Objective: Evaluate the effects of saponin-rich extracts from Panax notoginseng roots with steaming - NP (H) and without steaming - NP (O) on some hematological and immunological indices in sarcoma TG180-bearing mice. Methods: The mice were injected with Sarcoma TG180 cells in the thigh to cause tumors, then were oral administrated NP(O), NP(H), Letinan or normal saline from the 6th day to 22nd day since developed tumors. On the 22nd day, blood was taken for IL-2, TNF-α and hematological indicators, revealing and determining the relative weight of spleen and thymus. Results: NP(H) and NP(O) at both level of the doses of 300 mg/kg/day and 900 mg/kg/day increased red blood cell count, hemoglobin, hematocrit, adjusted white blood cell and platelet counts return to physiological level, increased blood IL-2 and TNF-α, increased relative weight of spleen and thymus. Conclusion: The saponin-rich extracts from Panax notoginseng roots had the effects of improving hematological index and enhancing immunity on sarcoma 180-bearing mice, in which the extract from steaming Panax notoginseng roots has a better effect compared with no-steaming Panax notoginseng roots ones.

The genotoxicity of the ethanolic extract of betel nut was evaluated using sarcoma 180 tumour bearing mouse considering sperm motility, sperm viability, biochemical estimation of fructose in seminal fluid and sperm head morphology assays. Sperm head morphology was studied by H-E staining and Toluidine blue staining method. But Toluidine blue staining method is a reliable method to evaluate the DNA damage of sperms. Ethanolic BNE (betel nut extract) can suppress the percentage of sperm motility, sperm viability and seminal fructose level. In addition, it can also enhance the percentage of DNA damaged sperms. Moreover, histological sections of testes have been studied in control and BNE treated sarcoma 180 tumour bearing mice to highlight the potential toxic effect of BNE. The significant decreasing rate of seminal fructose concentration, sperm motility as well as viability and increasing rate of sperm head abnormality in different doses of treated series may be as a result of different toxic alkaloid ingredients present in BNE. Therefore, the results showed the potential of the BNE to induce different types of germ cell abnormalities in tumour bearing male mice.

O Sarcoma 180 (S180) é um tipo de câncer de mama, que está associado à uma alta morbimortalidade e, por isso, a elucidação dos mecanismos moleculares é importante. O avanço da Química Medicinal no planejamento in silico de fármacos e vem despertando o interesse de cientistas e indústrias farmacêuticas. Assim, este trabalho tem como principal objetivo avaliar parâmetros farmacocinéticos (ADME) in silico, por meio do software PreADMET, para ligantes derivados de pirrole de anel A oito, substituídos de duocarmicina B2, com avaliação antitumoral em S180. Os resultados inéditos obtidos corroboram com informações moleculares que são essenciais para prever a biodisponibilidade oral dos potentes candidatos à fármacos.

Background: Alpha-terpineol is a monoterpene alcohol with anti-tumor activity against different tumor cell lines (lung, breast, leukemias and colorectal) through blockage of NF-kB expression, which play an important role in tumor cells growth. Objective: Evaluate the antitumor activity of alpha-terpineol in murine Sarcoma 180 cell line Methods: For the tests, different cytotoxic and genotoxic assays were used, including Trypan blue, cytokinesis-blocked micronucleus assay, comet assay, agarose gel DNA fragmentation, flow cytometry and cell viability using fluorescence. Ascitic fluid cells from sarcoma 180 were obtained from Mus musculus peritoneal cavity and Alpha-terpineol was tested at 100, 250 and 500 μg/mL. Doxorubicin and Cisplatin were used as positive controls. Results: Cytotoxic effects of alpha-terpineol were found in all concentrations tested, reducing cell viability in 50.9; 38.53; 30.82% at 100, 250 and 500 μg/mL, respectively. Alpha-terpineol induced genotoxic effects due to DNA fragmentation (increased frequency and index of damage), and was clastogenic by increased micronuclei formation, nucleoplasmic bridges and nuclear buds. DNA fragmentation and increased cell death indicated that alpha-terpineol can cause early, late, and necrotic apoptosis. Conclusion: Our data indicate that alpha-terpineol has antitumor activity revealed by cytogenetic mechanisms and / or loss of cell membrane integrity.
