Shanghai Cooperation Organization (SCO) is presently a Eurasian politico-economic bloc having two South Asian members, Pakistan and India. The aim of the study was to evaluate the prospective role of Pakistan in the SCO with qualitative study based on interviews. On June 2017, Pakistan was granted permanent membership in the SCO during its 16th Heads of State Summit in Astana. By associating itself as a permanent member, summit allows Pakistan to contribute to regional development alongside other key regional players, Russia and China and India. The study gave four findings that how multilateralism was having a calming effect on a regional conflict, Pakistan embracing independence from isolation through Trans-Afghanistan gas pipeline. Through CPEC, Pakistan was offering the SCO member countries an economic hallway. And how the Eurasian politico-economic ambitions to reach the warm waters of the Arabian Sea was perceived as hampering the Western economic interests, thereby offering a major impediment to Pakistan’s emerging role in the SCO. Moreover, SCO participation will help enhance Pakistan's worldwide socio-economic objectives. This study examines the socio-politico and economic aspirations of Pakistan and benefit due to ties with SCO member countries.

Biodiesel has lately gained popularity due to its environmental issues and the fact that it is generated from renewable resources. However, the cost of synthesis of biodiesel is the major impediment toward commercialization. The utilization of leftover cooking oils as raw material, the adaptation of a continuous transesterification process, and the use of cheap catalysts are the major possibilities for investigation the cost of biodiesel. In this work, a dolomite catalyst was prepared from natural dolomite rocks and used for the evaluation of continuous transesterification of biodiesel from waste cooking oil (WCO). The dolomite catalyst was prepared by activation under vacuum at a surface area of 34.5 m2/g. The characterization tests showed good thermal stability of the catalyst and evolution of the CaO and MgO compounds at high concentrations. A kinetic study was conducted to obtain kinetic parameters of catalytic transesterification of the WCO.

T cell exhaustion is a major impediment to anti-tumor immunity. However, it remains elusive how other immune cells in the tumor microenvironment (TME) contribute to this dysfunctional state. Here we show that the biology of tumor-associated macrophages (TAM) and exhausted T cells (Tex) in the TME is extensively linked. We demonstrate that in vivo depletion of TAM reduces exhaustion programs in tumor-infiltrating CD8+ T cells and reinvigorates their effector potential. Reciprocally, transcriptional and epigenetic profiling reveals that Tex express factors that actively recruit monocytes to the TME and shape their differentiation. Using lattice light sheet microscopy, we show that TAM and CD8+ T cells engage in unique long-lasting antigen-specific synaptic interactions that fail to activate T cells, but prime them for exhaustion, which is then accelerated in hypoxic conditions. Spatially resolved sequencing supports a spatiotemporal self-enforcing positive feedback circuit that is aligned to protect rather than destroy a tumor.

It has usually been accepted that the success of any organisation or nation depends on the ability of its leader to govern and manage its affairs. The failure of most developing countries, especially Nigeria, has often been blamed on bad leadership. Most Nigerians are of the opinion that the leadership problem in Nigeria has been the major impediment in the nation’s development. Most people also agree that corruption and bad governance, which are prevalent in Nigeria today, are the results of leadership failure. The consensus among scholars, critics and observers of the Nigerian state is that the current situation of the nation is the result of lack of good and purposeful leadership. It is because of leadership failure in the country that Nigerians today clamor for good and purposeful leadership. This paper is a contribution to the search for the long-awaited leader. This paper examines Barnabas’ character using the narrative method.Contribution: This article reveals that Barnabas was a bridge-builder, team player, non-discriminatory and not power or position drunk and these are the traits that should be used as a model in selecting or electing leaders within ecclesiastical and secular circles.

AbstractInfection is a major co-morbidity that contributes to impaired healing in diabetic wounds. Although impairments in diabetic neutrophils have been blamed for this co-morbidity, what causes these impairments and whether they can be overcome, remain largely unclear. Diabetic neutrophils, extracted from diabetic individuals, exhibit chemotaxis impairment but this peculiar functional impairment has been largely ignored because it appears to contradict the clinical findings which blame excessive neutrophil influx (neutrophilia) as a major impediment to healing in chronic diabetic ulcers. Here, we report that exposure to glucose in diabetic range results in impaired chemotaxis signaling through the FPR1 chemokine receptor in neutrophils, culminating in reduced chemotaxis and delayed neutrophil trafficking in wound in diabetic animals, and rendering diabetic wound vulnerable to infection. We further show that at least some auxiliary chemokine receptors remain functional under diabetic conditions and their engagement by the pro-inflammatory cytokine CCL3, overrides the requirement for FPR1 signaling and substantially improves infection control by jumpstarting the neutrophil response toward infection, and stimulates healing in diabetic wound. We posit that CCL3 may have real therapeutic potential for the treatment of diabetic foot ulcers if it is applied topically after the surgical debridement process which is intended to reset chronic ulcers into acute fresh wounds.

The modern outlook on emancipation has made its quest inseparable from a quest for endless enhancement, based on an ever-more intensive exploitation of the biophysical world. This accounts for how unsustainable ways of living are reiterated worldwide, in spite of evidence of their deleterious effects. The underpinnings of unsustainability, and a major impediment to conceiving alternatives, come from an account of the human as ontologically indeterminate, crushed on doing, both vulnerable and powerful towards the world. The impasse of such ambivalence hampers social theory critique, from post-humanist ontologies to the case for degrowth and lifestyle politics. The article outlines a different take on emancipation. An account is provided of form-of-life as a doing tailored to being – not as a self-enclosed monad but as a result of the encounter between own ‘inclination’ and the world. This theoretical perspective discloses a research program on emergent mobilisations.

Various types of character education theories have swiftly developed, such as Thomas Lickona's notion of moral knowing, moral emotion, and moral deeds. The Presidential Regulation No. 87 of 2017 on Strengthening of Character Education has also spawned a slew of derivative theories. Some of the new results in character education are holistic-integrative and character-interconnected. However, character education, particularly Islamic education, continues to be a source of concern for all parties. The challenges of Islamic education are increasing with the sophistication of technology. The community's faltering use of technology is a major impediment to increasing Islamic education's principles. This study's research technique is descriptive-qualitative, utilizing literature reviews from a variety of scientific data sources. According to the findings and conclusions of this study, Islamic education in Indonesia plays both a function and a challenge in the age of disruption. Problems in the sphere of educational technology, the dichotomy of science, educational evaluation, subject matter delivery, and geographical challenges of Islamic education are among the challenges in Islamic education. Thus, to create a golden generation of Indonesia in 2045, Islamic education plays an important role in maintaining Islamic values, character values and noble character.

Acquisition of resistance to cisplatin is a major impediment to the success of cisplatin-based combination therapies for cancer. Recent studies indicate that exosomal miRNAs derived from drug-resistant tumour cells can confer resistance properties to recipient cells by a horizontal transfer mechanism. Although the role of horizontal transfer of a few miRNAs has been described, little is known about the concerted action of horizontal transfer of miRNAs in conferring cisplatin resistance. The present study was designed to identify the role of miR-643, which is one of the most significantly increased miRNA in exosomes released from cisplatin-resistant Heptocarcinoma cells, in altering the cisplatin resistance properties of recipient cells. Drug-sensitivity assays involving miR-643 revealed that ectopic expression of miR-643 can desensitise the cells towards cisplatin. Furthermore, we identified APOL6 as a major target of miR-643. Further mechanistic studies showed that miR-643 can modulate APOL6 mRNA and protein levels, leading to a reversal of APOL6-mediated apoptosis. Altogether, our results suggest an APOL6-dependent mechanism for miR-643 mediated cisplatin resistance upon the horizontal transfer across cell types.

A dense reconstruction of neuronal synaptic connectivity typically requires high-resolution 3D electron microscopy (EM) data, but EM data alone lacks functional information about neurons and synapses. One approach to augment structural EM datasets is with the fluorescent immunohistochemical (IHC) localization of functionally relevant proteins. We describe a protocol that obviates the requirement of tissue permeabilization in thick tissue sections, a major impediment for correlative pre-embedding IHC and EM. We demonstrate the permeabilization-free labeling of neuronal cell types, intracellular enzymes, and synaptic proteins in tissue sections hundreds of microns thick in multiple brain regions from mice while simultaneously retaining the ultrastructural integrity of the tissue. Finally, we explore the utility of this protocol by performing proof-of-principle correlative experiments combining two-photon imaging of protein distributions and 3D EM.
