The fate of naturally occurring and added bacterial pathogens was determined in “soul foods” purchased at local supermarkets and farm families while the foods were stored under conditions simulating those used for retail distribution, home storage, and preparation before use. Viable count determinations for 10 samples at the end of a 5-day period at 10 C showed considerable decreases in comparison to the inoculum size, indicating that growth was not promoted. Escherichia coli survived in all the food samples but the populations decreased by 1 to 9 log cycles/g of food. Salmonella typhimurium survived in 59% of the food samples. Except for farm family collard greens and sausage (encased), Staphylococcus aureus remained viable in all of the foods tested an d was the only survivor in cracklings (cooked) obtained from both sources. Clostridium perfringens was detected in farm family sweet peas and 23% of the pig offal samples.

ABSTRACT Environmental testing for Salmonella Enteritidis is required for U.S. shell egg producers with ≥3,000 hens on a farm. The egg producer assumes all costs for the mandatory testing. According to the U.S. Food and Drug Administration (FDA) Egg Rule, either manure scraper or drag swabs can be collected according to published guidelines and requirements. The present study was undertaken to determine the efficacy of Salmonella detection with one-, two-, and four-swab pools of either manure scraper or drag swabs. Resistant isolates of Salmonella serovars Enteritidis (1,000 ppm of streptomycin), Heidelberg (200 ppm of nalidixic acid [NA]), Typhimurium (200 ppm of NA), and Kentucky (200 ppm of NA) were utilized. Low (approximately 8.4 CFU) and high (approximately 84 CFU) levels of inocula were introduced onto a single swab within a pool. Single flocks from each conventional cage (manure scraper swabs) and cage-free barn (drag swabs) were monitored throughout the study at the ages required under the FDA Egg Rule. The highest and most consistent recovery of inoculum was found in single swab samples. For low dose inocula, recovery of isolates was low from single manure scraper swabs (57.9 to 29.2%) and decreased as more swabs were added to the pool. Recovery of isolates from manure scraper swabs was higher for high dose inocula, although Salmonella Heidelberg was outcompeted by the naturally occurring flora and had the lowest rate of recovery among the isolates tested. One- and two-swab pools of drag swabs had similar rates of recovery at both low and high doses for Salmonella Enteritidis, Salmonella Heidelberg, and Salmonella Typhimurium. When Salmonella Enteritidis and Salmonella Kentucky were combined in an inoculum, Salmonella Enteritidis was recovered at a much higher rate than was Salmonella Kentucky for all types of swabs and doses of inocula. Pooling of two drag swabs allowed for similar detection of low and high dose Salmonella, but the pooling of manure scraper swabs decreased detection of low dose Salmonella. HIGHLIGHTS

Rectal stricture occurred in each of 23 pigs submitted for necropsy from eight Indiana farms. Each stricture was an annular cicatrization of the rectal wall, 2.0-5.0 cm anterior to the anorectal junction. Emaciation, colonic dilatation and compression atrophy of abdominal and thoracic viscera were a result of chronic obstipation and inanition. Most pigs with strictures had severe enteric disease 4-8 weeks before the strictures occurred. Salmonella typhimurium was isolated from seven of the eight groups of pigs examined. Ulcerative proctitis, a possible precursor of rectal stricture, was frequently in pigs with enterocolitis caused by 5. typhimurium.

SUMMARYSalmonella typhimuriumstrains of biotype 25x have been shown in transductional cross experiments to be clonal in the Nad character. The ancestral bacterium, probably of biotype 25a, mutated to a requirement for nicotinamide and subsequently diversified in phage type and secondary biotype characters. Such a sequence of events indicates interconversion among phage types 6, 16, 46, 49, 73, 76 and 135. Strains in biotypes 1x, 9ix, 17x, 17dx, 19dx and 25hix yielded Nad+recombinants in interbiotype crosses, suggesting that each originated as an independent mutant line.
