Bell pepper (Capsicum annuum L.) fruit are susceptible to various molds, including grey mold and Alternaria rot. In this work, antagonistic bacteria isolated from disease-suppressive composts were assessed for their ability to reduce the mycelial growth of Botrytis cinerea Persoon and Alternaria alternata (Fries) Keissler as well as to suppress grey mold and Alternaria rot on bell pepper. Results showed that different bacterial antagonists reduced mycelial growth of B. cinerea and A. alternata by up to 49% and 53%, respectively. The bacteria provided inhibition of grey mold, reducing incidence and severity by as much as 43% and 67%, respectively. The isolates also provided strong inhibitory effects against Alternaria rot, decreasing incidence and severity by as much as 77% and 80%, respectively. Pseudomonas and Arthrobacter spp. were particularly suppressive to Alternaria rot. Results of this study suggest antagonistic bacteria may represent a potential method for controlling fruit rot of bell pepper.

The essential oil of Lavandula stoechas was examined by GC and GC-MS. Discs (5 mmi.d.) of the tested fungi ( Alternaria alternata, Fusarium oxysporum and Botritys cinerea) were inoculated separately onto each assay plate and incubated at 25oC for 7 days. The oil yield of dried parts (v/dw) obtained by hydro-distillation was 2.9%. Thirty-two compounds representing 98.3% of the essential oil were determined. Linalool (49.9%), linalyl acetate (14.4%), lavandulyl acetate (5.7%), α-terpineol (5.6%), terpinene-4-ol (5.1%), lavandulol (3.7%), ( E)-β-ocimene (2.6%) and ( Z)-β-ocimene (2.4%) were identified as the main constituents of the oil. In addition, both doses of the lavender oil showed varying levels of inhibitory effects on the mycelial growth of tested fungi used in the experiment. The results demonstrated the strongest effect on B.cinerea, followed by A.alternata and F.oxysporum. The inhibitory effect is probably dependent on the concentration of essential oils.

To develop new natural fungicides, six monoterpenes and two phenylpropenes were tested for their antifungal activity against eight plant pathogenic fungi. The results of the mycelial growth inhibition assay showed that trans-cinnamaldehyde was the most potent compound against the eight tested fungi with EC<sub>50</sub> values ranging between 0.75 and 3.19 mg/l. This compound caused the higher mycelial growth inhibition than carbendazim. Furthermore, (–)-menthone exhibited strong antifungal activity against Alternaria solani (EC<sub>50</sub> = 9.31 mg/l), Penicillium digitatum (EC<sub>50</sub> = 16.14 mg/l), and Rhizoctonia solani (EC<sub>50</sub> = 24.69 mg/l). Likewise, eugenol showed potent antifungal activity against P. digitatum, R. solani, Fusarium solani, and A. solani, whereas EC<sub>50</sub> values were less than 30.0 mg/l. In a separate experiment, trans-cinnamaldehyde, p-cymene, eugenol, and (–)-menthone were evaluated for their inhibitory effects on pectin methyl esterase and cellulase. The tested compounds exhibited the pronounced inhibition of enzyme activities with trans-cinnamaldehyde being the most potent inhibitor for both enzymes.

Tomato fruits were infected by various pathogens especially fungal pathogens under storage conditions as well as in field conditions. It was revealed that tomato fruits are attacked by Alternaria alternata causing Alternaria rot of tomato and Mucor plumbeus causing Mucor rot of tomato. Antifungal activity of some fungicides was undertaken during the present study.  Different concentrations of fungicides brought about significant reduction in the mycelial growth and spore germination of Alternaria alternata and Mucor plumbeus under in vitro conditions. Hexaconozole proved highly effective in inhibiting the mycelial growth and spore germination of Alternaria alternata followed by carbendazim and copper oxychloride respectively. However, the most effective fungicide in inhibiting the mycelial growth and spore germination of Mucor plumbeus was hexaconozole followed by carbendazim and copper oxychloride respectively. Six chemical fungicides and extracts of 15 locally available plants were tested against three fruit rot pathogens viz. Fusarium oxysporium f. sp. capsici, Rhizopus artocurpi and Alternaria tenuis for the evaluation of inhibition of spore germination and mycelial growth. Among the fungicides tested, all the concentrations of Ridomil showed 100% inhibition of spore germination and the least inhibition was recorded in case of treatment with Bavistin for F. oxysporium f. sp. capsici. In case of R. artocarpi and A. tenuis, Dithene–M 45 showed good inhibitory effects but Thiovit had no inhibitory effect against A. tenuis. Both leaf and seed extracts of Azadirachta indica showed good inhibitory effect than tested with other plant extracts. Moringa oleifera extract showed least inhibitory effect against all the fungi tested. Datura metel, Plygonum oriantale, Tagestes patula and Micania scandens also showed promising inhibition on spore germination and mycelial growth of all the pathogens tested. Microorganisms causing anthracnose diseases have a medium to a high level of resistance to the existing fungicides. This study aimed to investigate neem plant extract (propyl disulfide, PD) as an alternative to the current fungicides against mango’s anthracnose. Microorganisms were isolated from decayed mango and identified as Colletotrichum gloeosporioides and Colletotrichum acutatum. Next, a pathogenicity test was conducted and after fulfilling Koch’s postulates, fungi were reisolated from these symptomatic fruits and we thus obtained pure cultures. Then, different concentrations of PD were used against these fungi in vapor and agar diffusion assays. Ethanol and distilled water were served as control treatments. PD significantly (p ≤ 0.05) inhibited more of the mycelial growth of these fungi than both controls. The antifungal activity of PD increased with increasing concentrations. The vapor diffusion assay was more effective in inhibiting the mycelial growth of these fungi than the agar diffusion assay. A good fit (R2, 0.950) of the experimental data in the Gompertz growth model and a significant difference in the model parameters, i.e., lag phase (λ), stationary phase (A) and mycelial growth rate, further showed the antifungal efficacy of PD. Therefore, PD could be the best antimicrobial compound against a wide range of microorganisms.

Abstract Background Artemisia hedinii is a well-known traditional Chinese medicine. It can be used to extract dihydroartemisinin (DHA). Objective The purpose of this study was to explore the optimal conditions for the homogenate extraction of DHA from A. hedinii and the antifungal activity of DHA. Methods In this study, single factor experiments and response surface method were used to determine the optimal extraction conditions of crude extract and DHA, the method of spore germination was used to study the antifungal activity of DHA to Alternaria alternata. Result The optimal conditions were found as fellow: ratio of liquid to material 22 mL/g; Extraction time 60 s; soaking time 34 min. Under these conditions, extraction yield of DHA was (1.76 ± 0.04%). When the concentration of crude extract were 0.5 and 8 mg/mL, the spore germination inhibition rates of Alternaria alternata were (17.00 ± 2.05%) and (92.56 ± 2.01%), which were 3.34 and 1.15 times that of DHA standard, respectively. Conclusion Homogenate extraction technology is a fast and efficient method to extract DHA from A. hedinii. The crude extract has significant antifungal activity against A. alternata with low cost, which provides a possibility for the use of DHA in the prevention and treatment of plant pathogenic fungi. Highlights The optimum conditions of the extraction of DHA from A. hedinii by homogenate extraction were obtained. DHA has antifungal activity against A. alternata. Compared with pure DHA, the crude extract has stronger antifungal activity against A. alternata.

The main objective of this study was to evaluate the ability of both antagonistic bacteria Bacillus amyloliquefaciens (SF14) and Alcaligenes faecalis (ACBC1) used in combination with salicylic acid (SA) to effectively control brown rot disease caused by Monilinia fructigena. Four concentrations of salicylic acid (0.5%, 2%, 3.5%, and 5%) were tested under in vitro and in vivo conditions. Furthermore, the impact of biological treatments on nectarine fruit parameters’ quality, in particular, weight loss, titratable acidity, and soluble solids content, was evaluated. Regardless of the bacterium, the results indicated that all combined treatments displayed a strong inhibitory effect on the mycelial growth of M. fructigena and disease severity. Interestingly, all SA concentrations significantly improved the biocontrol activity of each antagonist. The mycelial growth inhibition rate ranged from 9.79% to 88.02% with the highest reduction rate recorded for bacterial antagonists in combination with SA at both concentrations of 0.5% and 3.5%. The in vivo results confirmed the in vitro results with a disease severity varying from 0.00% to 51.91%. A significant biocontrol improvement was obtained with both antagonistic bacteria when used in combination with SA at concentrations of 0.5% and 2%. The lowest disease severity observed with ACBC1 compared with SF14 is likely due to a rapid adaptation and increase of antagonistic bacteria population in wounded sites. The impact of all biological treatments revealed moderate significant changes in the fruit quality parameters with weight loss for several treatments. These results suggest that the improved disease control of both antagonistic bacteria was more likely directly linked to both the inhibitory effects of SA on pathogen growth and induced fruit resistance.

Pepper (Capsicum annuum L.) is an important vegetable crop worldwide. Some Fusarium species can cause pepper fruit rot, leading to significant yield losses of pepper production and, for some Fusarium species, potential risk of mycotoxin contamination. A total of 106 diseased pepper fruit samples were collected from various pepper cultivars from seven provinces (Gansu, Hainan, Heilongjiang, Hunan, Shandong, Shanghai, and Zhejiang) in China during the 2012 growing season, where pepper production occurs on approximately 25,000 ha. Pepper fruit rot symptom incidence ranged from 5 to 20% in individual fields. Symptomatic fruit tissue was surface-sterilized in 0.1% HgCl2 for 1 min, dipped in 70% ethanol for 30 s, then rinsed in sterilized distilled water three times, dried, and plated in 90 mm diameter petri dishes containing potato dextrose agar (PDA). After incubation for 5 days at 28°C in the dark, putative Fusarium colonies were purified by single-sporing. Forty-three Fusarium strains were isolated and identified to species as described previously (1,2). Morphological characteristics of one strain were identical to those of F. concentricum. Aerial mycelium was reddish-white with an average growth rate of 4.2 to 4.3 mm/day at 25°C in the dark on PDA. Pigments in the agar were formed in alternating red and orange concentric rings. Microconidia were 0- to 1-septate, mostly 0-septate, and oval, obovoid to allantoid. Macroconidia were relatively slender with no significant curvature, 3- to 5-septate, with a beaked apical cell and a foot-shaped basal cell. To confirm the species identity, the partial TEF gene sequence (646 bp) was amplified and sequenced (GenBank Accession No. KC816735). A BLASTn search with TEF gene sequences in NCBI and the Fusarium ID databases revealed 99.7 and 100% sequence identity, respectively, to known TEF sequences of F. concentricum. Thus, both morphological and molecular criteria supported identification of the strain as F. concentricum. This strain was deposited as Accession MUCL 54697 (http://bccm.belspo.be/about/mucl.php). Pathogenicity of the strain was confirmed by inoculating 10 wounded, mature pepper fruits that had been harvested 70 days after planting the cultivar Zhongjiao-5 with a conidial suspension (1 × 106 spores/ml), as described previously (3). A control treatment consisted of inoculating 10 pepper fruits of the same cultivar with sterilized distilled water. The fruit were incubated at 25°C in a moist chamber, and the experiment was repeated independently in triplicate. Initially, green to dark brown lesions were observed on the outer surface of inoculated fruit. Typical soft-rot symptoms and lesions were observed on the inner wall when the fruit were cut open 10 days post-inoculation. Some infected seeds in the fruits were grayish-black and covered by mycelium, similar to the original fruit symptoms observed at the sampling sites. The control fruit remained healthy after 10 days of incubation. The same fungus was isolated from the inoculated infected fruit using the method described above, but no fungal growth was observed from the control fruit.
