In recent years, great strides have been made on the cultivation of spirochetes, and if relatively simple and completely reliable methods of growing them in vitro on both solid and liquid nutrient media have been developed for a number of pathogenic and non-pathogenic spirochetes, then on the issue of With the cultivation of the pale spirochete, we are moving forward very slowly, and the successes achieved recently in this area must be recognized as very modest.

Eight strains of Ceratocystis ulmi originating from different locations and host species were found to be highly sensitive to the antibiotic myxin in in vitro tests. By paper disc assay, amounts as low as 0.5–1.0 μg caused strong inhibition of the fungus on solid media. The minimum inhibitory concentration in liquid culture was 0.2 μg/ml and levels of antibiotic above this concentration proved to be fungicidal.

ABSTRACT A novel nucleoside phosphorylation process using the food additive pyrophosphate as the phosphate source was investigated. TheMorganella morganii gene encoding a selective nucleoside pyrophosphate phosphotransferase was cloned. It was identical to theM. morganii PhoC acid phosphatase gene. Sequential in vitro random mutagenesis was performed on the gene by error-prone PCR to construct a mutant library. The mutant library was introduced intoEscherichia coli, and the transformants were screened for the production of 5′-IMP. One mutated acid phosphatase with an increased phosphotransferase reaction yield was obtained. With E. coli overproducing the mutated acid phosphatase, 101 g of 5′-IMP per liter (192 mM) was synthesized from inosine in an 88% molar yield. This improvement was achieved with two mutations, Gly to Asp at position 92 and Ile to Thr at position 171. A decreasedKm value for inosine was responsible for the increased productivity.

Bone marrow cells from three preleukemic patients with prominent marrow karyotypic abnormalities were studied in liquid culture to determine if the neoplastic clones were capable of maturation. Parallel cytogenetic and cytologic studies were performed in sequentially harvested bone marrow cultures. Maturation, albeit delayed, occurred in cultures from all three patients. By 14 days of culture in vitro, morphologic, cytochemical, and functional evidence of maturation was observed in about 70% of the cells. By day 21, 85% of the cells were mature by these criteria. All but 2 of 249 metaphases from the cultured cells contained the cytogenetic abnormality of the neoplastic clone. We conclude that some preleukemic cells identified by a chromosomal abnormality can mature in vitro. Preleukemia may be viewed as a syndrome of “early leukemia” in which the neoplastic clone is established and manifested functionally as ineffective hematopoiesis. Hematopoietic cell differentiation becomes progressively abnormal with termination in the nearly complete maturational block characteristic of acute myelogenous leukemia.

There is a vast variety of microbes available in soil performing diverse functions for plants including root colonization for protection against pathogens and nutrient acquisition. In the present study, efficacy of rhizospheric microbial strains and plant extracts were evaluated against citrus nematode for their nematicidal activity. Three different concentrations of plant extracts were evaluated in vitro, results depicts that 10% solution of Tagetes erecta (root) has given 46% the highest percentage mortality at 48hrs followed by Tagetes erecta (leaf) and Datura stramonium killed 41% and 37% nematodes respectively, similarly for 20% of plant extract in distilled water, the highest mortality rate was recorded for T. erecta (root) 68% followed by D. stramonium 47% and Acacia nilotica 44% at 48hrs of application. While using a higher concentration 30% of SS. the percentage mortality elevated multifold i.e.  Marigold root has killed maximum 76% nematodes followed by Marigold leaves and Dhatura with mortality rate 69% and 58% respectively at 48 hrs of interval. While Eucalyptus killed lowest no of nematodes. Additionally, five bacterial isolates were evaluated for their nematicidal activity. Results indicated that for bacterial cell suspension, Pseudomonas putida has shown maximum mortality rate at 84% followed by Bacillus subtilis 73% & Pseudomonas geniculata 65% at 48hrs of interval. Similarly, in the 2nd set of experiment bacterial culture filtrates has exhibited promising results, maximum mortality rate was recorded for Bacillus spp. 81% followed by B. subtilis 76%, Ps putida 63%, after 48hrs of incubation. Moreover, an insight investigation of bacterial extracellular metabolites and plant extracts would enable researchers to develop purposeful products and compositions

Abstract 1. Phosphate exchange in red cells and plasma was studied in vitro using P32 in the form of sodium phosphate as a tracer. 2. No phosphate was added other than the isotopic preparation which was of high specific activity. 3. Inorganic phosphate exchanged freely between the plasma and the erythrocytes at 37.5 C. in a period of four hours. Minimal transfer occurred at 7 C. 4. Most of the added P32 which passed into the erythrocytes during this time remained in the inorganic fraction, less than 15 per cent being found in the organic acid soluble fraction. 5. The specific activity of the inorganic phosphate of the erythrocytes was equal to or greater than that obtaining for the inorganic phosphate of the plasma at the end of the four hour incubation period at 37.5 C.
