ABSTRACT: Eggshell quality is directly linked to the good acceptability of the product by consumers, whereas the industry is concerned with economic losses. Prolonged periods of laying, diseases, nutrition, bird age and genetics can influence egg quality. This study aimed to examine the influence of the environment, bird age and storage period on the physical characteristics of the shell of eggs from commercial laying hens. The experiment involved 288 eggs from Hy-line hens, which were distributed in a completely randomized design with a triple 3×2×4 factorial arrangement represented by three ages (31, 62 or 88 weeks), two storage temperatures (ambient or refrigerated), and four storage periods (one, 10, 20 or 30 days), totaling 24 treatments. Four replications were used with three eggs each. Eggshell strength, percentage and thickness were measured. Differences were observed, with the eggs stored at room temperature exhibiting lower shell strength. Laying eggs at an advanced age (88 weeks) resulted in decreased eggshell strength as compared with the other ages, whereas the eggs from young layers (31 weeks) had a thicker shell. The advancing age of the laying hens reduced eggshell quality, regardless of the storage environment. Nonetheless, the eggs stored in the refrigerator showed better eggshell quality and integrity than those stored at ambient temperature.

Recently developed myostatin (MSTN) mutant quail and chickens demonstrated similar effects of MSTN on muscle and fat developments between avian and mammalian species. However, the effect of MSTN mutation on the quality of eggshells, an important avian specific characteristic, has not yet been investigated although egg production traits of mutant quail have been studied. In this study, several parameters for eggshell quality, including eggshell size, eggshell weight, eggshell breaking strength (EBS), and eggshell thickness, were all compared between MSTN mutant and wild-type (WT) eggs. MSTN mutant eggs had greater height and width along with heavier eggshell weight compared to WT eggs, which shows proportional improvement in egg size as affected by the MSTN mutation. However, EBS and eggshell thickness were decreased in mutant eggs compared to WT eggs. In addition, the palisade layer, the thickest and most important layer for the strength of an eggshell, was also decreased without a change in the number of vesicular holes. These data indicated that decreases in the thickness of the eggshell and the palisade layer would be a main factor contributing to a lower EBS in mutant eggs. MSTN mutant quail provide a useful model to better understand the function of MSTN on avian uterine cell development and eggshell biomineralization.

Abstract Background Eggshell is a bio-ceramic material comprising columnar calcite (CaCO3) crystals and organic proteinaceous matrix. The size, shape and orientation of the CaCO3 crystals influence the microstructural properties of chicken eggshells. However, the genetic architecture underlying eggshell crystal polymorphism remains to be elucidated. Results The integral intensity of the nine major diffraction peaks, total integral intensity and degree of orientation of the crystals were measured followed by a genome-wide association study in 839 F2 hens. The results showed that the total integral intensity was positively correlated with the eggshell strength, eggshell thickness, eggshell weight, mammillary layer thickness and effective layer thickness. The SNP-based heritabilities of total integral intensity and degree of orientation were 0.23 and 0.06, respectively. The 621 SNPs located in the range from 55.6 to 69.1 Mb in GGA1 were significantly associated with TA. PLCZ1, ABCC9, ITPR2, KCNJ8, CACNA1C and IAPP, which are involved in the biological process of regulating cytosolic calcium ion concentration, can be suggested as key genes regulating the total integral intensity. Conclusions The findings greatly advance the understanding of the genetic basis underlying the crystal ultrastructure of eggshell quality and thus will have practical significance in breeding programs for improving eggshell quality.
