The consequences of mixed pollination on seed set in Vaccinium corymbosum

Self pollen of Vaccinium corymbosum germinates as well as outcross (compatible) pollen from the same population and more rapidly than compatible pollen from more distant populations. When a mixture of self and compatible pollen, either from the same or incontiguous populations, is applied to the stigma, the time required for berry ripening is significantly reduced compared with compatible pollen alone, although seed set also decreases significantly. Adding compatible pollen from different pollen donors significantly increased seed set but also increased the time for berry maturation by 5 days. Self pollen in conjunction with compatible pollen resulted in a 50% loss of viable seed production and an 8% gain in berry maturation time. Self-pollination resulted in embryo abortion in V. corymbosum. Key words: pollination, Vaccinium, seed set, outcrossing.

The reproductive biology of Schlumbergera truncata (Haworth) Moran and S. xbuckleyi (T. Moore) Tjaden was examined in a series of experiments. At anthesis, pollen grains are spherical, 54 to 62 μm in diameter, and tricellular. The receptive surface of the stigma is densely covered with elongated papillae and is devoid of exudate during the period of flower opening. When compatible pollen was applied to mature stigmas, germination occurred between 20 and 30 minutes after pollination and pollen tubes penetrated the stigma surface between 30 and 40 minutes after pollination. Pollen tubes exhibited a nonlinear pattern of growth in the upper two-thirds of the style, and the maximum rate of growth (≫1.9 mm·h-1) occurred between 12 and 18 hours after pollination. Full seed set was attained between 32 and 48 hours after pollination. Genotypic variation in the time required to achieve full seed set was partly attributable to differences in stylar length. Seeds were fully mature 6 months after pollination, but delaying fruit harvest until 8 months after pollination did not affect seed germination.

To find out the ability of Orobanche seeds to germinate immediately after seed set, seeds were germinated periodically at an interval of three months for one year in GR24. Some Orobanche seeds were capable of germination immediately after seed set but most required about nine months as after ripening or incubation period to be able to germinate. The phenomenon of after ripening in Orobanche seeds could be taken as an ecological measure to dormant over following unfavorable wet summer season. The growth hormone studies on Orobanche seed germination have shown that GA3 at a concentration of 100 ppm substantially enhanced seed germination when applied during pre-conditioning period. NAA showed some stimulatory effect at 0.5 - 1.0 ppm when applied during post-conditioning period but the hormone if applied during pre-conditioning period inhibited the germination. A study was made of barriers to hybridization between tetraploid V. corymbosum L., the cultivated highbush blueberry, and V. elliottii Chapm., a wild diploid blueberry which may be useful in breeding cultivars adapted to the southeastern United States. Both prefertilization and postfertilization barriers were detected. Prefertilization barriers were mainly observed when V. elliottii was the male parent and were manifested as an arrest of pollen tube growth at the base of the style. In cases where fertilization did take place, regardless of which species was used as the female, a strong postfertilization barrier usually prevented development of hybrid zygotes. Zygotes remained in a resting state after fertilization and usually aborted before dividing. Meanwhile, the endosperm underwent four to six cycles of cell division before it started to degenerate. A causal relationship between embryo abortion and endosperm malfunction was not clearly established. Endosperm malfunction and embryo abortion were delayed when V. corymbosum was the female parent.

The extractions of soil K by electroultrafiltration (EUF) and by chemical methods were compared as predictors of plant-available K for greenhouse-grown alfalfa on 38 Ontario soils. The relation of soil properties to the amount of K extracted by EUF fractions was also examined. The contents of silt and clay were negatively correlated with EUF-K at 50 V and 22 °C and positively correlated with EUF-K at 400 V and 80 °C. Soil pH and organic matter were not significantly correlated with the amounts of K extracted in the EUF-K fractions. The EUF extraction of K was influenced by the presence of carbonate or high exchangeable calcium in soils. Similar correlations were obtained between K uptake and K extracted by chemical methods and the sum of EUF-K fractions. When used along with other soil properties, EUF-K fractions and K extracted by chemical methods predicted availability of soil K with roughly equal ability. The model using the sum of K extracted by EUF at 50, 200 and 400 V is the simplest one and contains three variables, K, K2 and Ca2. The use of EUF is limited due to cost of equipment and time required for analysis, unless a number of nutrients can be accurately determined on one extract. Among the chemical methods, equations developed using three nonacidic extradants, NaCl, ABDTPA and NH4OAc explained more variation in K uptake than two acidic extractants, Mehlich 3 and 0.1 M HNO3. The NaCl model, ABDTPA model and NH4OAc model contained the same variables and had similar R2 values (0.88–0.91). Rainbow trout (Salmo gairdneri) homozygous for liver lactate dehydrogenase alleles ldhHαA and ldhHαB were artificially propagated and their swimming stamina compared. The time required to fatigue 50% of the HαAHαA phenotypes in fixed water velocity tests was 2.3 times greater on the average than that of HαAHαB phenotypes.Likewise, LDH phenotypes HαAHαA, HαAHαB, and HαAHαB of steelhead trout from the Thompson River were artifically propagated and their swimming stamina compared. In contrast to the rainbow trout, significant differences in stamina among the three phenotypes of steelhead were not evident in the stocks from this river nor between phenotypes HαAHαB and HαBHαB from another stream, the Vedder River, which has a very low frequency of the ldhHαA allele. The stamina of young steelhead from the Thompson River was, however, 3.8 times greater than that of those from the Vedder River. Recommendations for desiccation of alfalfa do not clearly define the stage at which desiccants should be applied. To obtain this information, diquat and glufosinate were applied to alfalfa (Medicago sativa L) from 1991 to 1994 at various stages of maturity to determine the effect of desiccant and stage of maturity at desiccation on seed yield 1000-seed weight and seed germinability. Alfalfa seed yields, 1000-seed weights, percent germination and percent viable seed were similar after desiccation with diquat and glufosinate. Desiccation when 60–75% of the alfalfa seed pods were brown permitted maximum seed yields. Percent germination increased slightly in seeds that were exposed to weathering under normal conditions until all pods turned brown. Both desiccants adequately desiccated alfalfa for harvesting but desiccation with glufosinate took 2 or 3 d longer than diquat. Desiccants did not reduce alfalfa growth in the spring after application. Key words: Germination, yield, hard seed, seed weight, stage

Lythrum cultivars Morden Pink, Morden Gleam, and Dropmore Purple, and a wild population of Lythrum salicaria from Lockport, Manitoba were artificially self- and cross-pollinated. Success of pollination and fertilization was measured in terms of the percentage of resulting capsules and the average numbers of viable seeds per capsule. Germination tests were used to measure the viability of seeds produced as a result of successful crosses. Cultivars rarely produced seeds as a result of selfing but many crosses with wild plants or with other cultivars were fertile. As expected, legitimate (i.e., different style morphs) wild crosses were highly fertile, but other crosses gave high fertility, especially those in which 'Morden Gleam' was the provider of pollen or ovules. Although some sterility was indicated in 'Dropmore Purple' and perhaps 'Morden Pink', neither are sufficiently sterile to be considered safe for ornamental flower gardens. Most illegitimate crosses yielded greatly reduced seed outputs in comparison to legitimate crosses, but there were some exceptions. Germinability of ripe seeds from any cross was high and averaged 98%. Cultivars of Lythrum are capable of contributing viable seed and (or) pollen to the spread of purple loosestrife. The sale of cultivars, regardless of parentage, should be prohibited. Key words: purple loosestrife, Lythrum spp., interfertility, hybridization.

Controlled pollination experiments were performed on the self-incompatible distylous herb Turnera ulmifolia L. to investigate the effects of pollination intensity and large amounts of incompatible pollen on seed set. In the first experiment, known numbers of compatible pollen grains ranging from 1 to 100 were applied to stigmas of the floral morphs. In both morphs, increasing amounts of pollen generally resulted in increased levels of seed set, although considerable variance was observed at all pollination intensities. Approximately two to seven pollen grains are required to produce a single seed and more than 95 grains are required to achieve maximum seed set in T. ulmifolia. Regression analysis of the seed set data failed to detect a difference in the response of the floral morphs to pollination intensity. In the second experiment, known proportions of compatible and incompatible pollen were applied to stigmas at various time intervals. Most treatments involving mixtures of compatible and incompatible pollen had no significant effect on seed set when compared with the controls. Clogging was only observed in the long-styled morph when one anther of compatible pollen was applied to stigmas 1.5 and 3.0 h after pollination with five anthers of incompatible pollen. The clogging of stigmas by incompatible pollen seems unlikely to have played a major role in the evolution and maintenance of distyly in Turnera ulmifolia.

Abstract Forty-one taxa of Miscanthus grown in USDA Hardiness Zones 4, 5, 6, and 7 were examined in 1996 and 1997 for seed set and viability. Although laboratory results varied widely between years, climatic zones, and cultivars, many plants set viable seed. Eleven types had <18% viable seed, including ‘Morning Light’, ‘Variegatus’ and ‘Zebrinus’, and appear to represent the least risk for becoming invasive plants, especially in northern climates. Other characteristics of Miscanthus are discussed in terms of invasive potential in the United States.

AbstractIn Arabidopsis, successful pollen-stigma interactions are dependent on rapid recognition of compatible pollen by the stigmatic papillae located on the surface of the pistil and the subsequent regulation of pollen hydration and germination, and followed by the growth of pollen tubes through the stigma surface. Here we have described the function of a novel gene, E6-like 1 (E6L1), that was identified through the analysis of transcriptome datasets, as one of highest-expressed genes in the stigma, and furthermore, its expression was largely restricted to the stigma and trichomes. The first E6 gene was initially identified as a highly-expressed gene during cotton fiber development, and related E6-like predicted proteins are found throughout the Angiosperms. To date, no orthologous genes have been assigned a biological function. Both the Arabidopsis E6L1 and cotton E6 proteins are predicted to be secreted, and this was confirmed using an E6L1:RFP fusion construct. To further investigate E6L1’s function, one T-DNA and two independent CRISPR-generated mutants were analyzed for compatible pollen-stigma interactions, and pollen hydration, pollen adhesion and seed set were mildly impaired for the e6l1 mutants. This work identifies E6L1 as a novel stigmatic factor that plays a role during the early post-pollination stages in Arabidopsis.Key MessageWe describe a function for a novel Arabidopsis gene, E6-like 1 (E6L1), that was identified as a highly-expressed gene in the stigma and plays a role in early post-pollination stages.
