0.62, 0.61, 0.59, and 0.58; P &lt; 0.01). Increasing Ca:aP also resulted in decreased BW on days 14 and 28 (P &lt; 0.01). The BMC of the femur decreased with increasing Ca:aP (6.2, 6.3, 5.7, 5.9, 5.5, 5.6, and 5.3 g; P &lt; 0.05). Regression analysis explained the impact of Ca:aP as follows on ADG (ADG [g/d] = 339 − 36x; r2 = 0.81), G:F (G:F = 0.61 – 0.03x; r2 = 0.72), and BMC (BMC [g] = 6.4 – 0.27x; r2 = 0.43), where x is the Ca:aP. In conclusion, all outcomes indicated that any level of calcium above the minimum used in this experiment impaired growth performance and skeletal development. Further research using even lower levels of dietary Ca is warranted.

Abstract In theory, supplementing xylanase in corn-based swine diets should improve nutrient and energy digestibility and fiber fermentability, but its efficacy is inconsistent. The experimental objective was to investigate the impact of xylanase on energy and nutrient digestibility, digesta viscosity, and fermentation when pigs are fed a diet high in insoluble fiber (&gt;20% neutral detergent fiber; NDF) and given a 46-d dietary adaptation period. Three replicates of 20 growing gilts were blocked by initial body weight, individually housed, and assigned to 1 of 4 dietary treatments: a low-fiber control (LF) with 7.5% NDF, a 30% corn bran high-fiber control (HF; 21.9% NDF), HF+100 mg xylanase/kg [HF+XY, (Econase XT 25P; AB Vista, Marlborough, UK)] providing 16,000 birch xylan units/kg; and HF+50 mg arabinoxylan-oligosaccharide (AXOS) product/kg [HF+AX, (XOS 35A; Shandong Longlive Biotechnology, Shandong, China)] providing AXOS with 3-7 degrees of polymerization. Gilts were allowed ad libitum access to fed for 36-d. On d 36, pigs were housed in metabolism crates for a 10-d period, limit fed, and feces were collected. On d 46, pigs were euthanized and ileal, cecal, and colonic digesta were collected. Data were analyzed as a linear mixed model with block and replication as random effects, and treatment as a fixed effect. Compared with LF, HF reduced the apparent ileal digestibility (AID), apparent cecal digestibility (ACED), apparent colonic digestibility (ACOD), and apparent total tract digestibility (ATTD) of dry matter (DM), gross energy (GE), crude protein (CP), acid detergent fiber (ADF), NDF, and hemicellulose (P&lt;0.01). Relative to HF, HF+XY improved the AID of GE, CP, and NDF (P&lt;0.05), and improved the ACED, ACOD, and ATTD of DM, GE, CP, NDF, ADF, and hemicellulose (P&lt;0.05). Among treatments, pigs fed HF had increased hindgut DM disappearance (P=0.031). Relative to HF, HF+XY improved cecal disappearance of DM (162 vs. 98g; P=0.008) and NDF (44 vs. 13g; P&lt;0.01). Pigs fed xylanase had a greater proportion of acetate in cecal digesta and butyrate in colonic digesta among treatments (P&lt;0.05). Compared with LF, HF increased ileal, cecal, and colonic viscosity, but HF+XY decreased ileal viscosity compared with HF (P&lt;0.001). In conclusion, increased insoluble corn-based fiber decreases digestibility, reduces cecal fermentation, and increases digesta viscosity, but supplementing xylanase partially mitigated that effect.

Abstract This study was conducted to determine the effects of a multi-species direct-fed microbial (DFM) product based on lactic acid bacteria and Bacillus subtilis on growth performance and carcass characteristics of grow-finish pigs. A total of 1,188 pigs (PIC 359 × 1050; initially 25.8 kg BW) were used in a 121-d growth trial with 27 pigs/pen and 22 pens/treatment. Pigs were allotted to treatments based on initial BW in a randomized complete block design. Treatments included a control diet and the control diet with added DFM (BiOWiSH Technologies Inc., Cincinnati, OH) included at 0.055% of the diet at the expense of corn. Diets were based on corn, distillers dried grains with solubles, and soybean meal and fed in four dietary phases. Data were analyzed using a linear mixed model (PROC GLIMMIX, SAS®) with treatment as fixed effect, block as random effect, and pen as experimental unit. Overall (d 0 to 121), pigs fed the control diet had greater ADG (P < 0.05) and final BW (P < 0.001) compared to pigs fed the DFM diet (Table 1). There was no evidence for differences (P > 0.05) in ADFI or G:F between treatments. The difference in final BW resulted in heavier (P < 0.05) HCW in control pigs compared to DFM pigs, but no evidence for differences (P > 0.05) was observed in carcass yield, backfat, loin depth, and percentage lean between treatments. In conclusion, the inclusion of this multi-species DFM in growing-finishing diets reduced ADG in this commercial study. This response could be related to inclusion rate, feeding duration, or other factors not identified in this study, warranting further research to characterize the effects on pig performance.

Abstract The objective of this study was to conduct a meta-analysis to quantitatively summarize the growth responses of broilers fed cDDGS and the efficacy of various types of dietary enzyme supplementation. A total of 12 publications with 69 observations were included in the database. Individual observations were analyzed using a multivariable linear mixed model. The mean differences (MD) of BWG, FI, and gain efficiency (G/F) were calculated by subtracting either the enzyme response in corn-soybean meal (CSB) or CSB+cDDGS based diets to the control, and was expressed as a percentage (MD = (enzyme – control)/control ×100%). A type of exogenous enzymes (xylanase; protease; carbohydrases; cocktail = proteases + carbohydrases), and feeding phase (starter = d 0 to d 21; finisher = d 21 to d 42 or 49; overall = d 0 to d 42 or more) were included as fixed effects. Dietary enzyme inclusion showed significant improvement on BWG (3.19%, P &lt; 0.01) and G/F (5.69%, P &lt; 0.01) in broilers fed cDDGS diet. However, no significant enzyme responses were observed in broilers fed CSB diet on growth performance. Broilers fed cDDGS diet had increased (P &lt; 0.01) BWG with the addition of protease (3.32 %) and cocktail (3.27 %), whereas addition of xylanased improved (P &lt; 0.01) G/F by (3.56 %) and carbohydrases (1.90 %). Broilers fed cDDGS diet with enzyme supplementation showed greater improvement in BWG (3.71 %, P &lt; 0.01) and G/F (3.78 %, P &lt; 0.01) at finisher phase compared with starter phase. Likewise, Broilers fed CSB diet with enzyme supplementation increased BWG (9.40 %, P &lt; 0.01) and G/F (3.11 %, P &lt; 0.01) at finisher phase. In conclusion, supplementation of xylanase and carbohydrases in cDDGS diet improved G/F, and the enzyme response can be maximized when fed during the finisher phase diet compared with the starter phase diet.

In vitro maturation of goat oocytes has traditionally involved the use of serum or BSA. However, these products introduce variability and complicate evaluation of the effects of other medium components. The objective of this study was to examine the effects of citrate and hyaluronate in the absence or presence of BSA during IVM on the developmental competence of goat oocytes. Abattoir-derived, cumulus-oocyte complexes (COC) were matured for 20–22h (6.0% CO2 in air, 38.7°C) in modified SOF medium (1.5mM glucose, 3.0mM L-lactate, 0.1mM pyruvate, 1.0mM glutamine, 0.1mM taurine) supplemented with 1×MEM nonessential amino acids, 0.5×MEM essential amino acids, 1×MEM vitamins, 0.1mM cysteamine, 5μg mL−1 insulin, 5μgmL−1 transferrin, 5ng mL−1 selenium, 50ngmL−1 EGF, 0.01U mL−1 LH and FSH, and 50μgmL−1 gentamicin. Treatments were: (1) 1mgmL−1 PVA (protein-free, defined); (2) 4mgmL−1 BSA (semi-defined); (3) 0.5mM citrate and 0.5mgmL−1 hylauronate (C+H, defined); and (4) 0.5mM citrate and 0.5mgmL−1 hylauronate with 4mgmL−1 BSA (C+H+BSA, semi-defined). At the end of IVM, COC were transferred to modified Brackett and Oliphant’s medium with 7.7mM Ca-(l)-lactate and 20% FCS for IVF. Frozen-thawed sperm were processed through a 45%:90% Percoll gradient and added to IVF drops (50μL) containing COC at a final concentration of 14–15×106 spermmL−1. Gametes were coincubated in the presence of heparin (25μgmL−1) for 22–24h in 7% CO2 in air at 38.7°C. After coincubation, cumulus cells were removed and zygotes were cultured (6% CO2, 5% O2, 89% N2, 38.7°C) in G1 v.3 for 3 days followed by 4 days in G2 v.3. Cleavage was evaluated when embryos were moved to G2, and development to the blastocyst stage was assessed at the end of culture. All blastocysts were fixed and stained with Hoechst 33342 for total cell counts. Analysis of variance was performed using the general linear mixed model macro of SAS. Means are presented ±SEM and probability values P&lt;0.05 were considered significant. The use of BSA did not improve (P&gt;0.05) the developmental potential of goat oocytes (Table 1). Furthermore, a similar proportion (P&gt;0.05) of oocytes developed to the blastocyst and hatching blastocyst stage after maturation under defined conditions compared to oocytes matured with BSA. In conclusion, developmentally competent goat oocytes can be produced by IVM under defined conditions. Table 1 Development of goat oocytes following IVM with different macromolecules.

Context. The wettability of the chemically modified dentin substrate is a condition that intervenes in dentin-vascular and cellular interaction across regenerative endodontics. Aims. To compare the effect of CS and CSnp on the wettability in root dentine with other irrigation protocols with an experimental in vitro model prior regenerative endodontics. Methods and Material. An in vitro experimental study that included eighty hemisected human root distributed into 8 groups: G1- distilled water; G2- 1% NaOCl/17% EDTA; G3- hypochlorous acid 0.025% HOCl, G4- 1% NaOCl/0.025% HOCl/17% EDTA, G5- 0.2 g/100 mL CS, G6- 1% NaOCl/0.2 g/100 mL CS, G7- CSnp, and G8- 1% NaOCl/CSnp. The wettability analysis calculated the contact angle (θ) between a drop of a blood-like and root dentinal surface; topographic characterization with scanning electron microscopy (SEM) quantified the diameter and number of tubules per area; spectroscopy infrared analyses (IR-S) identified chemical changes in the inorganic (phosphate/carbonate) and organic phase (amide/methyl). Statistical analysis: a linear mixed model, Kruskal–Wallis, and Holm–Bonferroni correction ( P  < 0.05) were used. Results. Significantly higher wettability for G2 (27.1 ( P  = 0.0001)) was found. A mean value of 67°±°for experimental groups ( P  = 0.07) was found, and we did not identify differences between them. The SEM identified greater tubular opening and erosion for G4 and greater dentinal permeability per area for NaOCl/CS. IR-S identified dentinal organic integrity with NaOCl-CS/CSnp compared to organic reduction promoted for NaOCl/EDTA. Conclusions. This in vitro dentin determined an indirect association between the wettability and organic contents. The oxidative effect of NaOCl could be neutralized by CS-CSnp, and consequently, the wettability of the substrate decreases.

Semen processed with procedures permitting a flexible thaw method is used to breed millions of cows yearly. “Pocket thawing” is widely used as an alternative to warm-water thawing with such semen. To pocket thaw, a straw is retrieved from cryostorage, immediately wrapped in a folded paper towel, and moved to a thermally protected pocket for 2 to 3 min of thawing within the pocket before AI gun loading. Published field data are lacking for comparisons of such a thaw method with those for semen prepared to permit flexible-thawing. We measured the effect of warm-water or pocket thaw on conception rate in four dairy heifer herds using semen prepared with methods previously optimized for flexible-thawing success. Semen processing (Anderson S et al. 1994 J. Dairy Sci. 77, 2302–2307) includes two-step whole-milk extension, static vapor tank freezing (0.5-mL straws), and IMV Digitcool mechanical freezing (0.25-mL straws). It is unclear which specific processing steps permit flexible thawing. These procedures have been developed using breeding results from decades of field trials by professional inseminators using both pocket and warm-water thaw. Semen prepared from each of 12 sires produced equal straw units at 10 and 15 million total sperm per straw, in both 0.5- and 0.25-mL straw packages. Professional inseminators used each combination evenly over 16 months. Additional commercial semen (55% of total) from the same source was used. The thaw methods alternated weekly. Thaw effect on conception status, from 70 day non-return data for 11,215 services (67.6% conception rate), was estimated by a generalized linear mixed model. Neither thaw method nor total sperm per straw significantly affected conception rate (P = 0.658, 0.769, respectively). Bull, herd, inseminator within herd, year, season, and straw size did significantly affect conception rate (P < 0.05). No thaw method interactions with herd, sperm number, season, and straw package size were significant (P = 0.297, 0.526, 0.365, 0.723, respectively). This suggests that if semen has been prepared with procedures specific to flexible-thawing, it can be either pocket thawed or warm-water thawed within a range of herdsman or inseminator practices, season, or straw packaging choices. Even at 10 million, the lowest total sperm per straw, pocket thaw was equally as successful as warm-water thaw. We generally observe that in vitro sperm quality, as expected, is maximal for rapidly thawed straws, with slower thawing resulting in lower values. However, while it appears that conventional measures of in vitro semen quality are improved with fast thaw rates, these measures do not appear to correspond to higher in vivo fertility for semen prepared intentionally to be flexibly thawed. We conclude that, for semen prepared with procedures that permit flexible thawing, the thaw method, whether pocket or warm-water thaw, does not affect conception under commercial conditions and with routine semen handling methods. In 2012, 6.7 million women worldwide were diagnosed with cancer, including young women and children. Almost 85% of the latter survive the disease. However, the necessary cytotoxic treatment reduces their reproductive potential by damaging the stock of, basically preantral, ovarian follicles. Current fertility preservation techniques try to overcome this problem. Still, the success rate is low due to a lack of optimization and standardization. Nowadays, culturing preantral follicles in vitro is not yet routinely possible. Therefore, we used an in vivo model based on xenotransplantation of bovine ovarian cortex tissue in recipient mice, as an alternative to studying human reproduction. To further characterise this model, we assessed the influence from the host on the graft at the protein level. Visualising murine annexin A1, an anti-inflammatory protein, in the transplanted bovine cortex tissue should help to elucidate the process of immunological rejection. Hereto its distribution around the present bovine follicles is measured. In total, 12 mice (6 conventional fluorescent C57BL/6-eGFP and 6 immune deficient Balb/c-Nu) were used as graft recipients. All mice were anesthetized with an intraperitoneal injection and subsequently sterilized. Small pieces (max. 9 mm3) of adult bovine ovarian cortex, retrieved from slaughterhouse ovaries, were grafted retroperitoneal. After a transplantation period of 14 (3 mice/mice strain) or 28 (3 mice/mice strain) days, the mice were killed and all grafts were successfully recovered. These were fixed (4% formaldehyde) and processed into histological paraffin slides. Murine annexin A1 (Sigma-Aldrich HPA011271) was localised using immunofluorescence through a rhodamine label (RITC filter). We measured the shortest distances between 10 annexin fluorescent signals and the basal membrane of each follicle, found in the graft. We calculated the average of these measurements, resulting in one data point per follicle. The dataset was fitted in a linear mixed model with annexin as dependent variable, and transplantation period and mice strain as fixed effects. Fluorescent signals of murine annexin A1 were found in all grafts form both mice strains, surrounding bovine follicles. Data show no interaction between mice strain and the duration of the transplantation period. The influence of the mice strain showed a trend towards significance (P = 0.08), possibly due to the immunological state of the host. To our knowledge, this is the first time an attempt has been made to characterise the host/donor interaction in xenografting procedures. We can conclude that annexin A1 from the murine host (whether immunodeficient or immunocompetent) invades the bovine graft tissue in a short period of time.
