ABSTRACTWe isolated and characterized sixty-five anaerobic gut fungi (AGF, Neocallimastigomycota) strains from fecal samples of five wild (W), one zoo-housed (Z), and three domesticated (D) herbivores in the US states of Texas (TX) and Oklahoma (OK), Wales (WA), and the Indian states of Kerala (KE) and Haryana (HA). Phylogenetic assessment based on D1-D2 region of the large rRNA subunit (LSU) identified seven distinct lineages, with strains recovered from Axis Deer (W-TX) clustering within theOrpinomyces-Neocallimastix-Pecoramyces-Feramycesclade; Boer Goat-domesticated Goat strains (W-TX, D-KE) clustering within theOontomyces-Anaeromyces-Liebetanzomycesclade; and domesticated Goat and Sheep strains (D-HA) as well as Nilgiri Tahr strains (W-KE) forming two distinct clades associated with genusBuwchfawromyces. The remaining three lineages, represented by strains recovered from Mouflon-Boer Goat (W-TX), White Tailed Deer (W-OK), and Zebra-Horse (Z-OK, and D-WA), displayed no specific suprageneric affiliation. All strains displayed monocentric thalli and produced mono/uniflagellate zoospores with the exception of Axis Deer strains, which produced polyflagellate zoospores. Isolates displayed multiple interesting microscopic features including sporangia with tightly constricted necks and fine septa at the base (Axis Deer), papillated and pseudo-intercalary sporangia (White-Tailed Deer), swollen sporangiophores and zoospores with long flagella (Mouflon-Boer Goat), zoospore release through an apical pore followed by either sporangial wall collapse (Axis Deer and Boer Goat-domesticated Goat) or sporangial wall remaining intact after discharge (Zebra-Horse), multi-sporangiated thalli with branched sporangiophores (Zebra-Horse), and short sporangiophores with subsporangial swellings (Nilgiri Tahr). Internal transcribed spacer-1 region (ITS-1) sequence analysis indicated that Zebra-Horse strains are representatives of the AL1 lineage, frequently encountered in culture-independent surveys of the alimentary tract and fecal samples from hindgut fermenters. The other six lineages, five of which were isolated from wild herbivores, have not been previously encountered in such surveys. Our results significantly expand the genus level diversity within the Neocallimastigomycota, and strongly suggest that wild herbivores represent a yet-untapped reservoir of AGF diversity. We propose the creation of seven novel genera and eight novel Neocallimastigomycota species to accommodate these strains, for which we propose the namesAgriosomyces longus(Mouflon and wild Boer Goat),Aklioshbomyces papillarum(White tailed Deer),Capellomyces foraminis(wild Boar Goat) andC. elongatus(domesticated Goat),Ghazallomyces constrictus(Axis Deer),Joblinomyces apicalis(domesticated Goat and Sheep),Khoyollomyces ramosus(Zebra-Horse), andTahromyces munnarensis(Nilgiri Tahr). The type species are strains Axs-31, WT-2, MS-4, BGB-11, GFKJa1916, GFH683, ZS-33, and TDFKJa193, respectively.

Abstract Rotavirus C (RVC) has been detected increasingly in humans and swine in different countries, including the US. It is associated with significant economic losses due to diarrheal disease in nursing piglets. In this study we aimed: (1) to determine the prevalence of RVC in healthy and diarrheic suckling piglets on US farms; and (2) to evaluate if maternal antibody (Ab) levels were associated with protection of newborn suckling piglets against RVC. There was a significantly higher prevalence (p = 0.0002) of litters with diarrhea born to gilts compared with those born to multiparous sows. Of 113 nursing piglet fecal samples tested, 76.1% were RVC RNA positive. Fecal RVC RNA was detected in significantly (p = 0.0419) higher quantities and more frequently in piglets with diarrhea compared with healthy ones (82.5 vs. 69.9%). With the exception of the historic strain Cowden (G1 genotype), field RVC strains do not replicate in cell culture, which is a major impediment for studying RVC pathogenesis and immunity. To circumvent this, we generated RVC virus-like particles (VLPs) for Cowden (G1), RV0104 (G3) and RV0143 (G6) and used them as antigens in ELISA to detect swine RVC Abs in serum and milk from the sows. Using RVC-VLP Ab ELISA we demonstrated that sows with diarrheic litters had significantly lower RVC IgA and IgG Ab titers in milk compared to those with healthy litters. Thus, our data suggest that insufficient lactogenic protection provided by gilts plays a key role in the development of and the increased prevalence of clinical RVC disease.

Analyses using culture-independent molecular techniques have improved our understanding of microbial composition. The aim of this work was to identify and quantify enterococci in fecal samples of wild marine species using real-time quantitative PCR. Seven Enterococcus species were examined in fecal DNA of South American fur seals (Arctocephalus australis), Subantarctic fur seals (Arctocephalus tropicalis), green turtles (Chelonia mydas), Magellanic penguins (Spheniscus magellanicus), snowy-crowned tern (Sterna trudeaui), white-backed stilt (Himantopus melanurus), white-chinned petrels (Procellaria aequinoctialis), red knot (Calidris canutus), and black-browed albatross (Thalassarche melanophris). All Enterococcus species evaluated were detected in all fecal samples of wild marine species, with a concentration ranging between 106 and 1012 copies/ng of total DNA. Differences in the enterococci distribution were observed. Enterococcus faecalis and Enterococcus mundtii were most abundant in marine mammals. Enterococcus faecalis was frequent in green turtle, Magellanic penguin, snowy-crowned tern, red knot, and black-browed albatross. Enterococcus hirae and Enterococcus gallinarum showed elevated occurrence in white-backed stilt, and Enterococcus faecium in white-chinned petrel. This study showed highest diversity of enterococci in feces of wild marine species than currently available data, and reinforced the use of culture-independent analysis to help us to enhance our understanding of enterococci in gastrointestinal tracts of wild marine species.

ABSTRACT Campylobacter species are important organisms in both human and animal health. The identification of Campylobacter currently requires the growth of organisms from complex samples and biochemical identification. In many cases, the condition of the sample being tested and/or the fastidious nature of many Campylobacter species has limited the detection of campylobacters in a laboratory setting. To address this, we have designed a set of real-time quantitative PCR (qPCR) assays to detect and quantify 14 Campylobacter species, C. coli, C. concisus, C. curvus, C. fetus, C. gracilis, C. helveticus, C. hyointestinalis, C. jejuni, C. lari, C. mucosalis, C. rectus, C. showae, C. sputorum, and C. upsaliensis, directly from DNA extracted from feces. By use of a region of the cpn60 (also known as hsp60 or groEL) gene, which encodes the universally conserved 60-kDa chaperonin, species-specific assays were designed and validated. These assays were then employed to determine the prevalence of Campylobacter species in fecal samples from dogs. Fecal samples were found to contain detectable and quantifiable levels of C. fetus, C. gracilis, C. helveticus, C. jejuni, C. showae, and C. upsaliensis, with the majority of samples containing multiple Campylobacter species. This study represents the first report of C. fetus, C. gracilis, C. mucosalis, and C. showae detection in dogs and implicates dogs as a reservoir for these species. The qPCR assays described offer investigators a new tool to study many Campylobacter species in a culture-independent manner.

Leachates from different soils extracted at similar moisture potentials varied in their ability to induce sporangium formation, zoospore release and sporangium breakdown in Phytophthora spp. Sporangium production by P. cinnamomi did not occur in soil leachates heated at 40–50°C for 10 min or in soils treated with steam-air at 60° for 30 min. No evidence was obtained that Pseudomonas or Chromobacterium spp. were involved in the induction of sporangium formation. Chemotaxis of bacteria to sporangia occurred at zoospore release. Where sporangial breakdown occurred, the chemotaxis persisted for a longer period as the cytoplasm was withdrawn from the sporangial wall to be released eventually, without zoospore formation, through the papilla. The bacteria were oriented at right angles to the sporangial wall. Electron micrographs showed that in the vicinity of a bacterium, the outer, thin, electron-dense layer of the sporangial wall had disappeared and the cytoplasm of the sporangium had withdrawn from the cell wall.

The gastrointestinal tract (GIT) microbiota has been identified as an important reservoir of antibiotic resistance genes (ARGs) that can be horizontally transferred to pathogenic species. Maternal GIT microbes can be transmitted to the offspring, and recent work indicates that such transfer starts before birth. We have used culture-independent genetic screenings to explore whether ARGs are already present in the meconium accumulated in the GIT during fetal life and in feces of 1-week-old infants. We have analyzed resistance to β-lactam antibiotics (BLr) and tetracycline (Tcr), screening for a variety of genes conferring each. To evaluate whether ARGs could have been inherited by maternal transmission, we have screened perinatal fecal samples of the 1-week-old babies’ mothers, as well as a mother–infant series including meconium, fecal samples collected through the infant’s 1st year, maternal fecal samples and colostrum. Our results reveal a high prevalence of BLr and Tcr in both meconium and early fecal samples, implying that the GIT resistance reservoir starts to accumulate even before birth. We show that ARGs present in the mother may reach the meconium and colostrum and establish in the infant GIT, but also that some ARGs were likely acquired from other sources. Alarmingly, we identified in both meconium and 1-week-olds’ samples a particularly elevated prevalence of mecA (>45%), six-fold higher than that detected in the mothers. The mecA gene confers BLr to methicillin-resistant Staphylococcus aureus, and although its detection does not imply the presence of this pathogen, it does implicate the young infant’s GIT as a noteworthy reservoir of this gene.

Chromophobe pituitary adenomas arise from adenohypophysial cells and fail to exhibit cytoplasmic staining with conventional acid or basic dyes by light microscopy. The aim of the present work was to study the electron microscopic features of these tumors, to separate them into distinct entities and to correlate their fine structural appearances with secretory activity.Among 48 surgically removed various pituitary adenomas 30 tumors were found which, based on the tinctorial characteristics of the cytoplasm, corresponded to chromophobe adenomas. For electron microscopic investigation pieces of these tumors were fixed in 2.5 per cent glutaraldehyde in Sorensen's buffer, post fixed in 1 per cent osmium tetroxide in Millonig's buffer, dehydrated in graded ethanol and embedded in Epon 812. Ultrathin sections were stained with uranyl acetate and lead citrate.By electron microscopy it was possible to separate chromophobe adenomas into 3 distinct entities: 1) adenomas consisting of sparsely granulated growth hormone cells (7 cases).

On September 15, 2003, the US. Court of Appeals for the Eleventh Circuit held that agreements between pharmaceutical and generic companies not to compete are not per se unlawful if these agreements do not expand the existing exclusionary right of a patent. The Valley DrugCo.v.Geneva Pharmaceuticals decision emphasizes that the nature of a patent gives the patent holder exclusive rights, and if an agreement merely confirms that exclusivity, then it is not per se unlawful. With this holding, the appeals court reversed the decision of the trial court, which held that agreements under which competitors are paid to stay out of the market are per se violations of the antitrust laws. An examination of the Valley Drugtrial and appeals court decisions sheds light on the two sides of an emerging legal debate concerning the validity of pay-not-to-compete agreements, and more broadly, on the appropriate balance between the seemingly competing interests of patent and antitrust laws.

A variant of ductal carcinoma of the human breast which has been designated apocrine carcinoma has distinctive light and electron microscopic features. Such tumors comprise approximately 0.5% of breast carcinomas. Abundant cytoplasmic membrane bound vesicles (400-600 nm) with dense homogeneous osmophilic cores characterize these tumors. These granules are also seen in apocrine metaplastic breast epithelial lesions1 and appear to be responsible for the finely granular eosinophilic cytoplasm observed by light microscopy. A high content of intermediate affinity non-saturable 4S progesteroneestrogen binding protein (PEBP) in apocrine carcinoma has been reported.2 The present ultrastructural study evaluates the presence of apocrine granules in infiltrating ductal carcinoma (NOS) to determine if a correlation exists between apocrine granule content and the quantity of PEBP present.
