There is a vast variety of microbes available in soil performing diverse functions for plants including root colonization for protection against pathogens and nutrient acquisition. In the present study, efficacy of rhizospheric microbial strains and plant extracts were evaluated against citrus nematode for their nematicidal activity. Three different concentrations of plant extracts were evaluated in vitro, results depicts that 10% solution of Tagetes erecta (root) has given 46% the highest percentage mortality at 48hrs followed by Tagetes erecta (leaf) and Datura stramonium killed 41% and 37% nematodes respectively, similarly for 20% of plant extract in distilled water, the highest mortality rate was recorded for T. erecta (root) 68% followed by D. stramonium 47% and Acacia nilotica 44% at 48hrs of application. While using a higher concentration 30% of SS. the percentage mortality elevated multifold i.e.  Marigold root has killed maximum 76% nematodes followed by Marigold leaves and Dhatura with mortality rate 69% and 58% respectively at 48 hrs of interval. While Eucalyptus killed lowest no of nematodes. Additionally, five bacterial isolates were evaluated for their nematicidal activity. Results indicated that for bacterial cell suspension, Pseudomonas putida has shown maximum mortality rate at 84% followed by Bacillus subtilis 73% & Pseudomonas geniculata 65% at 48hrs of interval. Similarly, in the 2nd set of experiment bacterial culture filtrates has exhibited promising results, maximum mortality rate was recorded for Bacillus spp. 81% followed by B. subtilis 76%, Ps putida 63%, after 48hrs of incubation. Moreover, an insight investigation of bacterial extracellular metabolites and plant extracts would enable researchers to develop purposeful products and compositions

To investigate nematicidal activity against the root-lesion nematode, Pratylenchus coffeae, the methanolic extracts were obtained from 5 medicinal plants in Vietnam. Methanol extracts of the 5 samples were screened for nematicidal activity against Pratylenchus coffeae in vitro. Of the plant extracts tested, a 5 mg/ml concentration of Terminalia nigrovenulosa extract showed the highest level (95.0%) of nematicidal activity against Pratylenchus coffeae at 9 h after treatment. The remainder of the plant extracts was followed by Cinnamomum camphora (66.7%), Jasminum suptriplinerve (30.8%), Premna integrifolia (6.7%), and Treptocaulon juventas (5.8%). A 2.0 mg/mlconcentration of T. nigrovenulosa resulted in 13.3, 42.5, 62.5 and 86.7% at 1, 3, 5 and 7 h after treatment, respectively against Pratylenchus coffeae. Concentration of 0.1, 0.5, 1.0 and 2.0 mg/ml of T. nigrovenulosa resulted in 26.7, 71.7, 78.3 and 86.7% effectiveness respectively, at 7 h after treatment. Also, at 3 days after exposure to T. nigrovenulosa, hatch was inhibited by 60.0, 45.8, 17.5 and 5.8% at 0.1, 0.5, 1.0 and 2.0 mg/ml, respectively. Hatch was inhibited by 65.0, 50.8, 21.7 and 6.7% at 0.1, 0.5, 1.0 and 2.0 mg/ml respectively, at 6 days after exposure to T. nigrovenulosa. The mortality rate and hatch inhibition increased with increasing incubation time for crude extract of T. nigrovenulosa. Trong nghiên cứu này, 5 cây thuốc (Cinnamomum camphora, Jasminum suptriplinerve, Premna integrifolia, Terminalia nigrovenulosa, Treptocaulon juventas) đã được sử dụng làm nguyên liệu chiết xuất bằng methanol để thử hoạt tính kháng tuyến trùng Pratylenchus coffeae. Kết quả cho thấy chiết xuất từ cây Terminalia nigrovenulosa biểu hiện hoạt tính kháng tuyến trùng cao nhất (95,0%) sau 9 giờ xử lý với nồng độ 5 mg/ml. Hoạt tính kháng tuyến trùng của các chất chiết xuất được biểu hiện theo thứ tự giảm dần lần lượt là C. camphora (66,7%), J. suptriplinerve (30,8%), P. integrifolia (6,7%) và T. juventas (5,8%). Với nồng độ 2 mg/ml của chất chiết xuất từ T. nigrovenulossa biểu hiện hoạt tính gây chết tuyến trùng Pratylenchuss coffeae là 13,3; 42,5; 62,5 và 86,7% sau 1, 3, 5 và 7 giờ xử lý theo thứ tự. Ở cùng thời điểm sau xử lý 7 giờ, các nồng độ 0,1; 0,5; 1,0 và 2,0 mg/ml chiết xuất của T. nigrovenulossa có hoạt tính gây chết tuyến trùng Pratylenchus coffeae lần lượt là 26,7; 71,7; 78,3 và 86,7%. Bên cạnh đó, chất chiết xuất của T. nigrovenulossa cũng cho thấy tỷ lệ ức chế trứng nở cao. Cụ thể là sau 3 ngày xử lý với chất chiết xuất của T. nigrovenulossa, tỷ lệ ức chế trứng không nở là 60,0; 45,8; 17,5 và 5,8% at 0,1; 0,5; 1,0 và 2,0 mg/ml theo thứ tự. Sau 6 ngày xử lý với chất chiết xuất của T. nigrovenulossa, tỷ lệ ức chế trứng nở là 65,0; 50,8; 21,7 và 6,7% tại nồng độ 0,1; 0,5; 1,0 và 2,0 mg/ml theo thứ tự. Tỷ lệ gây chết và tỷ lệ ức chế của chất chiết xuất phụ thuộc vào cả thời gian và nồng độ xử lý.

The antifungal activity of aqueous extracts of nine plants viz, Azadirachta indica, Parthenium hysterophorus, Momordica charantia, Allium sativum, Eucalyptus globules, Calotropis procera, Aloe vera, Beta vulgaris and Datura stramonium were assessed in vitro against Fusarium oxysporum f. sp. melongenae, Rhizoctonia solani and Macrophomina phaseolina, the soil borne phytopathogens. The assessment of fungitoxic effect was carried out by using three different concentrations i.e., 5, 10 and 20% against the test fungi, in terms of percentage of mycelial growth inhibition. The extract of A. sativum completely inhibited the mycelial growth of M. phaseolina at all the concentrations. The extracts of D. stramonium and E. globulus inhibited the mycelial growth of R. solani of 72%, and 70.7% respectively at 20% concentration, that of A. sativum, E. globulus and D. stramonium exhibited inhibition percentage of 63.3%, 61.8% and 61.1% respectively at 20% concentration on Fusarium oxysporum f. sp. melongenae. The application of plant extracts for disease management could be less expensive, easily available, non-polluting and eco-friendly.

Sarocladium oryzae (Sawada) W. Gams & D. Hawksworth, the causal agent of sheath rot of rice was isolated from sheath rot infected rice sheaths and grains. Among ten tested fungicides Tall 25 EC completely inhibited radial growth of the fungus even at the lowest concentration 100 ppm. Similarly ethanol leaf extract of ten plants, namely Allium sativum, Artocarpus heterophyllus, Asparagus racemosus, Azadirachta indica, Citrus medica, Datura metel, Mangifera indica, Nerium indicum, Senna alata and Tagetes erecta at 5, 10 and 20% concentrations were screened for their fungicidal activity against the test fungus. All the plant extracts completely inhibited the radial growth of the test fungus at 20% concentration except Asparagus racemosus and C. medica. Ethanol extract of Tagetes erecta and Mangifera indica also completely inhibited the radial growth of the test fungus at 10% concentration.Bangladesh J. Sci. Res. 29(1): 47-54, June-2016

O objetivo do trabalho foi avaliar o potencial de rizobactérias e Trichoderma spp. no controle biológico de Bursaphelenchus cocophilus. Foram avaliados 19 isolados de Pseudomonas spp. (P), 29 de Bacillus spp. (B) e 27 de Trichoderma spp. (T), obtidos de plantio comercial. Os isolados foram caracterizados quanto a produção de compostos bioquímicos. Foi avaliada a taxa de mortalidade de B. cocophilus in vitro e in vivo, neste último realizou-se também a quantificação das enzimas relacionadas à patogênese. Os isolados B14 e P41 proporcionaram 69% e 56% de taxa de mortalidade de nematoides, respectivamente e foram tanto proteolíticos como solubilizadores de fosfato. Os isolados T41 e T54 apresentaram 96% de mortalidade de B. cocophilus in vitro e foram produtores de quitinases, protease e sideróforos. No experimento in vivo, os isolados de rizobactérias B14, B23 e P23 promoveram maior taxa de mortalidade do B. cocophilus do que os demais tratamentos. As mudas de coqueiro inoculadas com o B. cocophilus apresentaram aumento nas atividades das enzimas peroxidases, quitinase e β-1,3-glucanases, quando comparadas com a testemunha (mudas não inoculadas). Os isolados B41, P14, T41 e T54 apresentaram-se como potenciais agentes de controle biológico do B. cocophilus.Palavras-chave: anel vermelho, antagonismo, nematoid. BIOLOGIC CONTROL OF Bursaphelenchus cocophilus WITH RHIZOBACTERIA AND Trichoderma isolates  ABSTRACT:The purpose of this study was to evaluate the potential of rhizobacteria Pseudomonas spp. and Bacillus spp. and Trichoderma spp. isolates on the biological control of the nematode Bursaphelenchus cocophilus. The potential biological control isolates (48 rhizobacteria and 27 Trichoderma spp.) were characterized biochemically. The biological control experiments were carried out "in vitro" and "in vivo", and in the later one, the mortality rate of the nematodes and the pathogenesis related enzymes were evaluated. Rhizobacterial isolates B14 and P41 provided a high nematode mortality rate (69% and 56%, respectively) and both were proteolytic and phosphate solubilizers. In the "in vivo" experiment, rhizobacterial isolates B14, B23 and P23 tested the highest mortality rates of B. cocophilus than the isolates of Trichoderma spp. and of the control as coconut saplings challenged with B. cocophilus showed indices higher in enzyme activities, peroxidases, chitinase and β-1,3-glucanases when compared to witness (seedling not inoculated). Summary Alkaloids and alkaloid-producing plants have the potential to reduce crop damage by plant-parasitic nematodes. In a series of in vitro experiments, the nematicidal activity of the tropane alkaloids, hyoscyamine and scopolamine, and a mixture of both on the root-knot nematode, Meloidogyne incognita, was tested. Solutions of 16 mg of compound per ml of solvent were used in concentrations of 80-1280 μg ml−1 of water. Inactivity of second-stage juveniles of M. incognita increased with increasing concentration and exposure time. Lethal concentrations (LC50) after 120 min of exposure ranged from 182.4 μg ml−1 for scopolamine to 318.4 μg ml−1 for hyoscyamine and 332.8 μg ml−1 for the combination of both alkaloids. Similarly, the same concentrations of scopolamine inhibited hatching of M. incognita to a greater extent than hyoscyamine. In a glasshouse experiment, M. incognita was not able to penetrate the roots, induce galls and reproduce on Datura stramonium, D. innoxia and D. tatula. Results indicate that the tropane alkaloids hyoscyamine and scopolamine contained in Datura plants express a strong nematicidal activity against M. incognita and could possibly be used for an alternative and sustainable nematode management.

Background: The Meloidogyne incognita is the most destructive pathogen and causes loss of yield of various economically important plants of various families. It is polyphagous in behaviour and has a high reproductive rate, these parameters are a hurdle in management of it. Various chemicals are used to control nematode worldwide that are very expensive and highly toxic to the environment and human beings. For ecologically safe method, three plants i.e. Verbesina encelioides, Moringa oleifera, Cassia fistula and their various parts such as leaf, stem and flower’s cold aqueous extracts were tried in vitro to test their nematicidal activity. Methods: Cold aqueous extracts (10% w/v) of each plant material were prepared by mixing 10g of each plant material in 100 ml of distilled water and kept aside for 48 hours. To estimate inhibition of egg hatching and mortality rate of second stage juveniles of Meloidogyne incognita, eggs and juveniles were exposed for 24, 48 and 72 hours in different concentrations (20 ppm to 200 ppm) of plant extracts. Result: All three tested plant extracts showed promising nematicidal activity against Meloidogyne incognita and the leaf extract of Moringa oleifera prevents the hatching of eggs. Inhibition of larval hatching from egg and nematode mortality rate were strongly influenced by concentration of extract, plant species and duration of exposure.

Abstract To investigate in vitro nematicidal activity against the pine wood nematode, Bursaphelenchus xylophilus, plant methanolic extracts were obtained from 26 medicinal plants and herbs in Vietnam. Of the plant extracts tested, a 10 mg ml−1 concentration of Cinnamomum cassia bark extract showed the highest level of nematicidal activity against juvenile and adult B. xylophilus after treatment for 7 h. The remainder of the plant extracts had little or no effect on juveniles or adults. Four solvents, n-hexane, chloroform, ethyl acetate and n-butanol, were used initially to partition the compounds obtained from C. cassia extracts. The compound (5 mg ml−1) in the n-hexane fraction showed the highest nematicidal activity at 60 min after treatment. Compounds III and IV were then separated from the n-hexane fraction using silica gel plates. These compounds showed very similar activity against the pine wood nematode. However, treatment with different concentrations of these compounds resulted in a significant difference in their effects on the mortality of the pine wood nematode, with greater concentrations leading to increased mortality after the same exposure time. Overall, treatment with 3 mg ml−1 of compounds III and IV resulted in greater than 95% mortality against juvenile and adult B. xylophilus at 50 min after treatment.
