Effects of in vitro simulated digestion on the flavonoid content and antioxidant activity of aged and fresh dried tangerine peel

This study was designed to evaluate the antioxidant activity of phenolic compounds for six cultivars of date from In Saleh region (Phoenix dactylifera L.), which were extracted by reflux with solvents, 80% methanol and 70% aceton after the phenolic and flavonoid content were estimated. The total phenolic content (TPC) and total flavonoid content (TFC) of the extracts were measured using Folin–Ciocalteau and aluminum chloride colorimetric methods, respectively. Antioxidant activity acetone/H2O and methanol/H2O extracts using ABTS, DPPH, FRAP, TAC and CUPRAC assays. The results showed that methanolic solvent reflux gives better yields. The highest level of phenolic compounds for all dates varieties was found in the 80% methanol extract. This level ranged from 8.04 ± 0.14 to 46.81 ± 1.22 mg GAE/100 g fresh weight and flavonoid content was observed relatively low for both two solvents. However, extraction into 70% acetone gave the highest antioxidant activity with all assay. These results showed that dates had strongly scavenging activity on DPPH reaching 3.12 µg/mL in methanol/H2O from Cul1 (6.9 µg/mL with ascorbic acid), also on ABTS reaching 6.51 µg/mL in acetone/H2O from Cul2 (104.98 µg/mL with ascorbic acid).

Cereus jamacaru D.C. (mandacaru) is a cactus used as food and in the traditional medicine. In the present study, hydroalcoholic extract of C. jamacaru was evaluated for its chemical composition, antioxidant activity, cytotoxic and anti-cytotoxic effects in human lymphocytes and sarcoma 180 cells in vitro by MTT assay and antitumoral, mutagenic and cytotoxic effects on mice sarcoma-induced in vivo. Phytochemical characterization showed positive reactions for coumarin, flavanol and tyramine and total flavonoid content of 0.51 µg/mL. C. jamacaru showed antioxidant activity following DPPH (EC50 = 427.74 µg/mL), ABTS (EC50 = 270.57 µg/mL) and Fe2+ chelating ions assays (EC50 = 41.18 µg/mL). C. jamacaru induced significant decrease of sarcoma 180 viability at 24 h and 48 h of treatment, did not induce cytotoxicity in human lymphocytes and inhibits the cytotoxicity of cisplatin in vitro. Following in vivo assays, C. jamacaru promoted tumor reduction (86.07% of tumor inhibition), without inducing mutagenic or cytotoxic damage on mice blood cells. We propose that phenolic and alkaloid compounds in the extract are related to antioxidant activity, increasing its ability in metal chelating activity and promoting anti-cytotoxic activity against cisplatin, as well as these compounds may act on the cell cycle of the tumor cells in vitro and in vivo, leading to anticancer effects and tumor reduction.

In the present study the extracts ofin vivoandin vitrogrown plants as well as callus tissue of red clover were tested for their antioxidant activities, using different extraction solvent and different antioxidant assays. The total flavonoid and phenolic contents as well as extraction yield of the extracts were also investigated to determine their correlation with the antioxidant activity of the extracts. Among all the tested extracts the highest amounts of total phenolic and total flavonoids content were found in methanol extract ofin vivogrown plants. The antioxidant activity of tested samples followed the orderin vivoplant extract > callus extract >in vitroextract. The highest reducing power, 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging, and chelating power were found in methanol extracts ofin vivogrown red clover, while the chloroform fraction ofin vivogrown plants showed the highest 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, superoxide anion radical scavenging and hydrogen peroxide scavenging compared to the other tested extracts. A significant correlation was found between the antioxidant activity of extracts and their total phenolic and total flavonoid content. According to the findings, the extract ofin vitroculture of red clover especially the callus tissue possesses a comparable antioxidant activity to thein vivocultured plants’ extract.

The aim of this study was to investigate the effect of raspberry juice addition on the content of phenolic compounds and the antioxidant activity of wafers. The research was carried out on non-supplemented wafers (control) and wafers in which water was replaced with raspberry juice in the amount of 10%, 20%, 50%, 75% and 100%. The potential bioavailability of the phenolic compounds after in vitro digestion was also determined. As shown by the consumer assessment, wafers in which the water was replaced with 100% raspberry juice turned out to be the best variant of the enriched wafers. The content of total phenolic compounds, flavonoids, phenolic acids, and anthocyanins in the tested products increased with the increasing amount of raspberry juice added to the wafers. The fortification of the wafers with raspberry juice had a positive effect on the antioxidant activity, expressed as the ability to neutralize free radicals ABTS (2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)) and DPPH (di(phenyl)-(2,4,6-trinitrophenyl)iminoazanium), the ability to chelate iron ions, and as the reduction power. The simulated digestion increased the content of phenolic compounds and increased the antioxidant activity of the wafers. The ability to inhibit lipoxygenase and cyclooxygenase 1 and 2 (i.e., enzymes involved in the induction of inflammation), varied and depended on both the amount of raspberry juice added and the type of extract.
