Pollen grains are male gametophytes, an ephemeral haploid generation of plants, commonly engaging in competition for a limited supply of ovules. Since differential male fertility may influence the direction and pace of population evolution, the relative fitness of pollen is regularly estimated as either pollen viability, the proportion of pollen containing intact cytoplasm's and regenerative nuclei, or pollen fertility, the frequency of pollen germinating under standardized conditions. Here, we estimated the relative fitness of pollen in a dioecious, wind-pollinated model system, Cannabis sativa, by characterizing pollen fertility and viability from multiple sires. Pollen fertility quickly declined within two weeks of anther dehiscence, and pollen stored under freezer conditions did not germinate regardless of storage time. In contrast, pollen viability declined slowly and persisted longer than the lifetime of a sporophyte plant under both room temperature and freezer conditions. Pollen samples that underwent both fertility and viability analysis displayed no significant correlation, implying researchers cannot predict pollen fertility from pollen viability, nor infer male gametophytic fitness from a single measure. Our work demonstrates two approaches to measure proxies of male fitness in C. sativa, and identifies new questions around what are valuable estimates of male fitness in plants.

Abstract. The zones of distribution of varieties and forms of quince on the territory of the Nakhchivan Autonomous Republic have been established, phenological observations have been carried out, their flowering and fruiting have been studied. On the territory of the Autonomous Republic, flowering of varieties and forms of quince begins in the second decade of April, depending on the distribution zone, with an average daily temperature of 12–13 °C and lasts 12–13 days, depending on weather conditions. Each flower has 10–12 stamens arranged in one row. The article also studied the viability of pollen in a number of quince varieties. Pollen viability was studied in the varieties Sary, Tursh, Ordubad, Gara and wild forms. Pollen fertility was determined by staining with acetocarmine. Pollen germinates in 2–5–10–15 and 20% glucose solution. Counting of germinated pollen grains was carried out under a microscope. The study showed that of all the experimental varieties, the pollen fertility of the Sary quince and Tursh quince varieties is high (up to 96.6–97.1%). The best medium for the germination of quince pollen is a 10–15% glucose solution. Pollen germination in this solution reaches 47.4–88.0%. In distilled water (control), the germination of quince pollen reached from 9.7% to 35.6% for varieties. Quince pollen remains viable for 31–43 days.

This research was conducted in 2006 in the Experimental Station of the Department of Genetics and Horticultural Plant Breeding in Felin near Lublin. It included 7 cultivars of strawberry ('Pastel', 'Salut', 'Teresa', 'Jota', 'Paula', 'Senga Sengana', 'Plena') and two selected clones: 2395 and 3995 that belonged to the Department. Analysis of pollen fertility was carried out on the basis of materials collected on five occasions: on May 17 (at the beginning of florescence), May 20 and May 24 (the peak of florescence) and on June 2 and June 13 (the end of florescence). Smear preparations stained with 2% acetocarmin and glycerin solution (1:1) were used to test pollen viability. The highest percent of viable pollen in 2006 was in cultivar 'Jota' (70.88%). The lowest viability had pollen of 'Teresa' cultivar (33.83%). The average content of viable pollen grains was the lowest on May 20 (36.79%). The highest pollen fertility was noted on June 2 (62.15%), the percent of fertile pollen on a similar level was observed on May 17 and 24 and June13. The cultivar 'Jota' was characterized by an increased level of pollen fertility that was quite high during the whole period of florescence. The number of seeds has a great influence on the proper development of spurious strawberry fruit. The mass of seeds from a single fruit has a great influence on the mass of fruits in both large and small fruit categories. Pollen viability affected the mass of seeds on big fruits (r = 0.444), but there is no clear direct relationship between pollen fertility and mass of strawberry fruit(r = -0.193 and r = -0.052).

The pollen–ovule ratios, pollen fertility rate, pollen viability and longevity, pollen histochemistry of Robinia pseudoacacia flowers were determined in the field on Zhen Mountain in Yantai, Shandong province in China. The main results were as follows: Robinia pseudoacacia is completely fertile and has starchy pollen. The pollen longevity was about 80h. The pollen viability was higher than 90% at the beginning of anthesis on sunny days. 80h later, most pollen grains lose their viability. Pollen–ovule ratio (P/O) is 192±19. Accoring to Cruden, R. pseudoacacia is facultative autogamous.

Abstract Pollen characteristics are very important for Iris interspecific hybridisation. In this study, the pollen viability and male meiosis were studied in yellow-flowered Iris dichotoma (Y2), I. domestica (S3) and their hybrids F1, F2 and BC1 (BC1-Y and BC1-S). The BC1-Y hybrids showed higher pollen viability than that of F1, F2 and BC1-S hybrids, which were between I. dichotoma (26.1%) and I. domestica (35.1%). Two sterile hybrids, F2-1 and BC1-S-1, exhibited more meiotic abnormalities (57.3% and 58.7%) than other individuals. During the first meiotic division, a diffuse diplotene stage was observed for the first time in the genus Iris. The meiotic abnormalities included non-congressed chromosomes, chromosome bridges, lagging chromosomes, unequal division, abnormally oriented spindle fibres, nonsynchronous division and polyad, and resulted in reduced pollen fertility. The relatively high frequency of 2n pollen grains was found in hybrids of BC1-Y-2, BC1-Y-1, BC1-S-2, BC1-S-3 and BC1-S-4. Our research provides a new resource for meiotic behaviour and pollen fertility of the genus Iris.

<p>There are many populations in a 16 km east-west distribution and populations are usually small with fewer than 100 plants/clumps. The pollen biology in terms of pollen production, pollen-ovule ratio estimation, pollen viability and pollen fertility was studied to determine the impact of plant and flower density on pollen viability and fruit set of <em>Calceolaria tripartita</em> Ruiz &amp;Pav. (Scrophulariaceae) in Darjeeling Himalaya. It is revealed that higher the density of the plants and flowers, the productivity of pollens and ovules and the viability and fertility of pollen grains become higher showing a positive correlation between plant-flower density vs. pollen biology. Fruit set is higher in high dense flowering site than low ones which might be due to higher pollen fertility rate, low sterility and greater number of pollen load upon stigma. Thus, it reflects that plant and flower density has a crucial role upon the reproductive fitness of this taxon.</p>

Although most flowering plants are polyploid, little is known of how the meiotic process evolves after polyploidisation to stabilise and preserve fertility. On wheat polyploidisation, the major meiotic gene ZIP4 on chromosome 3B duplicated onto 5B and diverged (TaZIP4-B2). TaZIP4-B2 was recently shown to promote homologous pairing, synapsis and crossover, and suppress homoeologous crossover. We therefore suspected that these meiotic stabilising effects could be important for preserving wheat fertility. A CRISPR Tazip4-B2 mutant was exploited to assess the contribution of the 5B duplicated ZIP4 copy in maintaining pollen viability and grain setting. Analysis demonstrated abnormalities in 56% of meiocytes in the Tazip4-B2 mutant, with micronuclei in 50% of tetrads, reduced size in 48% of pollen grains and a near 50% reduction in grain number. Further studies showed that most of the reduced grain number occurred when Tazip4-B2 mutant plants were pollinated with the less viable Tazip4-B2 mutant pollen rather than with wild type pollen, suggesting that the stabilising effect of TaZIP4-B2 on meiosis has a greater consequence in subsequent male, rather than female gametogenesis. These studies reveal the extraordinary value of the wheat chromosome 5B TaZIP4-B2 duplication to agriculture and human nutrition. Future studies should further investigate the role of TaZIP4-B2 on female fertility and assess whether different TaZIP4-B2 alleles exhibit variable effects on meiotic stabilisation and/or resistance to temperature change.

To study the flowering biology of Rhododendron pulchrum, we used scanning electron microscopy (SEM) and paraffin sectioning to observe the microstructures of its floral organs, a methyl thiazolyl tetrazolium (MTT) colorimetric assay to detect pollen viability in different periods, continuous observations to study flowering phenology, and artificial pollination and a benzidine-hydrogen peroxide method to determine stigma receptivity. R. pulchrum exhibited a centralized flowering phenology. The protogynous stigmas of R. pulchrum were able to receive pollen before flowering. The pollen grains of R. pulchrum fused into tetrads, the average ratio of the polar axis length to the equatorial axis length (P/E) was 1.05, and the pollen viability was highest in the initial flowering period, reaching 88.98%. The pollen/ovule (P/O) ratio was 266–328, and the outcrossing index (OCI) was 4; the vitality of R. pulchrum pollen remained high in the initial flowering and blooming periods. Compared with the lifespan of a single flower, pollen vitality remained high for most of the experimental period, thereby improving male fitness. The P/O ratio suggests that R. pulchrum may have a facultative outcrossing breeding system. The OCI estimation suggests that R. pulchrum is partially self-compatible, most likely requiring pollinators to complete pollination.

AbstractIn presence of rapid environmental changes, it is of particular importance to assess the adaptive potential of populations, which is mostly determined by the additive genetic variation (VA) in fitness. In this study we used Tribolium castaneum (red flour beetles) to investigate its adaptive potential in three new environmental conditions (Dry, Hot, Hot-Dry). We tested for potential constraints that might limit adaptation, including negative genetic covariance between female and male fitness. Based on VA estimates for fitness, we expected the highest relative fitness increase in the most stressful condition Hot-Dry and similar increases in single stress conditions Dry and Hot. High adaptive potential in females in Hot was reduced by a negative covariance with male fitness. We tested adaptation to the three conditions after 20 generations of experimental evolution and found that observed adaptation mainly matched our predictions. Given that body size is commonly used as a proxy for fitness, we also tested how this trait and its genetic variance (including non-additive genetic variance) were impacted by environmental stress. In both traits, variances were sex and condition dependent, but they differed in their variance composition, cross-sex and cross-environment genetic covariances, as well as in the environmental impact on VA.

The objective of this study was the reproductive characterization of Capsicum accessions as well as of interspecific hybrids, based on pollen viability. Hybrids were obtained between Capsicum species. Pollen viability was high in most accessions, indicating that meiosis is normal, resulting in viable pollen grains. The pollen viability of species C. pubescens was the lowest (27 %). The interspecific hybrids had varying degrees of pollen viability, from fertile combinations (C. chinense x C. frutescens and C. annuum x C. baccatum) to male sterile combinations. Pollen viability also varied within the hybrid combination according to accessions used in the cross. Results indicate that male sterility is one of the incompatibility barriers among Capsicum species since hybrids can be established, but may be male sterile.
