Chitosan is recognized as a multipurpose biomaterial because of its low allergenicity, non-toxicity, biodegradability and biocompatibility. The present study was designed to estimate the role of chitosan derived from Lucilia cuprina on burn healing in diabetic mice; using histopathological and microbiological studies at different time intervals. Chitosan was prepared from L. cuprina with high molecular weight (MW) and high degree of deacetylation (DD) to evaluate its burn wound healing potential; skin burn closure assessment, histological and microbiological studies in vivo in male diabetic mice. Chitosan topical treatment was superior in wound closure acceleration; mainly in insulin injected group at all the time intervals. Additionally, earlier epidermal remodelling with mature and intense collagen deposition was encountered in all chitosan treated animals as well as non-diabetic burned animals. There was a significant delay in hair growth and poor epidermal remodelling with impairment of wound closure in diabetic groups. Moreover, chitosan treated groups assert the chitosan antibacterial effects with protecting the burn against contamination that hinders healing especially in this diabetic condition. Further researches needed to interpret effects of possible synergistic combination therapy.

Abstract: Theoretically, batata, Ipomea batata L (Lam) theoritically can be used in wound healing since it acts as antiseptic, antibiotic, and anti-inflammatory agents. Batata ingredients can also stimulate epithelialization in wound bed. This study aimed to obtain the role of batata juice in burn wound healing noticed from the inflammatory response aspect. This was an experimental study conducted in the research laboratory of Faculty of Medicine Univeristy of Sam Ratulangi. Samples were six rabbits divided into 2 groups: treatment group with batata juice, and control group without treatment. The histological examination showed that the quality of inflammatory tissue of the treatment group was denser, settled, and associated with edema expansion compared to the control group with diminished inflammatory components, edema, and vasodilatation, meanwhile, there were angiogenesis, improvement of dermis, and regrowth of epidermis. Conclusion: Batata juice was not superior in healing burns than the natural responses.Keywords: batata (Ipomeas batata L), burns, skin, inflammation, wound healingAbstrak: Batata, Ipomea batata L (Lam) secara teoritik berfaedah untuk penyembuhan luka, yang bekerja sebagai antiseptik, antibiotik, dan anti inflamatorik. Batata berkhasiat dalam penyembuhan luka karena kandungannya yang secara bermakna merangsang pembentukan epitel jaringan luka. Penelitian ini bertujuan untuk mendapatkan peran batata terhadap penyembuhan luka bakar dilihat dari kualitas komponen radang pada jaringan luka. Sampel terdiri dari 6 ekor kelinci yang dibagi atas 2 kelompok: kelompok perlakuan dan kontrol. Kelompok perlakuan diberi jus batata pada luka bakar sedangkan kelompok kontrol tanpa perlakuan. Penelitian ini dilakukan di Laboratorium Riset Fakultas Kedokteran Universitas Sam Ratulangi dengan menggunakan metode eksperimental. Hasil identifikasi mikroskopik menunjukkan bahwa pada kelompok perlakuan kualitas komponen radang jaringan luka bakar lebih padat dan menetap disertai perluasan sembab kolagen dibandingkan kelompok kontrol dimana komponen radang dan sembab berkurang, dilatasi pembuluh darah berkurang, pertumbuhan kapiler baru, jaringan dermis yang membaik, dan epidermis yang mulai tumbuh kembali. Simpulan: Jus batata tidak memperlihatkan penyembuhan luka bakar yang lebih baik dibandingkan dengan penyembuhan luka oleh tubuh sendiri.Kata kunci: batata (Ipomeas batata L), luka bakar, kulit, radang, penyembuhan luka

Treatment of non-healing burn injuries is a major challenge for the current scientific research. Hydrogen sulfide (H2S) is an endogenous gasotransmitter, which regulates redox homeostasis and cytoprotection during pathophysiological conditions. Similarly, heat shock proteins (HSPs) are molecular chaperones, which also confer cytoprotection during the wound repair process. Notably, the role of H2S as a regulator of HSPs during burn wound healing is still elusive. The present study investigated the effects of H2S supplementation on molecular chaperones during full-thickness, third-degree burn wound healing in the experimental rats. The animals were treated with sodium hydrosulphide (NaHS) as H2S donor (5 mg/kg body weight, intraperitoneal) daily for 10 days prior to burn-induction and continued till the fifth-day post-wounding. Histopathological analysis (Masson’s trichrome) revealed enhanced wound healing evident by increased collagen fiber deposition, cellular proliferation and re-epithelialisation in NaHS administered group as compared to the burn control. Furthermore, immunoblot analyses demonstrated significantly increased protein expression of molecular chaperons viz. HSP90, HSP70, HSP27, and GRP78 in H2S treated group as compared to control. Therefore, the present study signifies that H2S supplementation upregulates the protein expression levels of molecular chaperones, which could facilitate the cytoprotection during the tissue repair process and accelerates the burn wound healing.

Abstract Crocus sativus is a spice with various pharmacological properties. Crocin, picrocrocin, and safranal are the main compositions of saffron that have recently been considered in the therapy of many diseases. High-performance liquid chromatography analysis revealed presence of these compounds in our saffron extract. This study was carried out to evaluate the effect of saffron on burn wound healing at an in vivo model. Saffron was topically applied on burn wounds in rats; the percentage of wound closure, wound contraction, and the levels of main cytokines and growth factors were measured. The saffron extract was also applied to evaluate the proliferation and migration of human dermal fibroblast (HDF) cells using in vitro scratch assay and resulted in active proliferation and migration of the HDF cells in a dose-dependent manner. A clear enhanced healing was observed in the saffron-treated wounds compared to the silver sulfadiazine and negative control groups. Decreased expression of interleukin-1β and transforming growth factor-β1 (TGF-β1) during the inflammatory phase demonstrated the role of saffron in promoting wound healing. In addition, enhanced TGF-β1 expression during the proliferative phase and basic fibroblast growth factor during the remodeling phase represented regenerative and anti-scarring role of saffron, respectively. Our histological and biochemical findings also confirmed that saffron significantly stimulated burn wound healing by modulating healing phases. Therefore, saffron can be an optimal option in promoting skin repair and regeneration. Application of this herbal medicinal drug should be encouraged because of its availability and negligible side effects.
