The role of Ca2+ in sturgeon sperm maturation and motility was investigated. Sperm from mature male sterlets (Acipenser ruthenus) were collected from the Wolffian duct and testis 24 h after hormone induction. Testicular spermatozoa (TS) were incubated in Wolffian duct seminal fluid (WDSF) for 5 min at 20°C and were designated ‘TS after IVM’ (TSM). Sperm motility was activated in media with different ion compositions, with motility parameters analysed from standard video microscopy records. To investigate the role of calcium transport in the IVM process, IVM was performed (5 min at 20°C) in the presence of 2 mM EGTA, 100 µM Verapamil or 100 µM Tetracaine. No motility was observed in the case of TS (10 mM Tris, 25 mM NaCl, 50 mM Sucr with or without the addition of 2 mM EGTA). Both incubation of TS in WDSF and supplementation of the activation medium with Ca2+ led to sperm motility. The minimal Ca2+ concentration required for motility activation of Wolffian duct spermatozoa, TS and TSM was determined (1–2 nM for Wolffian duct spermatozoa and TSM; approximately 0.6 mM for TS). Motility was obtained after the addition of verapamil to the incubation medium during IVM, whereas the addition of EGTA completely suppressed motility, implying Ca2+ involvement in sturgeon sperm maturation. Further studies into the roles of Ca2+ transport in sturgeon sperm maturation and motility are required.

AbstrakIndonesia memiliki banyak tumbuhan berpotensi obat salah satunya bisa dijadikan sebagai obatkontrasepsi tradisonal yang biasa digunakan untuk menjarangkan anak atau sterilisasi sepertirebusan akar Caesalpinia bonduc (Aru). Tujuan penelitian untuk mengetahui pengaruh obatkontrasepsi tradisional air rebusan akar aru terhadap kualitas pematangan sperma epididimis mencit(Mus musculus). Mencit dipilih secara acak untuk mewakili 4 kelompok dosis yaitu : 0,0 (K0), 16,0(K1) 24,0 (K2) dan 32,0 (K3) μl/gram berat badan/hari. Setiap kelompok perlakuan dilakukandengan 5 kali replikasi. Perlakuan diberikan melalui oral dengan menggunakan jarum gavage selama10 hari berturut-turut. Pembedahan dilakukan pada hari pertama setelah perlakuan selesai.Pengamatan kualitas sperma epididimis dengan perhitungan produksi sperma, % sperm motil, %sperma hidup, % sperma abnormal,dan rangking keaktifan sperma. Untuk mengetahui ada atautidaknya perbedaan yang bermakna antar perlakuan dalam kelompok dilakukan uji Anava satu arah,bila terdapat perbedaan bermakna dilanjutkan uji BNJ untuk membandingkan angka rata-rata antarkelompok perlakuan. Pemberian air rebusan akar Caesalpinea bonduc pada mencit menunjukanpengaruh perbedaan secara signifikan pada jumlah (%) sperma abnormal antara dosis 0,0 μl dengan24,0 dan 32,0 μl serta antara 16,0 μl dengan 24,0 dan 32,0 μl; Jumlah (%) sperma hidup terdapatperbedaan antara dosis 0,0 μl dengan 16,0 , 24,0 dan 32,0 μl; Jumlah (%) sperma motil terdapatperbedaan antara dosis 0,0 μl dengan 24,0 dan 32,0 μl; Rangking keaktifan sperma terdapatperbedaan antara dosis 0,0 μl dengan 24,0 dan 32,0 μl. The aims of this study is to know the effect of root boiled water ofCaesalpinia bonduc as traditional contraception medicine on quality of sperm maturation ofepididymis of mice (Mus musculus). Mice were choosed radomly and doses gouped: 0,0 (K0 ), 16,0(K1) 24,0 (K2) and 32,0 (K3) μl/gram body weight/day. Each of group was replicated 5 times.Treatment were given by oral with using gavage needle for ten days. Surgery was carried out on firstday after completing treatments. Examination of epidymis sperm by counting number of spermproduction, percentage of motil, live, abnormal sperm, and rank of sperm motility. In order to knowthe deferences between control and treated group, was used one way anava analysis and analysedvalue for comparing between teratment group. The treatments of root boiled water of Caesalpiniabonduc have effect significantly on percentage of abnormal sperm between dose of 0,0 μl with 24,0and 32,0 μl, also between 16,0 μl with 24,0 and 32,0 μl. Percentage of live sperem is different betweendose of 0,0 μl with 16,0 , 24,0 and 32,0 μl. Percentage of motile sperm is defferent significantlybetween dose of 0,0 μl with 24,0 and 32,0 μl. Meanwhile percentage of motile rank has differencebetween dose of 0,0 μl with 24,0 and 32,0 μl. The treatment of root boiled water of Caesalpiniabonduc has effect signifiantly on increasing percentange of sperm abnormality, decreasingpercentage of life sperm, motile and rank of motile of mice sperm.

The development of intracytoplasmic sperm injection has made the use of testicular sperm a viable option for infertile men with obstructive and nonobstructive azoospermia. Over the past decade, testicular biopsies have been handled and processed using a variety of different methods. Whole biopsy pieces can be effectively cryopreserved in a 10% glycerol diluent (Schiewe et al. 1997 67, S115 abst); however, the ability to find viable, motile sperm post-thaw is improved when prefreeze motility exists. The purpose of this study was to comparatively document in vitro sperm motility enhancement over time at different temperatures, and to prove that an intermediate temperature (28 to 30°C) would optimize sperm longevity for up to 1 week. In this study, 10 men with obstructive azoospermia underwent a surgical, open testicular biopsy procedure. Each biopsy was placed in HEPES buffered-human tubal fluid (mHTF) medium supplemented with 5% human serum albumin (HSA; Irvine Sci., Santa Ana, CA, USA) and transported to the laboratory at room temperature. Each testis biopsy (TBx) was dissected into 8 equal pieces (approximately 2 × 2 × 1 mm). Five intact pieces of TBx were cryopreserved in separate cryovials for future use. The remaining TBx tissue was subdivided into 1 of 3 temperature treatment groups (24, 30, or 37°C) for extended IVM. The 30°C incubation condition was achieved by placing the dish(es) in a Styrofoam box placed on a 37°C warming plate. Each TBx was placed into a separate 100 × 35 mm Falcon dish in 150-μL droplets of mHTF under oil and shredded by needle dissection to disperse the contents of the seminiferous tubules. Reminant tissue was placed into another droplet for additional dissection, as needed. Sperm were analyzed for motility (graded as Type I = twitching, II = undulating, III = slow progression, and IV = rapid progression) at 0, 24, 96, and 144 h without replacing the IVM medium. The percentage increase in motility, compared with 0 h, was statistically contrasted across temperature treatments by chi-squared analysis. Testicular sperm motility ranged from 5 to 25% at 0 h and increased in all groups at 24 h. There was no difference in total motility at 24 h, but progressive motility was higher (P < 0.05) at 37°C compared with 24°C. Significant differences (P < 0.05) were observed in total percentage of motility/percentage of progressive motility at 96 h with treatment differences (*) being 30°C (66%*/44%*) > 24°C (42%*/14%) > 37°C (18%*/10%). This statistical trend continued at 144 h with 30°C (42%*/23%*) > 24°C (24%*/8%) > 37°C (9%*/5%). During this study, a viable pregnancy was achieved using a 30°C sample 8 days post biopsy, exceeding 2 previous healthy triplet pregnancies, which were successful using 4-day-old TBx specimens in 1996 and 2004. This study confirms that an intermediate IVM temperature of 30°C is optimal to enhance TBx cryopreservation, which in our laboratory is generally performed at 24 to 72 h of IVM. Our routine pregnancy success in applying ICSI with IVM, thawed TBx sperm discounts the concern some individuals express regarding DNA fragmentation of sperm over time.

Abstract Study question Would the use of theophylline have an effect on ICSI outcomes? Summary answer The cumulative pregnancy rate after the transfer of fresh and frozen embryos (FET) becomes more important justifying the addition of theophylline as an efficacy variable. What is known already Absolute immotile spermatozoa is one of the most important causes of reduced fertilization and pregnancy rates after ICSI, immotility of testicular spermatozoa is a physiological event resulting from metabolic sperm immaturity. Over the years, there have been numerous attempts to resolve this problem by identification of pharmacological agents that might improve sperm motility. In particular, theophylline turned out to be an effective tool in stimulating motility in human semen and identifying viable sperm in testicular sperm extraction sperm (TESE) patients. aim of this study is to evaluate ICSI outcomes after the use of theophylline to select viable spermatozoa Study design, size, duration This prospective, comparative randomised study was conducted in Fertillia ART center in Tunisia, between november 2017 to november 2020. All patients underwent ICSI cycles with testicular sperm were included. The exclusion criteria consist of cycles with female partner age &gt;42 years and/or cycles with no information about pregnancy outcomes. In our study, the cycles were categorized into two groups according to sperm selection method used in ICSI procdure. Participants/materials, setting, methods The present study include 678 cycles. This cohort was randomly divided into two groups according to method of spermatozoa selection. Treatements arms were performed by theophylline to improve sperm motility (Group A) and the Hyposmotic swelling (HOS) test to indicate sperm vitality (Group B). Clinical and biological parameters, the duration of sperm selection and ICSI outcomes were compared between the two groups. A statistical significant difference was accepted when the p value was &lt;0.05. Main results and the role of chance Baseline clinical parameters were found to be comparable in the two groups. No differences regarding number of oocytes retrieved or MII oocytes were reported . he sperm selection was easier in the group A (time interval : 12±7,2 minutes) than group B (time interval : 22±10 minutes) (p &lt; 0,05). No significative difference was observed between goups in the Fertilization rate (group A : 66% vs group B : 68%) ; Cleavage rate (group A : 76.20% vs. Group B : 75.56%) and Blasulation rate (group A : 63.60% vs. Group B 60.61%) p &gt; 0 ,05. The Cumulative Pregnancy rate for group A (31. 5%) was higher than group B (30%, p &gt; 0 ,05). Indeed, The rate of frozen cycles is significantly higher for group A compared to group B (30% vs. 4.5% p &lt; 0,05). Limitations, reasons for caution Need large sample size. Wider implications of the findings: Theophylline reduces significantly the time needed for sperm selection, Which positively affects the ICSI result. Trial registration number Not applicable

Summary This study examined the effect of a cryoprotectant with and without pentoxifylline supplementation on the motility and viability of human testicular sperm, both before and after freezing. Testicular samples were obtained from 68 patients with azoospermia who came to the Andrology Service of West China Second University Hospital, Sichuan University, for testicular biopsies from December 2019 to April 2020. All patients were assigned randomly to two groups: experimental, whose testicular sperm were added to the cryoprotectant with pentoxifylline, and the control, whose testicular sperm were added to the cryoprotectant without pentoxifylline. Both groups used the same freezing and thawing methods. Testicular sperm motility in the experimental group was significantly higher than that of the control group, both before and after cryopreservation. The recovery rate of sperm motility in the experimental group was significantly higher than that of the control group. The percentage of samples with motile testicular sperm in the experimental group was significantly higher than that of the control group after thawing. Sperm viability was unchanged between the experimental and control groups, both before and after freezing. Overall, a pentoxifylline-supplemented cryoprotectant can significantly improve the motility of testicular sperm before and after cryopreservation.
